We report the anti-osteosarcoma and anti-osteosarcoma stem cell (OSC) properties of a nickel(II) complex, 1 . Complex 1 displays similar potency towards bulk osteosarcoma cells and OSCs, in the micromolar range. Notably, 1 displays similar or better OSC potency than the clinically approved platinum(II) anticancer drugs cisplatin and carboplatin in two- and three-dimensional osteosarcoma cell cultures. Mechanistic studies revealed that 1 induces osteosarcoma cell death by necroptosis, an ordered form of necrosis. The nickel(II) complex, 1 triggers necrosome-dependent mitrochondrial membrane depolarisation and propidium iodide uptake. Interestingly, 1 does not evoke necroptosis by elevating intracellular reactive oxygen species (ROS) or hyperactivation of poly ADP ribose polymerase (PARP-1). ROS elevation and PARP-1 activity are traits that have been observed for established necroptosis inducers such as shikonin, TRAIL and glutamate. Thus the necroptosis pathway evoked by 1 is distinct. To the best of our knowledge, this is the first report into the anti-osteosarcoma and anti-OSC properties of a nickel complex.  相似文献

A dynamic modeling approach was used to estimate in-situ model parameters, which describe the degradation of methyl tert-butyl ether (MTBE) in a laboratory packed bed reactor. The measured dynamic response of MTBE pulses injected at the reactor's inlet was analyzed by least squares and parameter response surface methodologies. Response surfaces were found to be statistically significant and thus suitable for estimating the global minimum as well as the 95% parameter uncertainty regions. Electrode biasing system was designed, constructed, and installed on the IR-T1 tokamak, and then biasing experiments were carried out. Also, using a Mach probes the effects of radial electric field (produced by biased electrode) on the poloidal and toroidal components of the edge plasma velocity were investigated. The results showed an increase in both toroidal and poloidal components of the edge plasma velocity during biasing regime. Results compared and discussed. During positive biasing, increased Er tends to slow the poloidal rotation in the electron diamagnetic drift direction, i.e., to speed up rotation in the ion diamagnetic drift direction. An increased toroidal rotation velocity has the opposite effect on the poloidal rotation.  相似文献

This paper explains and evaluates the potential and limitations of conducting cross-well radar (CWR) in sandy soils. Implementing the experiment and data collection in the absence of any scattering object, and in the presence of an acrylic plate [a representative of dielectric objects, such as dense nonaqueous phase liquid (DNAPL) pools, etc.], as a contrasting object in a water-saturated soil is also studied. To be able to image the signature of any object, more than one pair of receiving and transmitting antennas are required. The paper describes a method to achieve repeatable, reliable, and reproducible laboratory results for different transmitter-receiver combinations. Different practical methods were evaluated for collecting multiple-depth data. Similarity of the corresponding results and problems involved in each method are studied and presented. The data show that the frequency response of a saturated coarse-grained soil is smooth due to the continuous and dominant nature of water in saturated soils. The repeatability and potential symmetry of patterns across some borehole axes provide a valuable tool for validation of experimental results. The potential asymmetry across other borehole axes is used as a tool to evaluate the strength of the perturbation on the electromagnetic field due to hidden objects and to evaluate the feasibility of detecting dielectric objects (such as DNAPL pools, etc.) using CWR. The experimental simulation of this paper models a real-life problem in a smaller scale, in a controlled laboratory environment, and within homogeneous soils that are uniformly dry or fully water saturated, with a uniform dielectric property contrast between the inclusion and background. The soil in the field will not be as homogeneous and uniform. The scaling process takes into consideration that as the size is scaled down; the frequency needs to be scaled up. It is noteworthy that this scaling process needs to be extensively studied and validated for future extension of the models to real-field applications. For example, to extend the outcome of this work to the real field, the geometry (antenna size, their separation and inclusion size) needs to be scaled up back to the field size, while soil grains will not. Therefore, soil, water, and air coupling effects and interactions observed at the laboratory scale do not scale up in the field, and may have different unforeseen effects that require extensive study.  相似文献

Ralstonia eutropha was able to degrade p-nitrophenol (PNP) at concentrations ranging from 5 to 15mg l ?1 in the presence of phenol which was kept at the constant concentration of 200 mg l ?1. More than 90% of phenol was degraded within 30 h and in the absence of PNP. While in this time period and in the presence of 15 mg l ?1 less than 30% of phenol was degraded and PNP removal ability of the test bacterium was about 20%. Kissiris as a natural source of silicon dioxide having a very rigid structure with many micropores irregularly distributed throughout its surface was used to evaluate effectiveness of the cell immobilization using a Kissiris-immobilized cell bioreactor [ICB]. By applying phenol-feeding regime in the ICB operated in a batch recycling mode, simultaneous degradation of phenol in total amount of 1,000 mg l ?1 with 15 mg l ?1 PNP was achieved within 40 h.  相似文献

In this contribution we presented the first biasing experiments performed on the IR-T1 tokamak. For this purpose, a movable biasing system was designed, constructed, and installed, and then the positive voltage applied to an electrode inserted inside the tokamak limiter and the plasma current and also the plasma horizontal displacement in the absence and presence of the biased electrode based on the multipole moments and magnetic probes techniques were measured. Measurements result has shown a modification of plasma confinement by decreasing the plasma displacement.  相似文献

Nitrification is an acidity producing process. It has been shown theoretically that the diffusional resistance to the transport in the biofilm of the inorganic carbon species as affected by the acidity production in a nitrifying biofilm gives rise to a decreased pH in the interior of the biofilm. These theoretical results have been verified on biofilms developed on the surfaces of a rotating drum under well controlled laboratory conditions. The results show clearly the drop in pH as predicted by theory. The phenomenon can give rise to unexpected effects on the performance of nitrifying biofilms, when most of the bacteria work under a much lower pH than the pH measured in the bulk water.  相似文献

对3 mm厚的AZ31-O镁合金板和6061-T6铝合金板进行异种材料搅拌摩擦焊。实验中搅拌针的旋转速度为600~1400 r/min,前进速度为20~60 mm/min。结果表明,在旋转速度1000 r/min、前进速度40 mm/min的工艺条件下,可以获得无缺陷的焊接接头。组织观察发现搅拌区的晶粒尺寸要比基材区的明显小很多。在搅拌区形成了复杂的金属流动。显微硬度测试表明搅拌区的硬度分别是不均匀的。拉伸实验结果表明,接头的拉伸强度约为基材AZ31镁合金的76%,或6061铝合金的60%。接头拉伸断口SEM形貌观察表明为脆性断裂。  相似文献

Co-metabolic biodegradation of cis-dichloroethylene (cis-DCE) was investigated in a bench-scale fixed-film bioreactor inoculated with a mixed culture of methane oxidising bacteria. The aim of this work was to identify factors that affect the cis-DCE biodegradation. It was observed that the presence of methane was necessary to enhance the biodegradation of cis-DCE, but an excess of methane inhibited the cis-DCE removal. cis-DCE did not inhibit the methane biodegradation at concentrations up to 300 microg/L. Maximum cis-DCE removal was observed with a methane bulk concentration ranging from 0.2 to 0.7 mg/L. It was found that the activity of the biofilm was located in the upper 100 microm of the biofilm. On the basis of this study it is concluded that careful control of the oxygen and methane concentrations as well as of the biofilm thickness is necessary in order to optimise the biodegradation of cis-DCE in fixed film bioreactors.  相似文献

The varicella-zoster virus (VZV) genes ORF47 and ORF66 are predicted to encode serine/threonine protein kinases, which are homologs of herpes simplex virus 1 (HSV-1) UL13, and US3. When mutants were constructed by inserting stop codons into ORF47 and ORF66, the recombinants ROka47S and ROka66S, as well as intact ROka replicated in tissue culture. In contrast, inoculation of human thymus/liver or skin implants in SCID-hu mice showed that ORF47 protein was required for viral growth in human T cells and skin. Eliminating ORF66 expression inhibited VZV infectivity for T cells partially but did not impair replication in skin compared with ROka. Infectivity for T cells and skin was restored when ROka47S virus was complemented by insertion of ORF47 into a distant, noncoding site. The ORF47 gene product is the first VZV protein identified as necessary for T cell tropism. It also is essential for skin infectivity in vivo, as is glycoprotein C. Expression of ORF66 did not compensate for the absence of the ORF47 protein. The requirement for ORF47 expression in T cells and skin indicates that this gene product, which is dispensable in vitro, has a critical role within differentiated cells that are essential targets for VZV pathogenesis in vivo.  相似文献
