A 3D virtual fruit tissue generator is presented that can distinctly define the microstructural components of a fruit tissue and that can be used to model important physical processes such as gas transport during controlled atmosphere storage. The model is based on the biomechanics of plant cells in tissues. The main merit of this algorithm is that it can account for typical differences in intercellular air space networks and in cell size and shape found between different fruit species and tissues. The cell is considered as a closed thin walled structure, maintained in tension by turgor pressure. The cell walls of adjacent cells are modeled as parallel, linear elastic elements which obey Hooke's law. A 3D Voronoi tessellation is used to generate the initial topology of the cells. Intercellular air spaces of schizogenous origin are generated by separating the Voronoi cells along the edges where three Voronoi cells are in contact; while intercellular air spaces of lysigenous origin are generated by deleting (killing) some of the Voronoi cells randomly. Cell expansion then results from turgor pressure acting on the yielding cell wall material. To find the sequence of positions of each vertex and thus the shape of the tissue with time, a system of differential equations for the positions and velocities of each vertex is established and solved using a Matlab ordinary differential equation solver. Statistical comparison with synchrotron tomography images of fruit tissue is excellent. The virtual tissues can be used to study tissue mechanics and exchange processes of important metabolites.  相似文献

Recognition of carbohydrates by proteins is a ubiquitous biochemical process. In legume–rhizobium symbiosis, lipochitin oligosaccharides, also referred to as nodulation (nod) factors, function as primary rhizobial signal molecules to trigger root nodule development. Perception of these signal molecules is receptor mediated, and nod factor receptor 5 (NFR5) from the model legume Lotus japonicus is predicted to contain three LysM domain binding sites. Here we studied the interactions between nod factor and each of the three NFR5 LysM domains, which were chemically synthesized. LysM domain variants (up to 58 amino acids) designed to optimize solubility were chemically assembled by solid‐phase peptide synthesis (SPPS) with microwave heating. Their interaction with nod factors and chitin oligosaccharides was studied by isothermal titration calorimetry and circular dichroism (CD) spectroscopy. LysM2 showed a change in folding upon nod factor binding, thus providing direct evidence that the LysM domain of NFR5 recognizes lipochitin oligosaccharides. These results clearly show that the L. japonicus LysM2 domain binds to the nod factor from Mesorhizobium loti, thereby causing a conformational change in the LysM2 domain. The preferential affinity for nod factors over chitin oligosaccharides was demonstrated by a newly developed glycan microarray. Besides the biological implications, our approach shows that carbohydrate binding to a small protein domain can be detected by CD spectroscopy.  相似文献

Information fusion is an essential part of nearly all systems whose goal is to derive decisions from multiple sources. Often, a fusion solution has parameters and the goal is to learn them from data. Herein, we propose efficient evolutionary algorithm (EA) operators to facilitate learning the Choquet integral (ChI). Whereas many EAs provide a way to solve complex, unconstrained optimization tasks, most tend to perform relatively poor in light of constraints. Recently, a few EA-based approaches to optimizing the ChI have appeared. Namely, these methods focus on fixing the values of variables so conditions are met or feasible candidate pairs are identified for steps such as crossover. Herein, we introduce a new set of transparent operators that are guaranteed to naturally preserve constraints, thus eliminating the need to resort to costly evaluations and fixing of constraint violations. In particular, our method scales well to large numbers of inequality constraints, something that prior work does not. The proposed algorithm, coined efficient ChI genetic algorithm (ECGA), is evaluated on several synthetic data sets and it is compared with state-of-the-art algorithms. This paper presents the implementation of a multiple analyte enzyme assay, based on the sequential injection of the different enzyme solutions, in an electrokinetic driven microfluidic chip. The assay methodology for the simultaneous quantification of d-glucose and d-fructose was reported in previous publications but here the real integration of both enzyme assays was achieved. When assays were executed separately, good reproducibility was observed with average CV values of 5.2% and 4.5% for the d-glucose and d-fructose assay, respectively. Next, the assays for the quantification of d-glucose and d-fructose were integrated simultaneously on chip, where each assay was executed consecutively in the same microreactor by applying a specific sequence of potentials at the reservoirs. This article proves the integration of a sequential based quantification approach in continuous microfluidic chips with electrokinetic actuation.  相似文献

Bats fly using a thin wing membrane composed of compliant, anisotropic skin. Wing membrane skin deforms dramatically as bats fly, and its three-dimensional configurations depend, in large part, on the mechanical behaviour of the tissue. Large, macroscopic elastin fibres are an unusual mechanical element found in the skin of bat wings. We characterize the fibre orientation and demonstrate that elastin fibres are responsible for the distinctive wrinkles in the surrounding membrane matrix. Uniaxial mechanical testing of the wing membrane, both parallel and perpendicular to elastin fibres, is used to distinguish the contribution of elastin and the surrounding matrix to the overall membrane mechanical behaviour. We find that the matrix is isotropic within the plane of the membrane and responsible for bearing load at high stress; elastin fibres are responsible for membrane anisotropy and only contribute substantially to load bearing at very low stress. The architecture of elastin fibres provides the extreme extensibility and self-folding/self-packing of the wing membrane skin. We relate these findings to flight with membrane wings and discuss the aeromechanical significance of elastin fibre pre-stress, membrane excess length, and how these parameters may aid bats in resisting gusts and preventing membrane flutter.  相似文献

As the American population ages, older adults are accounting for a larger percentage of the drug-abusing population, but little attention has been given to this age group especially in regards to evaluating responsivity to different treatment modalities. Contingency management (CM) is a highly effective behavioral treatment that provides positive tangible reinforcers for objective evidence of behavior change. The purpose of this study was to examine main and interactive effects of age on outcomes in cocaine-dependent patients receiving CM with standard care (SC) or SC alone. Patients (N = 393) participating in 1 of 3 randomized trials of CM for cocaine dependence were divided into young, middle, and older age cohorts. Baseline characteristics and outcomes were compared across the age groups. The oldest age group had more medical problems than the youngest and middle age groups but had fewer legal difficulties and psychiatric symptoms. The oldest age group remained in treatment significantly longer than the other age groups, regardless of the type of treatment received. Although all age groups benefited from CM in terms of retention and longest duration of abstinence achieved, a significant age by treatment interaction effect emerged, with the older cohort improving relatively less from CM than the younger age groups. These findings demonstrate that age may play a role in moderating intervention outcomes, and tailoring CM to the needs of older and middle-aged substance abusers may be important for improving outcomes in this growing population. The effect of on‐chain ketone defects on the charge transport of the polyfluorene derivative poly(9,9‐dioctylfluorene) (PFO) is investigated. Using MoO3 as ohmic hole contact, the hole transport in a pristine PFO diode is observed to be limited by space‐charge, whereas fluorenone contaminated PFO (PFO‐F) is shown to be trap limited by the occurrence of an exponential trap distribution with a trap depth of 0.18 eV. The electron transport in PFO is also observed to be trap limited, but in order to describe the electron transport of PFO‐F, an additional trap level with a depth of 0.46 eV must be introduced. The obtained energy levels of the fluorenone trapping sites are in close agreement with cyclic voltammetry (CV) measurements reported in literature. As a result, the fluorenone defects are shown to simultaneously act as hole‐ and electron trap. Moreover, through ideality factor measurements, the green emission associated with these defects is observed to originate from trap‐assisted recombination.  相似文献

Water loss of fruit during storage has a large impact on fruit quality and shelf life and is essential to fruit drying. Dehydration of fruit tissues is often accompanied by large deformations. One-dimensional water transport and large deformation of cylindrical samples of apple tissue during dehydration were modeled by coupled mass transfer and mechanics and validated by calibrated X-ray CT measurements. Uni-axial compression–relaxation tests were carried out to determine the nonlinear viscoelastic properties of apple tissue. The Mooney–Rivlin and Yeoh hyperelastic potentials with three parameters were effective to reproduce the nonlinear behavior during the loading region. Maxwell model was successful to quantify the viscoelastic behavior of the tissue during stress relaxation. The nonlinear models were superior to linear elastic and viscoelastic models to predict deformation and water loss. The sensitivity of different model parameters using the nonlinear viscoelastic model using Yeoh hyperelastic potentials was studied. The model predictions proved to be more sensitive to water transport parameters than to the mechanical parameters. The large effect of relative humidity and temperature on the deformation of apple tissue was confirmed by this study. The validated model can be employed to better understand postharvest storage and drying processes of apple fruit and thus improve product quality in the cold chain.  相似文献
