Sequestration of m2 receptors (muscarinic acetylcholine receptor m2 subtypes), which was assessed as loss of N-[3H]methylscopolamine ([3H]NMS) binding activity from the cell surface, was examined in COS 7 and BHK-21 cells that had been transfected with expression vectors encoding the m2 receptor and, independently, vectors encoding a G protein-coupled receptor kinase (GRK2) (beta-adrenergic receptor kinase 1) or a GRK2 dominant-negative mutant (DN-GRK2). The sequestration of m2 receptors became apparent when the cells were treated with 10(-5) M or higher concentrations of carbamylcholine. In this case, approximately 40% or 20-25% of the [3H]NMS binding sites on COS 7 or BHK-21 cells, respectively, were sequestered with a half-life of 15-25 min. In cells in which GRK2 was also expressed, the sequestration became apparent in the presence of 10(-7) M carbamylcholine. Approximately 40% of the [3H]NMS binding sites on both COS 7 and BHK-21 cells were sequestered in the presence of 10(-6) M or higher concentrations of carbamylcholine. When DN-GRK2 was expressed in COS 7 cells, the proportion of [3H]NMS binding sites sequestered in the presence of 10(-5) M or higher concentrations of carbamylcholine was reduced to 20-30%. These results indicate that the phosphorylation of m2 receptors by GRK2 facilitates their sequestration. These results are in contrast with the absence of a correlation between sequestration and the phosphorylation of beta-adrenergic receptors by the GRK2 and suggests that the consequences of phosphorylation by GRK2 are different for different receptors. 相似文献

Provision of Quality‐of‐Service (QoS) guarantees is an important and challenging issue in the design of integrated‐services
packet networks. Call admission control is an integral part of the challenge and is closely related to other aspects of networks
such as service models, scheduling disciplines, traffic characterization and QoS specification. In this paper we provide a
theoretical framework within which call admission control schemes with multiple statistical QoS guarantees can be constructed
for the Generalized Processor Sharing (GPS) scheduling discipline. Using this framework, we present several admission control
schemes for both session‐based and class‐based service models. The theoretical framework is based on recent results in the
statistical analysis of the GPS scheduling discipline and the theory of effective bandwidths. Both optimal schemes and suboptimal
schemes requiring less computational effort are studied under these service models. The QoS metric considered is loss probability.

A New Process to Make a Porous PTFE Structure from Aqueous PTFE Dispersion with the Help of Hydrogel
A novel method to make a porous material having relatively large cell diameter (200–300 m), which consisted of mainly poly(tetrafluoroethylene) (PTFE), was developed from aqueous PTFE dispersion by using the characteristics of hydrogel with the addition of carbon nanofiber (CNF). The porous material was produced as follows: firstly, an aqueous agar gel containing PTFE and CNF was prepared; secondly, the gel was freeze-dried; thirdly, the dried gel was heat-treated at 400°C where the agar was almost decomposed and PTFE became molten. The porous material showed electric conductivity (about 50 ), high porosity (about 96 vol%), and relatively uniform cell structures without shrinkage during freeze drying and heat treatment. While the method without CNF resulted in large shrinkage during heat treatment, meaning that CNF prevented the shrinkage. It was explained by the idea that the existence of rigid CNF, which was dispersed in the cell wall, prohibited the shrinkage of PTFE during heat treatment. It was unexpectedly found by SEM analysis that the porous materials had another macro-porous structure inside the cell wall, suggesting that the developed materials had a double porous structure. 相似文献

Selective modification by ozonation for the surface of polyvinyl chloride (PVC) was evaluated to separate PVC from the other plastics, polyethylene terephthalate (PET), polycarbonate (PC) and polymethyl methacrylate (PMMA), with almost the same density as PVC by the froth flotation process. Ozonation could selectively decrease the contact angles of flexible PVC from 87.5 degrees to 68.4 degrees and rigid PVC from 90.3 degrees to 66.9 degrees, whereas little decreases in the contact angle were observed for other plastics. This would be due to the replacement of the chloride group on the surface of PVC, into hydrophilic functional groups; carbonyl, carboxyl and ester group. The PVC was successfully separated from the other plastics by the froth flotation process after the selective surface modification by ozonation. 相似文献

We provide a formal framework for studying broadcasting schemes and design a family of schemes for broadcasting popular videos,
the greedy disk-conserving broadcasting (GDB) family. We analyze the resource requirements for GDB, i.e., the number of server
broadcast channels, the client storage space, and the client I/O bandwidth required by GDB. Our analysis shows that all of
our proposed broadcasting schemes are within a small factor of the optimal scheme in terms of the server bandwidth requirement.
Furthermore, GDB exhibits a tradeoff between any two of the three resources. We compare our scheme with a recently proposed
broadcasting scheme, skyscraper broadcasting (SB). With GDB, we can reduce the client storage space by as much as 50% or the
number of server channels by as much as 30% at the cost of a small additional increase in the amount of client I/O bandwidth.
If we require the client I/O bandwidth of GDB to be identical to that of SB, GDB needs only 70% of the client storage space
required by SB or one less server channel than SB does. In addition, we show that with small client I/O bandwidth, the resource
requirements of GDB are close to the minimum achievable by any disk-conserving broadcasting scheme. 相似文献

Acoustic emission (AE) and microseismic (MS) events are indicators of rock fracturing or damage as the rock is brought to failure at high stress. By capturing the AE/MS events, underground excavation induced rock mass degradation or damage can be located and evaluated. A better understanding of the extent and shape of the excavation damaged zone (EDZ) or yield zone around caverns helps to arrive at safe and economic design and construction of the caverns. For this purpose, one needs to understand the AE mechanism associated with the excavation process.In the present study, a coupled numerical method is used to study AE at the Kannagawa underground powerhouse cavern in Japan. Two codes, Fast Lagrangian Analysis of Continua (FLAC), a finite difference code and Particle Flow Code (PFC), a distinct element code, are coupled. The motive to apply the FLAC/PFC coupled approach is to take advantage of each modeling scheme while at the same time minimizing the requirement for computational resources. The coupling is realized through an exchange of displacements, velocities, and forces in each cycling step. The rock mass surrounding an AE sensor is modeled using PFC and the remaining rock mass is modeled with FLAC to consider the geological complexity and the excavation sequence. In this manner, the AE activities at AE sensor locations of the Kannagawa cavern were simulated and found to be in good agreement with field monitoring results. This approach takes account of stress redistribution and provides stress and displacement patterns in the rock mass that are consistent with AE observations for excavation design. The observed AE activities in the rock mass can thus be utilized to assess the effectiveness of the rock support system and the overall stability of the cavern. 相似文献

A two-dimensional numerical simulation of a spray flame formed in a laminar counterflow is presented, and the flamelet characteristics are studied in detail. The effects of strain rate, equivalence ratio, and droplet size are examined in terms of mixture fraction and scalar dissipation rate. n-Decane (C10H22) is used as a liquid spray fuel, and the droplet motion is calculated by the Lagrangian method without the parcel model. A one-step global reaction is employed for the combustion reaction model. The results show that there appear large differences in the trends of gaseous temperature and mass fractions of chemical species in the mixture fraction space between the spray flame and the gaseous diffusion flame. The gas temperature in the spray flame is much higher than that in the gaseous diffusion flame. This is due to the much lower scalar dissipation rate and the coexistence of premixed and diffusion-limited combustion in the spray flame. For the spray flames, gas temperature and mass fractions of chemical species are not unique functions of the mixture fraction scalar dissipation rate. This is because the production rate of the mixture fraction, namely evaporation rate of the droplets, in the upstream region is not in proportion to its transport-diffusion rate in the downstream region. The behavior shows marked differences as the strain rate decreases, the equivalence ratio increases, or the droplet size decreases. 相似文献
