Alzheimer’s disease (AD) is the most common neurodegenerative disease worldwide. Histopathologically, AD presents with two hallmarks: neurofibrillary tangles (NFTs), and aggregates of amyloid β peptide (Aβ) both in the brain parenchyma as neuritic plaques, and around blood vessels as cerebral amyloid angiopathy (CAA). According to the vascular hypothesis of AD, vascular risk factors can result in dysregulation of the neurovascular unit (NVU) and hypoxia. Hypoxia may reduce Aβ clearance from the brain and increase its production, leading to both parenchymal and vascular accumulation of Aβ. An increase in Aβ amplifies neuronal dysfunction, NFT formation, and accelerates neurodegeneration, resulting in dementia. In recent decades, therapeutic approaches have attempted to decrease the levels of abnormal Aβ or tau levels in the AD brain. However, several of these approaches have either been associated with an inappropriate immune response triggering inflammation, or have failed to improve cognition. Here, we review the pathogenesis and potential therapeutic targets associated with dysfunction of the NVU in AD.  相似文献

Alzheimer’s disease (AD) is a neurodegenerative disease, characterized histopathologically by intra-neuronal tau-related lesions and by the accumulation of amyloid β-peptide (Aβ) in the brain parenchyma and around cerebral blood vessels. According to the vascular hypothesis of AD, an alteration in the neurovascular unit (NVU) could lead to Aβ vascular accumulation and promote neuronal dysfunction, accelerating neurodegeneration and dementia. To date, the effects of insoluble vascular Aβ deposits on the NVU and the blood–brain barrier (BBB) are unknown. In this study, we analyze different Aβ species and their association with the cells that make up the NVU. We evaluated post-mortem AD brain tissue. Multiple immunofluorescence assays were performed against different species of Aβ and the main elements that constitute the NVU. Our results showed that there are insoluble vascular deposits of both full-length and truncated Aβ species. Besides, insoluble aggregates are associated with a decrease in the phenotype of the cellular components that constitute the NVU and with BBB disruption. This approach could help identify new therapeutic targets against key molecules and receptors in the NVU that can prevent the accumulation of vascular fibrillar Aβ in AD.  相似文献

The wettability of granular materials has a direct effect on the workability of pastes, mortars and concretes containing such materials. Given this, the wetting behaviour of two Portland cements, two mineral admixtures (limestone filler and silica fume) and different sand types (siliceous, limestone and granite) was evaluated through the measurement of the contact angle between these materials and water. The results show that all the materials possess high wettability. A chemical agent, dichlorodimethylsilane, was used to modify the surface properties of a 0–1 mm siliceous sand, making it more hydrophobic and, therefore, unable to be spontaneously wetted by water. The properties of a mortar produced with the silane-treated sand were evaluated, showing improvements on workability as a result of the chemical treatment.  相似文献

The limited industrial processing and export to European countries of fresh Mexican ‘Ataulfo’ mangoes is attributed in part to the lack of homogeneous ripening among fruit from the same lot. A viable technology to alleviate the situation is the application of exogenous ethylene. In this work, vacuum (34 kPa) was applied for 20 min to ‘Ataulfo’ mangoes that were later exposed to exogenous ethylene (500, 1000 and 1500 μL L−1) for 30 min and ripening was monitored. Application of vacuum did not produce apparent visual damage to the fruit; when 1500 μL L−1 ethylene were applied for 30 min after the vacuum, induced production of internal ethylene with a concomitant increase in respiration rate. Firmness and acidity loss proceeded faster after the fruits were exposed to vacuum and 1500 μL L−1 ethylene; similarly, total soluble solids increased and pulp and peel color development was 100% in the whole lot. An overall reduction of three days from the normal ripening time was observed attributed to the treatments. It is proposed that short exposures to vacuum and ethylene could improve the uniformity in mango ripening.  相似文献

Basically, nonorthogonal multiple access (NOMA) allows controllable interferences by nonorthogonal resource allocation with a tolerable increase in receiver complexity, using successive interference cancellation (SIC). In this paper, authors show a novel mathematical model of teletraffic for users accessing, simultaneously, to Wi‐Fi and LTE services by using NOMA, calculating the birth–death process by order statistics. This methodology has not been previously used. The mathematical model is represented by a two‐dimensional Markovian process where the users can born or die in one system or another, independently, and results are validated through simulations. The outcomes demonstrate an increment of the sum‐rate gain for a system where users are ordered in the power domain using order statistics, according to the signal interference plus noise ratio (SINR), compared to a system where these statistics are not employed. By using the proposed mathematical model, the sum‐rate gain increases around 15%, and it is also demonstrated that this algorithm foresees a steady behavior of the sum rate, even when the number of channels is augmented, contrary when no order statistics are employed, in which the sum rate decreases considerably. On the other hand, the blocking probability is also demonstrated based on the chance of moving from one technology to another.  相似文献

Abstract

This article offers a framework for understanding how energy is used to meet water demand in countries. Specifically, the relationships between energy use and water scarcity, the location of renewable water resources, and aggregate water demand are explored. The article also examines how policy options such as water price reforms, agriculture subsidies and crop elimination may influence the energy use and energy intensity of water withdrawals. Conclusions suggest that while policy options exist, certain uncontrollable factors such as severe water scarcity or substantial freshwater abundance limit the ability of some countries to significantly improve the aggregate energy efficiency of water provision.  相似文献

Solutions of methyl orange azo dye were degraded by electrochemical oxidation using a 3 L flow plant with a boron-doped diamond (BDD)/stainless steel cell operating at constant current density, ambient temperature and liquid flow rate of 12 L min?1. A 23 factorial design considering the applied current density, azo dye concentration and electrolysis time as variable independents was used to analyze the process by response surface methodology. LC–MS analysis revealed the formation of seven oxidation products from the cleavage of the NN group of the dye, followed by deamination, formation of a nitro group and/or desulfonation of the resulting aromatics.  相似文献

Bacterial ATP binding cassette (ABC) transporters mediate the influx of numerous substrates. The cluster A-I ABC transporters are responsible for the specific uptake of the essential metals zinc, manganese or iron, making them necessary for survival in metal-limited environments, which for pathogens include the animal host. In Paracoccus denitrificans, there are two zinc ABC transporter systems: ZnuABC and AztABCD with apparently redundant functions under zinc-limited conditions. The unusual presence of two zinc ABC transporter systems in the same organism allowed for the investigation of specificity in the interaction between the solute binding protein (SBP) and its cognate permease. We also assessed the role of flexible loop features in the SBP in permease binding and zinc transport. The results indicate that the SBP–permease interaction is highly specific and does not require the flexible loop features of the SBP. We also present an expanded table of the properties of characterized cluster A-I SBPs and a multiple sequence alignment highlighting the conserved features. Through this analysis, an apparently new family of binding proteins associated with ABC transporters was identified. The presence of homologues in several human pathogens raises the possibility of using it as a target for the development of new antimicrobial therapies.  相似文献
