In this study, we characterized polystyrene-binding peptides (PS-tags) that possess a specific binding affinity for hydrophilic polystyrene (phi-PS) plates. Both the FITC-labeled PS19-1 (RAFIASRRIKRP) and PS19-6 (RIIIRRIRR) peptides showed strong binding affinity for commercially available hydrophilic, but not hydrophobic, PS plates in the presence of the non-ionic surfactant Tween 20. The dissociation constants (Kd) of the PS19-1 and PS19-6 peptides for the hydrophilic PS-A plate were 169 and 86 nM, respectively, and the Kd of both peptides increased with the concentration of NaCl or urea. Based on adsorption yield and residual activity of glutathione S-transferase (GST) after fusion with the PS19-6 peptide or its variants, it was found that the basic amino acid in the PS-tags, i.e., Arg was essential for the strong binding affinity of PS-tags in both the peptide and peptide-fused protein forms The aliphatic amino acids in PS19-6 and PS19-6L, such as Ile or Leu, were also effective. Thus, a series of PS-tags that possess this unusual feature, especially the peptides PS19-6 (RIIIRRIRR) and PS19-6L (RLLLRRLRR), are potential candidate affinity peptide tags for site-specific immobilization of proteins onto hydrophilic PS plates, which show potential as solid supports for protein-based biochips.  相似文献

Various synthetic esters are widely applied as lubricating fluid to reduce friction and wear at tribological contact. Among them low viscous synthetic esters are expected to improve fuel efficiency by minimizing the fluid friction. These low viscous esters are composed of short-chain fatty acids. Therefore, low viscous synthetic esters are inherently polar molecules. Since efficiency of anti-wear additives decreases with increase of polarity of the base oil, new additive technology is requested.In this work, hydroxyalkyl phosphates [P(O)(OCHRCH2OH)3], and hydroxyalkyl phosphonates [P(OH)n(OCHRCH2OH)3−n, where n=1,2] are proposed as novel anti-wear additives for polar synthetic esters. The anti-wear additives are evaluated under the boundary conditions. The additives prevent wear in polar esters, in which conventional anti-wear additives do not work at all. Interestingly, effects of substituent in additive molecule on anti-wear properties are found. Alkyl and aryl derivatives reduce wear remarkably, whereas allyl derivatives exhibit poor results. It is speculated that the anti-wear inefiiciency of allylic compounds is due to auto-oxidation of the additives.A facile preparative method for hydroxyalkyl derivatives characterizes the present additive system. They are prepared in situ by simply mixing phosphonic acid and substituted epoxides. Flexiblity of lubricant design can be made possible by the present additive system.  相似文献

针对日本国鬼怒河流域小流量河流,通过对河床上附着生物膜的组成、底栖生物、细菌活性分析和AGP试验,研究了污水厂二级处理排放水对小流量河流生态的影响.研究表明,当污水厂二级处理排放水占河流流量比例较大时(研究河流所占比例为35%),同样会引发河流富营养化和影响河流水体的生态.因此,从保护水环境及其生态考虑,有必要对污水进行深度处理.  相似文献

In Japan, various countermeasures have been taken to improve the water quality of public waters such as rivers and lakes. Though water quality has improved, it is still insufficient. In summer, eutrophication is seen in lakes and inner bays, as well as rivers. As a countermeasure to prevent eutrophication, the removal treatment of nutrient salts such as nitrogen and phosphorus is done, in addition to organic substance elimination in the domestic sewerage system. This report will show the water quality characteristics of rivers and ponds in Japan It is considered that these investigative results are effective when the water quality improvement of the stabilization ponds where eutrophication occurs are examined in China.  相似文献

采用混凝-沉淀-过滤-消毒处理工艺处理洗浴排水,在水温10℃时,主要控制指标浊度、COD、BOD5和ABS平均平除率分别为98.1%,95.2%,93.3%和68.2%,出水水质完全满足中水控制指标要求,试验中发现出水浊度与COD和BOD5有着很好的相关性,可以出水浊度监测COD和BOD5指标,使运行管理简便、结果完全可靠。  相似文献

Algal cells of Dunaliella tertiolecta with a moisture content of 78.4 wt% were converted directly into oil by thermochemical liquefaction at around 300°C and 10 MPa. The oil yield was about 37% on an organic basis. The oil obtained at a reaction temperature of 340°C and holding time of 60 min had a viscosity of 150–330 mPas and a calorific value of 36 kJ g−1, comparable to those of fuel oil.  相似文献

There are several PWB technologies in the market that enables high density interconnection for product miniaturization, and this paper focuses on two HDI technologies, SSP and FVSS®, and discuss the new PWB technologies and the reliability evaluation results for mobile applications. SSP is manufactured in simple lamination process using conventional FR4 materials, which enables future cost reduction and high reliability. FVSS contains filled buried via hole (BVH) and filled micro blind via (μvia) process. By combining the two process FVSS accomplishes high vertical design capability with Stacked μvia-on-μvia and stacked μvia-on-BVH designs.Key reliability requirements for final product quality is drop and temperature cycling reliability in board level in addition to evaluate PWB specific tests to compare performance of new materials used in PWB.  相似文献

The binding of a monoclonal antibody to an epitope peptide wascontrolled by the conformational change of the epitope peptideinduced by anions. We synthesized peptides in which the epitopesequence DTYRYI for the monoclonal antibody AU1 is located betweenamphiphilic peptides (KKLL)n and (LLKK)n. In the absence ofan appropriate anion, the peptide was in a random coil stateand the epitope was linear. In contrast, in the presence ofan appropriate anion, the peptide exhibited an anti-parallel-helical structure and the epitope was subsequently `bent'.In the presence of 41 µM triphosphate, the associationconstant between the antibody and the peptide was decreasedby one order of magnitude in the case of n = 3 and at leastthree orders of magnitude in the case of n = 4 or 5. Oligo-DNAs,as anions, dissociated the antibody–peptide complex, whereastriphosphate did not. The DNA concentrations required for 50%dissociation of the antibody–peptide complex at pH 7.5were 4x10–8, 1x10–7 and 6x10–6 M for decamer,octamer and hexamer DNA, respectively.  相似文献

The effects of sulfur sources on the desulfurization activity of Rhodococcus erythropolis KA2-5-1 were investigated by using an exponential fed-batch culture technique. The feed rate of a sulfur source was controlled independently of the feed rate of ethanol, which was used as a carbon and energy source. Among the sulfur sources examined were dibenzothiophene (DBT), ammonium sulfate, L-cysteine, L-methionine, and 2-amino-ethanesulfonic acid. When the fed-medium contained DBT as the sole sulfur source, KA2-5-1 cells showed a maximum desulfurization activity of approximately 130 mmol 2-HBP kg-cell(-1) h(-1). Similar levels of enzyme activity were also achieved with inexpensive ammonium sulfate by using the exponential fed-batch culture technique. In addition, higher levels of desulfurization activity were achieved by increasing the dosage of the DBT desulfurization (dsz) operon and dszD gene in R. erythropolis KA2-5-1. The recombinant strain showed a maximum desulfurization activity of approximately 250 mmol 2-HBP kg-cell(-1) h(-1) in the exponential fed-batch cultures.  相似文献

An effective method for cell harvesting of the halotolerant microalga, Dunaliella tertiolecta ATCC30929, was investigated. By increasing the pH of the culture broth using NaOH solution, suspended D. terfolecta cells rapidly coagulated and settled within a few minutes, which allowed simple harvesting of the concentrated cells from the culture broth. The pHs for successful coagulation were between 8.6 and 10.5 and more than 90% of cells was recovered from the culture broth. Clarified culture broth could be reused for the successive cultivation of D. tertiolecta simply by supplying fresh medium and neutralizing alkali with HCl without the requirement for an additional inoculum.  相似文献
