ABSTRACT

For the first time, we have fabricated insect-derived cell sheets by using temperature-responsive culture surfaces having a phase-transition temperature below 25°C. We prepared the temperature-responsive cell culture surfaces (tissue culture polystyrene, TCPS) by grafting a copolymeric gel consisting of hydrophobic N-tert-butylacrylamide (tBAAm) and N-isopropylacrylamide (IPAAm) units. First, to characterize the hydrophilic and hydrophobic properties of the copolymeric gel-grafted surfaces, static water contact angles of each surface were measured at various temperatures. By increasing the amount of tBAAm in the grafted copolymeric gel, the transition temperature of the gel was shifted to lower temperatures. At 25°C, the grafted copolymeric gel was dehydrated, and the insect-derived cells (AeAl2 cells) adhered on all the copolymeric gel-grafted surfaces. At 20°C, AeAl2 cells cannot adhere on the P(IPAAm-1.62tBAAm)-TCPS surface (the initial molar ratio of IPAAm and tBAAm (tBAAm?=?1.62 mol%)) better than on other surfaces (TCPS and tBAAm?=?4.88, 8.17 mol%). These two findings implied that the lower critical solution temperature of the copolymeric gel-grafted-TCPS existed from 20°C to 25°C. The laminin-coated P(IPAAm-1.62tBAAm)-TCPS surface showed temperature-dependent cell attachment and detachment properties, while AeAl2 cells were not detached from the extracellular matrix uncoated P(IPAAm-1.62tBAAm)-TCPS surface. AeAl2 cells and insect muscle cells were harvested as the respective sheets.  相似文献

Pulse tube refrigerators do not have moving parts in the cold section, and they have low vibration, high reliability, and long life. The expander in refrigerators typically has an inverted U or coaxial shape because this attains a wider absorber area, lower height, and compactness. However, the performance of a Stirling-type pulse tube refrigerator is inferior to that of a Stirling refrigerator. Cooling characteristics of the pulse tube refrigerator greatly depend on the shape of the expander. In this study, an inertance-type refrigerator, which uses ambient air for the working gas, was developed to examine the effect of expander shape. This refrigerator model with changeable expander operated with a Stirling cycle, and it was composed of a reciprocating compressor, after-cooler, regenerator, absorber, pulse tube, hot-end, and inertance tube with reservoir. The following expander shapes were tested: in-line, L shape, L-L shape, and coaxial shape. The effect of expander shape on cooling capacity was examined experimentally and numerically using the model pulse tube refrigerator. The results of experiments showed that the L shape expander had the highest performance and the coaxial expander had the lowest performance. In addition, the characteristics of the gas flow in each expander were confirmed by fluid dynamics analysis.  相似文献

Many additives are commercially used to add more favorable qualities to films. The bleeding process by which the additive in a film comes to the surface is considered. A new bleeding model of additives in a polypropylene film under atmospheric pressure was investigated. Solubility and diffusion are found to be important for explaining this bleeding process. It was found that the experimental results were explained more precisely by assuming a two‐step transport process between the crystalline regions and the amorphous ones. The solubilities and diffusion coefficients of UV‐stabilizers such as 2‐(2H‐benzotriazol‐2‐yl)‐4‐(1,1,3,3‐tetramethylbutyl)phenol and 2‐(2H‐benzotriazol‐2‐yl)‐4‐methylphenol were determined at 40°C. The difference between the saturation solubilities and the diffusion coefficients of UV‐stabilizers was discussed by comparing with the results of molecular dynamics (MD) simulation. © 2007 Wiley Periodicals, Inc. Abstract— The growth behaviour of small fatigue cracks has been investigated on aluminum alloy 7075-T6 at stress ratios R of 0, −1 and −2. The effects of stress ratio are discussed with special interest in the stage I region of small crack growth. Cracks which initiated at R =−1 and −2, grew by a stage I mechanism up to a certain depth followed by stage II crack growth. The stage I to stage II transition occurred under a constant maximum stress intensity factor which was approximately consistent with the threshold effective stress intensity range, λ K eff,th, for large cracks. At R = 0, on the other hand, stage I crack growth was not observed because of crack initiation at inclusions. Small cracks grew more rapidly than large cracks subjected to the same nominal stress intensity ranges at all the stress ratios, and they grew below the threshold stress intensity range, λ K th, for large cracks. Stage I cracks, in particular, showed much higher growth rates than large cracks and grew even below λ K eff,th. It is suggested that stage II crack growth rates should be characterized in terms of an effective stress intensity range, while a micromechanics approach will be necessary to evaluate stage I crack growth rates.  相似文献

A 4-nitrophenol (4-NP)-degrading bacterium was isolated from activated sludge and identified as a Rhodococcus sp. This bacterium, designated as strain PN1, could utilize 4-NP as a sole carbon, nitrogen and energy source. Degradation tests of 4-NP using cell suspensions of strain PN1 revealed that the degradation was induced by 4-NP and that 4-nitrocatechol (4-NC) was one of the metabolites. A gene library was constructed from the total DNA of strain PN1 and introduced into Rhodococcus rhodochrous ATCC 12674. Two recombinant strains showed 4-NP hydroxylase activity, and a 9.1-kb DNA fragment encoding the activity was isolated from one of the strains. In addition, a 2.4-kb smaller fragment expressing the activity was subcloned from the 9.1-kb fragment and sequenced. The sequence analysis showed that the fragment encodes a two-component 4-NP hydroxylase, the predicted amino acid sequence of which exhibits significant similarity to those of phenol hydroxylases and 4-hydroxyphenylacetate 3-hydroxylases belonging to the two-component flavin diffusible monooxygenase (TC-FDM) family proposed by Galán et al. (J. Bacteriol., 182, 627-636, 2000).  相似文献

Despite the structural similarities between cholesterol oxidasefrom Streptomyces and that from Brevibacterium, both enzymesexhibit different characteristics, such as catalytic activity,optimum pH and temperature. In attempts to define the molecularbasis of differences in catalytic activity or stability, substitutionsat six amino acid residues were introduced into cholesteroloxidase using site-directed mutagenesis of its gene. The aminoacid substitutions chosen were based on structural comparisonsof cholesterol oxidases from Streptomyces and Brevibacterium.Seven mutant enzymes were constructed with the following aminoacid substitutions: L117P, L119A, L119F, V145Q, Q286R, P357Nand S379T. All the mutant enzymes exhibited activity with theexception of that with the L117P mutation. The resulting V145Qmutant enzyme has low activities for all substrates examinedand the S379T mutant enzyme showed markedly altered substratespecificity compared with the wild-type enzyme. To evaluatethe role of V145 and S379 residues in the reaction, mutantswith two additional substitutions in V145 and four in S379 wereconstructed. The mutant enzymes created by the replacement ofV145 by Asp and Glu had much lower catalytic efficiency forcholesterol and pregnenolone as substrates than the wild-typeenzyme. From previous studies and this study, the V145 residueseems to be important for the stability and substrate bindingof the cholesterol oxidase. In contrast, the catalytic efficiencies(kcat/Km) of the S379T mutant enzyme for cholesterol and pregnenolonewere 1.8- and 6.0-fold higher, respectively, than those of thewild-type enzyme. The enhanced catalytic efficiency of the S379Tmutant enzyme for pregnenolone was due to a slightly high kcatvalue and a low Km value. These findings will provide severalideas for the design of more powerful enzymes that can be appliedto clinical determination of serum cholesterol levels and assterol probes.  相似文献

Propionibacterium acidipropionici TISTR442 produced the highest amount of 5-aminolevulinic acid (ALA) when cultivated in medium supplemented with glycine at 18g/l. ALA production correlated with ALA synthase activity, whereas ALA dehydratase activity was maintained at a low level. ALA yield reached 405mg/l after prolonged cultivation for 1 month.  相似文献

Since the chemical synthesis of vitamin B12 requires more than 70 steps, the production of vitamin B12 has been achieved by microorganism fermentation with additional brief chemical modifications. In an effort to increase the productivity of vitamin B12, we tried to express 10 genes belonging to the hem, cob and cbi gene families involved in the synthesis of vitamin B12 in Propionibacterium freudenreichii, which is a known producer of vitamin B12. In a recombinant P. freudenreichii clone that harbored the expression vector containing a cobA, cbiLF, or cbiEGH, we obtained an increase in vitamin B12 production of 1.7-, 1.9-, and 1.5-fold higher, respectively, than that in the microorganism without any cloned genes in the expression vector pPK705. The cobU and cobS genes caused a slight increase in the production of vitamin B12. Furthermore, we achieved multigene expression in P. freudenreichii. In a recombinant P. freudenreichii clone that harbored an exogenous gene, hemA, from Rhodobacter sphaeroides and endogenous hemB and cobA genes, we successfully achieved the production of about 1.7 mg/l vitamin B12, 2.2-fold higher than that produced by P. freudenreichii harboring pPK705.  相似文献
