Insulin is a peptide hormone with many physiological functions, besides its use in diabetes treatment. An important role of insulin is related to the wound healing process—however, insulin itself is too sensitive to the external environment requiring the protective of a nanocarrier. Polymer-based nanoparticles can protect, deliver, and retain the protein in the target area. This study aims to produce and characterize a topical treatment for wound healing consisting of insulin-loaded poly-DL-lactide/glycolide (PLGA) nanoparticles. Insulin-loaded nanoparticles present a mean size of approximately 500 nm and neutral surface charge. Spherical shaped nanoparticles are observed by scanning electron microscopy and confirmed by atomic force microscopy. SDS-PAGE and circular dichroism analysis demonstrated that insulin preserved its integrity and secondary structure after the encapsulation process. In vitro release studies suggested a controlled release profile. Safety of the formulation was confirmed using cell lines, and cell viability was concentration and time-dependent. Preliminary safety in vivo assays also revealed promising results. 相似文献

The focus of this triple‐blind study was on evaluating the effect of chitosan combined with Dysphania ambrosioides (A) extract on the bone repair process in vivo. In total, 60 male Wistar rats (Rattus norvegicus albinus) weighing between 260 and 270 g were randomly selected for this study and distributed into four groups (n = 15). Group C (chitosan), Group CA5 (chitosan + 5% of D. ambrosioides), Group CA20 (chitosan + 20% of D. ambrosioides), and Group CO (Control‐Blood clot). In each animal, bone defects measuring 2 mm in diameter were performed in both tibias for placement of the substances. After 7, 15, and 30 days, the animals were sedated and sacrificed using the cervical dislocation technique and the tissues were analyzed under optical microscope relative to the following events: inflammatory infiltrate, necrosis, osteoclasts, osteoblasts, fibroblasts, periosteal, and endosteal bone formation. The data were evaluated to verify distribution using the Kolmogorov–Smirnov test, and variance, using the Levene test; as distribution was not normal, data were subjected to the Kruskal–Wallis and Dunn nonparametric tests (p < .05). A significant inflammatory infiltrate was observed in Group CA5 (p = .008) in the time interval of 7 days, and in Group C at 15 (p = .009) and 30 (p = .017) days. Osteoblastic activity was more significant in Group CA20 (p = .027) compared with CA5 in the time interval of 7 days. Group CA20 demonstrated a significantly higher endosteal and periosteal bone formation value in the time interval of 7 (p = .013), 15 (p = .004), and 30 days (p = .008) compared with the other groups. The null hypothesis was refuted, bone regeneration was faster in spheres with an association of chitosan and 20% extract, and complete bone repair occurred clinically at 15 days and histologically at 30 days. 针对复合板结构中弯曲板波谐振频率计算相对复杂的问题,建立并分析了基于弯曲板波延迟线振荡器的理论模型,计算得到A0(0阶反对称)模式板波的谐振频率为41 MHz;为了验证理论分析的结果,利用有限元软件ANSYS建立了基于弯曲板波的延迟线振荡器的3D有限元模型,并对其进行了谐响应分析以及瞬态动力学分析。谐响应分析得到A0模式的谐振频率为37.2 MHz,与理论计算结果的相对误差为9.3%;瞬态动力学分析得到A0模式的谐振频率为39.1 MHz,与理论计算结果的相对误差为4.6%。计算和仿真为基于弯曲板波的延迟线振荡器的结构设计提供了重要的依据。 相似文献

A direct carbon fuel cell (DCFC) is a variation of the molten carbonate fuel cell (MCFC) which converts the chemical energy of carbon directly into electrical energy. Thus, the energy conversion efficiency is very high and correspondingly CO2 emission is very low for given power output. DCFC as a high temperature fuel cell performs better at elevated temperatures (>800 °C) but because of the corrosive nature of the molten carbonates at elevated temperatures the degradation of cell components becomes an issue when DCFC is operated for an extended period of time.We explored the DCFC performance at lower temperatures (at 700 °C and less) using different sources of carbon, different compositions of electrolytes and some additives on the cathode surface to increase catalytic activity. Experiments showed that with petroleum coke as a fuel at low temperatures the ternary eutectic (43.4 mol % Li2CO3 - 31.2 mol% Na2CO3 - 25.4 mol % K2CO3) spiked by 20 wt % Cs2CO3 performed better than any binary or ternary eutectics described in the published work by other researchers. Maximum power output achieved at 700 °C was 49 mW/cm2 at a current density of 78 mA/cm2 when modified cathode was fed with O2/CO2 gases. 相似文献

Cystoliths are amorphous calcium carbonate bodies that form in the leaves of some plant families. Cystoliths are regularly distributed in the epidermis and protrude into the photosynthetic tissue, the mesophyll. The photosynthetic pigments generate a steep light gradient in the leaf. Under most illumination regimes the outer mesophyll is light saturated, thus the photosynthetic apparatus is kinetically unable to use the excess light for photochemistry. Here we use micro‐scale modulated fluorometry to demonstrate that light scattered by the cystoliths is distributed from the photosynthetically inefficient upper tissue to the efficient, but light deprived, lower tissue. The results prove that the presence of light scatterers reduces the steep light gradient, thus enabling the leaf to use the incoming light flux more efficiently. MicroCT and electron microscopy confirm that the spatial distribution of the minerals is compatible with their optical function. During the study we encountered large calcium oxalate druses in the same anatomical location as the cystoliths. These druses proved to have similar light scattering functions as the cystoliths. This study shows that certain minerals in the leaves of different plants distribute the light flux more evenly inside the leaf. 相似文献

The urgent need for new antibiotics poses a challenge to target un(der)exploited vital cellular processes. Thymidylate biosynthesis is one such process due to its crucial role in DNA replication and repair. Thymidylate synthases (TS) catalyze a crucial step in the biosynthesis of thymidine 5‐triphosphate (TTP), an elementary building block required for DNA synthesis and repair. To date, TS inhibitors have only been successfully applied in anticancer therapy due to their lack of specificity for antimicrobial versus human enzymes. However, the discovery of a new family of TS enzymes (ThyX) in a range of pathogenic bacteria that is structurally and biochemically different from the “classic” TS (ThyA) has opened the possibility to develop selective ThyX inhibitors as potent antimicrobial drugs. Here, the interaction of the known inhibitor 5‐(3‐octanamidoprop‐1yn‐1yl)‐2′‐deoxyuridine‐5′‐monophosphate ( 1 ) with Mycobacterium tuberculosis ThyX enzyme is explored using molecular modeling starting from published crystal structures, with further confirmation through NMR experiments. While the deoxyuridylate (dUMP) moiety of compound 1 occupies the cavity of the natural substrate in ThyX, the rest of the ligand (the “5‐alkynyl tail”) extends to the outside of the enzyme between two of its four subunits. The hydrophobic pocket that accommodates the alkyl part of the tail is formed by displacement of Tyr 44.C, Tyr 108.A and Lys 165.A. Changes to the resonance of the Lys 165 NH3 group upon ligand binding were monitored in a titration experiment by 2D HISQC NMR. Guided by the results of the modeling and NMR studies, and inspired by the success of acyclic antiviral nucleosides, compounds where a 5‐alkynyl uracyl moiety is coupled to an acyclic nucleoside phosphonate (ANP) were synthesized and evaluated. Of the compounds evaluated, sodium (6‐(5‐(3‐octanamidoprop‐1‐yn‐1‐yl)‐2,4‐dioxo‐3,4‐dihydropyrimidin‐1(2H)‐yl)hexyl)phosphonate ( 3 e ) exhibited 43 % of inhibitory effect on ThyX at 50 μM . While only modest activity was achieved, this is the first example of an ANP inhibiting ThyX, and these results can be used to further guide structural modifications to this class to develop more potent compounds with potential application as antibacterial agents acting through a novel mechanism of action. 相似文献

Composites of blast furnace slag (BFS) based cement mortar reinforced with vegetable fibres are presented. Roofing components are produced with these composites through a simple and low-energy consuming method, including ordinary vibration and curing in a wet chamber. Composites reinforced with eucalyptus pulp, coir fibres and with a mixture of sisal fibre and eucalyptus pulp gave a suitable performance, with compressive strength higher than 20 MPa and modulus of rupture (MOR) higher than 3 MPa. The performance of tiles made with these composites is in accordance with international requirements, with maximum load higher than 450 N, in wet conditions. 相似文献

The upgrading potential of quince (Cydonia oblonga Miller) wastes was evaluated for production of fiber-rich powders with useful functional and physiological properties. As a first approach, three products were obtained by applying different drying conditions without or with a previous extraction either with water or ethanol. Chemical compositions as well as physical and functional properties of the isolated fractions were evaluated in relation to water or oil absorption for characterization. The products obtained presented interesting hydration properties comparable to those reported for citrus and apple pulps. At the same time, all dried fractions showed high spontaneous water absorption rate in the kinetics assay. Oil absorption seemed to essentially depend on the microstructural characteristics of the fiber powders, whereas parameters involving water absorption were really determined by the material’s hydrophilicity. Specific volume, which was also in part a direct function of structural differences, was associated to the ability for oil uptake. 相似文献
