The targeting of karyophilic proteins to nuclear pores is mediated via the formation of a nuclear pore-targeting complex, through the interaction of nuclear localization signal (NLS) with its NLS receptor. Recently, a novel human protein, Qip1, was identified from a yeast two-hybrid system with DNA helicase Q1. This study demonstrates that Qip1 is a novel third class of NLS receptor that efficiently recognizes the NLS of the helicase Q1. Moreover, the data obtained in this study show that the specific interaction between Qip1 and the NLS of the helicase Q1 requires its upstream sequence of the minimal essential NLS. By using purified recombinant proteins alone in the digitonin-permeabilized cell-free transport system, it was demonstrated that the two known human NLS receptors, Rch1 and NPI-1, are able to transport all the tested NLS substrates into the nucleus, while Qip1 most efficiently transports the helicase Q1-NLS substrates, which contain its upstream sequence in so far as we have examined the system. Furthermore, in HeLa cell crude cytosol, it was found that endogenous Rch1 binds to all the tested NLS substrates, while the binding of endogenous NPI-1 is restricted to only some NLSs, despite the fact that NPI-1 itself shows binding activity to a variety of NLSs. These results indicate that at least three structurally and functionally distinct NLS receptors exist in the human single cell population, and suggest that the nuclear import of karyophilic proteins may be controlled in a complex manner at the NLS recognition step by the existence of a variety of NLS receptors with various specificities to each NLS.  相似文献

DNA-wrapped single-wall carbon nanotubes (DNA-SWNT hybrids) prepared from different diameter HiPco- and Arc-SWNTs were investigated by atomic force microscopy. The mean diameter of DNA-HiPco-SWNT hybrids is 1.94 nm that is consistent with one HiPco-SWNT (~ 0.9 nm) wrapped by DNA (~ 1 nm). On the other hand, the mean diameter of DNA-Arc-SWNT hybrids is 3.74 nm that can correspond to one Arc-SWNT (~ 1.4 nm) wrapped by several layers of DNA. It is suggested that the DNA-wrapping mechanism for large diameter Arc-SWNTs is different from that for small diameter HiPco-SWNTs.  相似文献

If this is indeed the case, then this form of human activity should be considered to be an important element of our lives and therefore worthy of intervention when circumstances restrict an individual's engagement with it. To investigate the therapeutic nature of work for people with enduring mental health problems, the author conducted a pilot case study of a work rehabilitation scheme in the United Kingdom. Cyanobacteriochromes (CBCRs) are promising optogenetic tools for their diverse absorption properties with a single compact cofactor-binding domain. We previously uncovered the ultrafast reversible photoswitching dynamics of a red/green photoreceptor AnPixJg2, which binds phycocyanobilin (PCB) that is unavailable in mammalian cells. Biliverdin (BV) is a mammalian cofactor with a similar structure to PCB but exhibits redder absorption. To improve the AnPixJg2 feasibility in mammalian applications, AnPixJg2_BV4 with only four mutations has been engineered to incorporate BV. Herein, we implemented femtosecond transient absorption (fs-TA) and ground state femtosecond stimulated Raman spectroscopy (GS-FSRS) to uncover transient electronic dynamics on molecular time scales and key structural motions responsible for the photoconversion of AnPixJg2_BV4 with PCB (Bpcb) and BV (Bbv) cofactors in comparison with the parent AnPixJg2 (Apcb). Bpcb adopts the same photoconversion scheme as Apcb, while BV4 mutations create a less bulky environment around the cofactor D ring that promotes a faster twist. The engineered Bbv employs a reversible clockwise/counterclockwise photoswitching that requires a two-step twist on ~5 and 35 picosecond (ps) time scales. The primary forward Pfr → Po transition displays equal amplitude weights between the two processes before reaching a conical intersection. In contrast, the primary reverse Po → Pfr transition shows a 2:1 weight ratio of the ~35 ps over 5 ps component, implying notable changes to the D-ring-twisting pathway. Moreover, we performed pre-resonance GS-FSRS and quantum calculations to identify the Bbv vibrational marker bands at ~659,797, and 1225 cm−1. These modes reveal a stronger H-bonding network around the BV cofactor A ring with BV4 mutations, corroborating the D-ring-dominant reversible photoswitching pathway in the excited state. Implementation of BV4 mutations in other PCB-binding GAF domains like AnPixJg4, AM1_1870g3, and NpF2164g5 could promote similar efficient reversible photoswitching for more directional bioimaging and optogenetic applications, and inspire other bioengineering advances.  相似文献

A new hot dip Zn-7Al alloy coating was performed on a structural steel by double coating of fine Zn and Zn-7 wt.% Al alloy, to prevent severe corrosion in coastal area. The alloy-coated steels were exposed to seaside, quasi-industrial, and rural districts to compare with conventional Zn coating. Double coating was significantly effective in preventing corrosion, particularly in a seaside. It was estimated from the exposure test for 10 years that the life of the Zn-7Al alloy-coated steel would be almost four times that of the Zn-coated steel in the seaside. A bending test showed that no exfoliation occurred at the interface between the coated alloy and substrate steel. TEM observation revealed that the excellent adhesiveness of the doubly coated fine Zn and Zn-7Al alloy to the steel substrate was due to formation of the interface region consisting of heterogeneous fine phase mixture of zinc, aluminium and iron.  相似文献

Pulsed plasma thrusters (PPTs) have the advantages of mechanical simplicity and robustness compared to other electric propulsion systems. However the thrust power ratio of PPTs is lower than that of other electric propulsion systems. To enhance the thrust performance of the PPT, we propose to use several combustible solid chemicals as coaxial PPT propellants to replace Teflon® (Polytetrafluoroethylene: PTFE). With the design, the force obtained thermodynamically is expected to augment the PPT thrust power ratio with the help of the chemical energy contained in the propellants. As a result, the thrust power ratio is increased using hydroxyl-terminated polybutadiene-ammonium perchlorate (HTPB-AP)-based propellants compared to the case of ordinary Teflon. The discharge current and voltage waveform does not change even when the propellant is changed. These findings could indicate that the impulse bits by gasdynamic contribution are lager in the case of chemical solid propellants than in the case of Teflon.  相似文献

Safety demonstration tests using the High Temperature Engineering Test Reactor (HTTR) will be conducted for the purpose of demonstrating inherent safety features of High Temperature Gas-cooled Reactors (HTGRs) as well as providing the core and plant transient data for validation of HTGR safety analysis codes. The first phase safety demonstration test items include the reactivity insertion test and the coolant flow reduction test. In the reactivity insertion test, which is the control rod withdrawal test, one pair out of 16 pairs of control rods is withdrawn, simulating a reactivity insertion event. The coolant flow reduction test consists of the partial loss of coolant flow test and the gas circulators trip test. In the partial loss of coolant flow test, primary coolant flow rate is slightly reduced by control system. In the gas circulators trip test one and two out of three gas circulators are run down, simulating coolant flow reduction events. The gas circulators trip tests, in which position of control rods are kept unchanged, are simulation tests of anticipated transients without scram (ATWS).  相似文献

Polymerized ionic liquids (PILs) having guanidinium cations with different counter‐anions, such as PF6? and N(CF3SO2)2? (TFSI?), were synthesized by copolymerization of a guanidinium ionic liquid monomer with methyl acrylate followed by an anion exchange reaction. Furthermore, incorporating a guanidinium ionic liquid, LiTFSI salt and nano‐size SiO2, a quaternary gel polymer electrolyte based on one of the PILs as the polymer host was prepared. The quaternary gel polymer electrolyte was chemically stable even at a higher temperature of 80 °C in contact with the lithium anode. In particular, the electrolyte exhibited high lithium ion conductivity, wide electrochemical stability window and good lithium stripping/plating performance. Li/LiFePO4 batteries with the quaternary gel polymer electrolyte at 80 °C had capacities of 140 and 130 mA h g?1 respectively at 0.1 and 0.2 C current rates. Due to rapid construction, necessity for raw materials of concrete, especially coarse aggregate, tends to increase the danger of early exhaustion of the natural resources. An alternative source of raw materials would perhaps delay the advent of this early exhaustion. Recycled coarse aggregate (RCA) plays a great role as an alternative raw material that can replace the natural coarse aggregate (NCA) for concrete. Previous studies show that the properties of RCA concrete are inferior in quality compared to NCA concrete. This article attempts to study the improvement of properties of RCA concrete with the addition of bacteria named as Bacillus subtilis. The experimental investigation was carried out to evaluate the improvement of the compressive strength, capillary water absorption, and drying shrinkage of RCA concrete incorporating bacteria. The compressive strength of RCA concrete is found to be increased by about 20% when the cell concentration of B. subtilis is 106 cells/ml. The capillary water absorption as well as drying shrinkage of RCA are reduced when bacteria is incorporated. The improvement of RCA concrete is confirmed to be due to the calcium carbonate precipitation as observed from the microstructure studies carried out on it such as EDX, SEM, and XRD.  相似文献

The effects of water chemistry distribution on the potential of a reference electrode and of the potential distribution on the measured potential should be known qualitatively to obtain accurate electrochemical corrosion potential (ECP) data in BWRs. First, the effects of oxygen on a platinum reference electrode were studied in 553 K pure water containing dissolved hydrogen (DH) concentration of 26–105 μg kg?1 (ppb). The platinum electrode worked in the same way as the theoretical hydrogen electrode under the condition that the molar ratio of DH to dissolved oxygen (DO) was more than 10 and that DO was less than 100 ppb. Second, the effects of potential distribution on the measured potential were studied by using the ECP measurement part without platinum deposition on the surfaces connected to another ECP measurement part with platinum deposition on the surfaces in 553 K pure water containing 100–130 ppb of DH or 100–130 ppb of DH plus 400 ppb of hydrogen peroxide. Measured potentials for each ECP measurement part were in good agreement with literature data for each surface condition. The lead wire connecting point did not affect the measured potential. Potential should be measured at the nearest point from the reference electrode in which case it will be not affected by either the potential distribution or the connection point of the lead wire in pure water.  相似文献
