Resveratrol is a biologically active diphenolic compound exerting multiple beneficial effects in the organism, including anti-diabetic properties. This action is, however, not fully elucidated. In the present study, we examined effects of resveratrol on some parameters related to insulin signaling, and also on diabetes-associated dysregulation in Goto-Kakizaki (GK) rats with congenital type 2 diabetes. Resveratrol was given at the dose of 20 mg/kg b.w. for 10 weeks. It was shown that the expression and phosphorylation levels of insulin receptor in the skeletal muscle of GK rats were significantly decreased, compared with control animals. However, these changes were totally prevented by resveratrol. Liver expression of the insulin receptor was also reduced, but in this case, resveratrol was ineffective. Resveratrol was also demonstrated to significantly influence parameters of insulin binding (dissociation constant and binding capacity) in the skeletal muscle and liver. Moreover, it was shown that the expression levels of proteins related to intracellular glucose transport (GLUT4 and TUG) in adipose tissue of GK rats were significantly decreased. However, treatment with resveratrol completely abolished these changes. Resveratrol was found to induce normalization of TUG expression in the skeletal muscle. Blood levels of insulin and GIP were elevated, whereas proinsulin and GLP-1 diminished in GK rats. However, concentrations of these hormones were not affected by resveratrol. These results indicate that resveratrol partially ameliorates diabetes-associated dysregulation in GK rats. The most relevant finding covers the normalization of the insulin receptor expression in the skeletal muscle and also GLUT4 and TUG in adipose tissue.  相似文献

In this paper, with the use of Euler‐Lagrange formalism, it has been proven which joint variables can influence entries of the inertia matrix and the gravity vector of dynamics of a manipulator relative to the kinematic structure of the manipulator. Some formulas, alternative to well‐known ones, are presented that allow us to compute the inertia matrix and the gravity vector without using a trace operator. These formulas are computationally inexpensive and provide an inspiration for interesting interpretations. Complexity indices are defined that are able to evaluate the complexity of dynamics of manipulators based on their kinematics. A key role of the accumulated pseudoinertia matrix of the manipulator has been shown. © 2002 Wiley Periodicals, Inc.  相似文献

The effect of the isoflavone, genistein, on the lipid metabolism of ovariectomized rats was studied. Three types of experiments were performed. In the first one, the rats were fed diets supplemented with 0.01 or 0.1% of genistein for 14 days. In the second and third experiments, the direct effect of genistein on the liver and fat tissue were measured respectively by means of liver perfusion or incubation of isolated adipocytes with the isoflavone. Genistein in food significantly decreased blood serum and muscle triglyceride concentrations and increased the level of free fatty acids in serum. Serum free cholesterol was diminished and liver cholesterol was enhanced after genistein ingestion. When genistein acted directly on the liver during perfusion, a smaller incorporation of 14C-glucose into lipids was observed, and in parallel a greater output of free fatty acids into the medium was noticed. These changes were accompanied by diminution of the liver triglyceride contents. Genistein, acting on the adipocytes strongly depressed both basal and insulin-induced lipid synthesis, when glucose was used as a substrate. The effect of the isoflavone alone on the lipolysis in the adipocytes was negligible. However, it intensified lipolysis induced by epinephrine. The results obtained let us conclude that genistein in food can reduce the fattening processes in ovariectomized rats. This effect of genistein may be attributed, at least in part, to its direct influence on lipid metabolism in the liver and adipose tissue.  相似文献

Montmorillonite (MMT) was organically modified with tributyl citrate (TBC). Organoclays (OMMTs) were processed with diisononyl phthalate (DINP)‐plasticized polyvinyl chloride (PVC) to form polymer nanocomposites. The produced composite materials showed a contradictory change in properties to that expected of a layered silicate nanocomposite, with a decreased E‐modulus and increased gas permeability compared with a material without OMMT. It was experimentally shown that the TBC modifier was extracted from the OMMT and was dispersed in the PVC/DINP matrix, whereupon the OMMT collapsed and formed micrometer‐sized agglomerates. Further investigation revealed that TBC has a significant effect on the gas permeability and the E‐modulus, even at low additions to a DINP‐plasticized PVC. A PVC nanocomposite with the TBC acting as both the OM for MMT and as the primary plasticizer was produced. This material showed a significantly increased E‐modulus as well as a decrease in gas permeability, confirming that it is possible to develop a nanocomposite based on plasticized PVC, if both the organo‐modification of the MMT and the formulation of the matrix are carefully selected. © 2015 Wiley Periodicals, Inc. We describe a new approach to measuring DNA hybridization using surface plasmon-coupled emission (SPCE). Excited fluorophores are known to couple with surface oscillations of electrons in thin metal films, typically 50 nm thick silver on a glass prism. These surface plasmons then radiate into the glass at a sharply defined angle determined by the emission wavelength and the optical properties of the glass and metal. This radiation has the same spectral profile as the emission spectrum of the fluorophores. We studied the emission due to Cy3-labeled DNA oligomers bound to complementary unlabeled oligomers which were themselves bound to the metal surface. Hybridization resulted in SPCE due to Cy3-DNA into the prism. Directional SPCE was observed whether the sample was illuminated from the sample side or through the glass substrate at the surface plasmon angle for the excitation wavelength. A large fraction of the total potential emission is coupled to the surface plasmons resulting in improved sensitivity. When illuminated through the prism at the surface plasmon angle, the sensitivity is increased due to the enhanced intensity of the resonance evanescent field. It is known that SPCE depends on proximity to the silver surface. As a result, changes in emission intensity are observed due to fluorophore localization even if hybridization does not affect the quantum yield of the fluorophore. The use of SPCE resulted in suppression of interfering emission from a noncomplementary Cy5-DNA oligomers due to weaker coupling of the more distant fluorophores with the surface plasmons. We expect SPCE to have numerous applications to nucleic acid analysis and for the measurement of bioaffinity reactions.  相似文献

Abstract

Significant evidence from epidemiological investigations showed that dietary polyphenols might manage and prevent type 2 diabetes (T2D). This review summarizes human studies and clinical trials of polyphenols as anti-diabetic agents. Polyphenols from coffee, guava tea, whortleberry, olive oil, propolis, chocolate, red wine, grape seed, and cocoa have been reported to show anti-diabetic effects in T2D patients through increasing glucose metabolism, improving vascular function as well as reducing insulin resistance and HbA1c level. However, individual flavonoid or isoflavonoid compounds appear to have no therapeutic effect on diabetes, based on the limited clinical data. Preliminary clinical trials provided evidence that resveratrol had anti-diabetic activity in humans by improving glycemic control in subjects with insulin resistance. Besides, anthocyanins exhibited anti-diabetic properties by reducing blood glucose and HbA1c levels or the improvement of insulin secretion and resistance. The structure-activity relationship of polyphenols as anti-diabetic agents in humans has been rarely reported.  相似文献

In this paper a pre-control form of the generalized Campbell–Baker–Hausdorff–Dynkin (gCBHD) formula is presented. This form is dedicated to nonholonomic (affine) systems often encountered in robotics. The simplest possible expression for the pre-control gCBHD is obtained as control-dependent functions pre-multiply elements of the Ph. Hall basis. Algorithmic aspects of deriving the formula are highlighted. An application of the pre-control form of the gCBHD formula in motion planning of nonholonomic systems is also reported.  相似文献

Environmental and economic reasons make the use of bioplastics and biocomposites increasingly coveted in sectors other than packaging. Recycling of all wasted or rejected durable plastics is highly desired and biobased plastics are no exception. Therefore, the investigation of pre‐ and post‐consumer recycling of products made from biobased plastics is of great interest. Polylactic acid (PLA) and its blends have been chosen for this study because it is an excellent representative of mass‐produced bioplastics for industrial applications. As part of the “Sustainable Recycling of ‘Green’ Plastics” project, the current study addresses the durability issues related to the reprocessing and post‐consumer recycling of a PLA virgin resin and two commercially available blends of PLA namely one with polycarbonate (PC) and one with polyethylene (PE). The materials were investigated using methods that simulate post‐processing and post‐consumer recycling. Accelerated ageing was performed at elevated temperature and humidity to simulate the usage period of the materials. The materials were analyzed using differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), and their mechanical strength was evaluated by tensile and impact testing. The flow properties of the materials were characterized by the melt flow index (MFI). Multiple processing of pure PLA did not affect the impact strength or the glass transition temperature (Tg), but caused crystallization and increase in the MFI, indicating that degradation occurred during processing. DSC thermograms of the blends revealed that the components in the blends were not miscible. Multiple processing of the blends did not significantly affect the elastic modulus of the materials, but affected the elongation at break. The results indicated that multiple processing of the PLA/HDPE blend caused increased dispersion and thus increased elongation at break, while the dominating mechanism in the PLA/PC blend was degradation that caused a decrease in elongation at break. Post‐consumer recycling of the PLA/PC blend was simulated and the results clearly showed that ageing corresponding to one year of use caused a significant degradation of PLA. Pure PLA was severely degraded after only one ageing cycle. Although the PLA/PC blend showed some improved mechanical properties and resistance to degradation compared with pure PLA, one ageing cycle still caused a severe degradation of the PLA and even the PC was degraded as indicated by the formation of small amounts of bisphenol A. © 2016 Wiley Periodicals, Inc.
