We discovered that a food aversion could be conditioned in anesthetized sheep. Sheep were allowed to eat a familiar food (alfalfa-grain pellets) for 30 min, and 90 min later they were given either an intraruminal (IR) injection of water (C), an IR injection of LiCl (L), anesthesia followed by an IR injection of water (A), or anesthesia followed by an IR injection of LiCl (A+L). Induction of anesthesia was by an intravenous injection of pentobarbitone sodium, and maintenance of deep anesthesia was by halothane. Sheep were maintained in deep anesthesia for 2 h to ensure that the effects of LiCl on the acquisition of a food aversion, which occur within about 1 h, were completed before they awakened. When tested 5 days later, sheep that received LiCl (treatments L and A+L) consumed less alfalfa-grain pellets than sheep that did not receive LiCl (treatments C and A) (241 g vs. 306 g; p = 0.057). Intake of sheep that were anesthetized (treatments A and A+L) did not differ from that of sheep that were not anesthetized (treatments C and L) (295 g vs. 252 g; p = 0.183). Nor was there an interaction between LiCl and anesthesia (p = 0.423). Thus, we conclude that changes in preferences for foods caused by postingestive feedback occur automatically every time food is ingested (i.e., they are noncognitive), and the kind and amount of feedback is a function of the match between the food's chemical characteristics and its ability to meet the animal's current demands for nutrients. 相似文献

BACKGROUND: Fluorophore dyes are used extensively in biomedical research to sensitively assay cellular constituents and physiology. We have created, as proof of principle, fluorophore dye binding peptides that could have applications in fluorescent dye-based approaches in vitro and in vivo. RESULTS: A panel of Texas red, Rhodamine red, Oregon green 514 and fluorescein binding peptides, termed here 'fluorettes', was selected via biopanning of a combinatorial library of 12-mer peptides fused to a minor coat pIII protein of the filamentous bacteriophage M13. The 'best' fluorette sequences from each of the groups were subjected to further mutagenesis, followed by a second biopanning to select a new generation of improved fluorettes. Phage were selected that had higher avidity for each fluorophore except Rhodamine red. Of these, peptides were characterized that could specifically and with high affinity bind at least one dye, Texas red, in solution. In addition, the binding of certain peptides to Texas red shifted the peak excitation and/or the emission spectra of the bound dye. CONCLUSIONS: Peptides in the context of phage display could readily be selected that could bind to small-molecule fluorophores. The affinities of selected mutant fluorettes could be increased by mutation and further selection. Only a subset of the free peptides could bind free dyes in solution, suggesting that phage context contributed to the selection and ability of certain peptidic regions to independently bind the dyes. Future screens might lead to the creation of other dye-binding peptides with novel characteristics or Texas red derivatives with cross-linking substituents might be designed to increase the utility of the system. 相似文献

To achieve this, volunteers download and install a software application capable of sensing periods of low local processor activity. The Wabash River Integrated Methanol and Power Production from Clean Coal Technologies (IMPPCCT) project is investigating an Early Entrance Coproduction Plant (EECP) concept to evaluate integrated electrical power generation and methanol production from coal and other carbonaceous feedstocks. Research, development and testing (RD&T) that is currently being conducted under the project is evaluating cost effective process systems for removing contaminants, particularly sulfur species, from the generated gas which contains mainly synthesis gas (syngas), CO2 and steam at concentrations acceptable for the methanol synthesis catalyst. The RD&T includes laboratory testing followed by bench-scale and field testing at the SG Solutions Gasification Plant located in West Terre Haute, Indiana. The creation of idealised, dimensionally reduced meshes for preliminary design and optimisation remains a time-consuming, manual task. A dimensionally reduced model is ideal for assessing design changes through modification of element properties without the need to create a new geometry or mesh. In this paper, a novel approach for automating the creation of mixed dimensional meshes is presented. The input to the process is a solid model which has been decomposed into a non-manifold assembly of smaller volumes with different meshing significance. Associativity between the original solid model and the dimensionally reduced equivalent is maintained. The approach is validated by means of a free-free modal analysis on an output mesh of a gas turbine engine component of industrial complexity. Cylindrical buttuohead specimens of an advanced silicon nitride were tested in uniaxial tension at temperatures between 1422 and 1673 K. In the range 1477 to 1673 K, creep deformation was reliably measured using high-temperature contact probe extensometry. Extensive scanning and transmission electron microscopy has revealed the formation of lenticular cavities at two-grain junctions at all temperatures (1422–1673 K) and extensive triple-junction cavitation occurring at the higher temperatures (1644–1673 K). Cavitation is believed to be part of the net creep process. The stress rupture data show stratification of the Monkman–Grant lines with respect to temperature. Failure strain increased with increase in rupture time or temperature, or decrease in stress. Fractography showed that final failure occurred by subcritical crack growth in all specimens. 相似文献

The removal of sulphur and ash from coal treated with aqueous hydrogen peroxide/sulphuric acid solutions has been studied at ambient temperature, under a variety of experimental conditions. Almost complete elimination of the sulphate and the pyritic sulphur was observed in most cases, as well as substantial reduction in the ash content. The other components of the organic coal matrix were not affected to a significant extent, indicating high selectivity of the system towards sulphur oxidation. Detailed measurements of the flow instability of dilute shear-thinning viscoelastic aqueous solutions, with relatively low zero-shear viscosities, in an obstructed microchannel flow are reported. We examine the flow behaviour resulting from a 100 μm post placed in the channel centreline over a range of Reynolds numbers (\(5<Re<300\)) and Weissenberg numbers (\(20<Wi<10^3\)). Micro-particle image velocimetry measurements show the onset of an upstream instability within a Reynolds number range and at a critical elasticity number corresponding to polymer concentrations above 25 ppm of long-chain polyacrylamide. The instability results in significant local fluctuations in the flow field approaching 30 % of the mean velocity. The magnitude of the local viscosity ratio in the region upstream of the post is proposed as a driving mechanism for the instability which resembles a buckling flow. Additionally, the classical instability owing to separation and vortex formation downstream of the post in Newtonian flow is suppressed and a very long stable wake is observed extending over 10 post-diameters downstream. 相似文献
