Core-shell hybrid nanomaterials have shown new properties and functions that are not attainable by their single counterparts.Nanoscale confinement effect by porous inorganic shells in the hybrid nanostructures plays an important role for chemical transformation of the core nanoparticles.However,metal-organic frameworks(MOFs)have been rarely applied for understanding mechanical insight into such nanoscale phenomena in confinement,although MOFs would provide a variety of properties for the confining environment than other inorganic shells such as silica and zeolite.Here,we examine chemical transformation of a gold nanorod core enclosed by a zeolitic imidazolate framework(ZIF)through chemical etching and regrowth,followed by quantitative analysis in the core dimension and curvature.We find the nanorod core shows template-effective behavior in its morphological transformation.In the etching event,the nanorod core is spherically carved from its tips.The regrowth on the spherically etched core inside the ZIF gives rise toformation of a raspberry-like branched nanostructure in contrast to the growth of an octahedral shape in bulk condition.We attribute the shell-directed regrowth to void space generated at the interfaces between the etched core and the ZIF shell,intercrystalline gaps in mult-domain ZIF shells,and local structural deformation from the acidic reaction conditions.  相似文献

Immuno-electron microscopy (Immuno-EM) is a powerful tool for identifying molecular targets with ultrastructural details in biological specimens. However, technical barriers, such as the loss of ultrastructural integrity, the decrease in antigenicity, or artifacts in the handling process, hinder the widespread use of the technique by biomedical researchers. We developed a method to overcome such challenges by combining light and electron microscopy with immunolabeling based on Tokuyasu’s method. Using cryo-sectioned biological specimens, target proteins with excellent antigenicity were first immunolabeled for confocal analysis, and then the same tissue sections were further processed for electron microscopy, which provided a well-preserved ultrastructure comparable to that obtained using conventional electron microscopy. Moreover, this method does not require specifically designed correlative light and electron microscopy (CLEM) devices but rather employs conventional confocal and electron microscopes; therefore, it can be easily applied in many biomedical studies.  相似文献

Adult zebrafish is a well-known small animal model for studying heart regeneration. Although the regeneration of scars made by resecting the ventricular apex has been visualized with histological methods, there is no adequate imaging tool for tracking the functional recovery of the damaged heart. For this reason, high-frequency Doppler echocardiography using dual mode pulsed wave Doppler, which provides both tissue Doppler (TD) and Doppler flow in a same cardiac cycle, is developed with a 30 MHz high-frequency array ultrasound imaging system. Phantom studies show that the Doppler flow mode of the dual mode is capable of measuring the flow velocity from 0.1 to 15 cm s−1 with high accuracy (p-value = 0.974 > 0.05). In the in vivo study of zebrafish, both TD and Doppler flow signals were simultaneously obtained from the zebrafish heart for the first time, and the synchronized valve motions with the blood flow signals were identified. In the longitudinal study on the zebrafish heart regeneration, the parameters for diagnosing the diastolic dysfunction, for example, E/Em < 10, E/A < 0.14 for wild-type zebrafish, were measured, and the type of diastolic dysfunction caused by the amputation was found to be similar to the restrictive filling. The diastolic function was fully recovered within four weeks post-amputation.  相似文献

A facile and rapid method for the synthesis of porous gold (PAu) electrodes on cellulose-filters is proposed. Morphology and active surface area of the electrode greatly influence the electrochemical properties of sensor. Compared to smooth electrode and glassy carbon, PAu and Pt/PAu electrode showed high roughness due to porous structure, which is helpful in electrochemical sensing of target molecules. The electrochemical sensing performance of Pt/PAu electrode was evaluated to detect hydrogen peroxide, glucose, and perchloric acid. The amperometric results with various concentrations of target molecules showed potential applications of Pt/PAu electrode in sensitive and effective chemical sensors.  相似文献

Fluorescent labeling is widely used to investigate the structural stability and changes to DNA nano- and microstructures. Despite this, the conventional method for observing DNA structures has several limitations in terms of cost-efficiency. This paper introduces a DNA spherical particle stained with DNA intercalating dyes (SYBR Green and SYTOX Orange) as tracers and reports the interaction between multiple dyes. The interference between the dyes was analyzed in terms of Förster resonance energy transfer (FRET) and competition. The changes in the fluorescence intensity by FRET were uniform, regardless of the sequence. The competition effect could occur when several dyes were added simultaneously. These properties are expected to help in the design of multicolor tracers in bioimaging and environmental applications.  相似文献

A novel petrosiacetylene analog (petrosiacetylene E) has been isolated from the Korean marine sponge Petrosia sp., along with petrosiacetylene A, B and C. Their structures were elucidated on the basis of spectroscopic methods and the stereochemistry of the new compound was determined by using the modified Mosher’s method. Petrosiacetylene E showed higher cytotoxicity against five human cancer cell lines than petrosiacetylene A and B, presumably due to the additional hydroxy group located at C-16.  相似文献

Fibrous poly(1‐butene) membranes were prepared from solutions using mixed solvents via electrospinning. The electrospun fibrous poly(1‐butene) membranes were stretched at elongations of 8, 16 and 24%, respectively. Scanning electron microscopy and stress–strain analysis showed that the deformation was elastic at an elongation of 8%, while plastic deformation occurred above 16%. The crystalline transformation induced by mechanical stretching was investigated using wide‐angle X‐ray diffraction. The poly(1‐butene) membrane stretched at 24% exhibited decreased diffraction intensity at 2θ = 11.8° and 18.3°, corresponding to the (200) and (213) crystalline reflections, indicating that crystalline transformation from metastable form II to either stable form I or I′ occurred due to mechanical stretching.
