Our study aimed to examine the effects of hypertension and the chronic administration of the fatty acid amide hydrolase (FAAH) inhibitor URB597 on vascular function and the endocannabinoid system in spontaneously hypertensive rats (SHR). Functional studies were performed on small mesenteric G3 arteries (sMA) and aortas isolated from SHR and normotensive Wistar Kyoto rats (WKY) treated with URB597 (1 mg/kg; twice daily for 14 days). In the aortas and sMA of SHR, endocannabinoid levels and cannabinoid CB1 receptor (CB1R) expression were elevated. The CB1R antagonist AM251 diminished the methanandamide-evoked relaxation only in the sMA of SHR and enhanced the vasoconstriction induced by phenylephrine and the thromboxane analog U46619 in sMA in SHR and WKY. In the sMA of SHR, URB597 elevated anandamide levels, improved the endothelium-dependent vasorelaxation to acetylcholine, and in the presence of AM251 reduced the vasoconstriction to phenylephrine and enhanced the vasodilatation to methanandamide, and tended to reduce hypertrophy. In the aortas, URB597 elevated endocannabinoid levels improved the endothelium-dependent vasorelaxation to acetylcholine and decreased CB1R expression. Our study showed that hypertension and chronic administration of URB597 caused local, resistance artery-specific beneficial alterations in the vascular endocannabinoid system, which may bring further advantages for therapeutic application of pharmacological inhibition of FAAH.  相似文献

Even though immunotherapy has radically changed the search for anticancer therapies, there are still many different pathways that are open to intervention with traditional small molecules. To expand our investigation in the anticancer field, we report here a new series of compounds in which our previous pyrazole and imidazopyrazole scaffolds are linked to a differently decorated phenyl ring through an acylhydrazone linker. Preliminary tests on the library were performed at the National Cancer Institute (USA) against the full NCI 60 cell panel. The best compounds among the imidazopyrazole series were then tested by immunofluorescence staining for their inhibition of cell proliferation, apoptosis induction, and their effect on the cell cycle and on microtubules. Two compounds, in particular 4-benzyloxy-3-methoxybenzyliden imidazopyrazole-7-carbohydrazide showed good growth inhibition, with IC50 values in the low-micromolar range, and induced apoptosis. Both compounds altered the cell-cycle phases with the appearance of polyploid cells. Immunofluorescence analysis evidenced microtubules alterations; tubulin polymerization assays and docking studies suggested the tubulin system to be the possible, although not exclusive, target of the new acylhydrazone series reported here.  相似文献

Physical exercise is known to influence hormonal mediators of appetite, but the effect of short-term maximal intensity exercise on plasma levels of appetite hormones and cytokines has been little studied. We investigated the effect of a 30 s Wingate Test, followed by a postprandial period, on appetite sensations, food intake, and appetite hormones. Twenty-six physically active young males rated their subjective feelings of hunger, prospective food consumption, and fatigue on visual analogue scales at baseline, after exercise was completed, and during the postprandial period. Blood samples were obtained for the measurement of nesfatin-1, ghrelin, leptin, insulin, pancreatic polypeptide (PP), human growth factor (hGH) and cytokine interleukin-6 (IL-6), irisin and plasma lactate concentrations, at 30 min before exercise, immediately (210 s) after exercise, and 30 min following a meal and at corresponding times in control sedentary males without ad libitum meal intake, respectively. Appetite perceptions and food intake were decreased in response to exercise. Plasma levels of irisin, IL-6, lactate, nesfatin-1 and ghrelin was increased after exercise and then it was returned to postprandial/control period in both groups. A significant rise in plasma insulin, hGH and PP levels after exercise was observed while meal intake potentiated this response. In conclusion, an acute short-term fatiguing exercise can transiently suppress hunger sensations and food intake in humans. We postulate that this physiological response involves exercise-induced alterations in plasma hormones and the release of myokines such as irisin and IL-6, and supports the notion of existence of the skeletal muscle–brain–gut axis. Nevertheless, the detailed relationship between acute exercise releasing myokines, appetite sensations and impairment of this axis leading to several diseases should be further examined.  相似文献

Micro-Raman light scattering experiments on PbZrO3 (PZO) single crystal doped with Nb5+ have been investigated. Special attention was paid to the paraelectric (PE) phase in which nominally forbidden first-order Raman spectra were detected at temperatures far above the phase transition TC. Complex Raman spectra were observed in the vicinity of three structural phase transitions. These results mainly from the coexistence of phases with different symmetries in a wide temperature range below TC. The Raman measurements have been compared with dielectric and optical observations and proved that polar nanoregions in a centrosymmetric lattice appear well above TC. It was shown that doping ABO3 perovskites with heterovalent ions like Nb5+ unbalances charge neutrality of the lattice and strongly extends the temperature range of polar regions. The investigations performed point out that in the PE matrix the interaction between electrons and lattice vibrations, as recently suggested for pure PZO, plays an important role.  相似文献

Tissues and biofluids are important sources of information used for the detection of diseases and decisions on patient therapies. There are several accepted methods for preservation of tissues, among which the most popular are fresh‐frozen and formalin‐fixed paraffin embedded methods. Depending on the preservation method and the amount of sample available, various specific protocols are available for tissue processing for subsequent proteomic analysis. Protocols are tailored to answer various biological questions, and as such vary in lysis and digestion conditions, as well as duration. The existence of diverse tissue‐sample protocols has led to confusion in how to choose the best protocol for a given tissue and made it difficult to compare results across sample types. Here, we summarize procedures used for tissue processing for subsequent bottom‐up proteomic analysis. Furthermore, we compare protocols for their variations in the composition of lysis buffers, digestion procedures, and purification steps. For example, reports have shown that lysis buffer composition plays an important role in the profile of extracted proteins: the most common are tris(hydroxymethyl)aminomethane, radioimmunoprecipitation assay, and ammonium bicarbonate buffers. Although, trypsin is the most commonly used enzyme for proteolysis, in some protocols it is supplemented with Lys‐C and/or chymotrypsin, which will often lead to an increase in proteome coverage. Data show that the selection of the lysis procedure might need to be tissue‐specific to produce distinct protocols for individual tissue types. Finally, selection of the procedures is also influenced by the amount of sample available, which range from biopsies or the size of a few dozen of mm2 obtained with laser capture microdissection to much larger amounts that weight several milligrams.  相似文献

Legal definitions exert a significant impact on the waste management strategy. Waste that is technically suitable for recovery does not automatically become a raw material if there is no market for it, or its use is not commercially effective and, hence, they should be disposed of. The majority of disposed wastes, including recyclable waste, are not environmentally safe. Waste as a freshly generated anthropogenic material is not geochemically stable. Przezchlebie fly ash surface pond (Upper Silesia, Poland) in the post-closure stage was subject to field validation of the results of laboratory leaching/extraction tests and long-term column experiments on fly ash (FA) leaching behaviour under controlled conditions for environmental risk assessment. The study showed: (i) the possibility of a discontinuous non-linear time delayed increase of pollution potential of disused 'non-hazardous' large-volume waste in the dumping sites to the hazardous level; (ii) inconsistency of the laboratory leaching tests and the actual leaching behaviour of trace metals, particularly when equilibria conditions are dictated by kinetically determined reactions where the test results reflected entirely wash-out (I) and dissolution (II) phases, but did not comprise delayed release (III) phase; and (iii) necessity of life-cycle screening/monitoring of 'non-hazardous' dumping sites for contaminant release as a function of the primary (pH-Eh, ionic strength, ionic composition of solute) and secondary controlling factors (L/S-liquid to solid ratio, water flow conditions) along the vertical profile of an anthropogenic or natural vadose zone. These data are to be used to develop long-term predictive hydrogeochemical models and their field validation, and for providing an early warning and remedial actions with respect to the particular site. The formation of pH (and Eh) as a function of time-dependent (kinetically defined) processes appeared to be a key issue for a correct prediction of the leaching behaviour of waste. The presented case study on FA shows that waste, even those considered non-hazardous and fit for use in a commercially proven applications, should not be treated the same way as a natural raw material. To facilitate waste utilisation and disposal in environmentally safe way and to prioritise its use, reliable environmental risk assessment prediction models and testing procedures, as well as special enforcement strategy and regulations, should be developed with respect to waste and not 'materials.'  相似文献

The purpose of this study is to investigate the effect of treated natural zeolite on the flow behavior, morphology, thermal, and mechanical properties in recycled polyolefin blends. A model polymer blend consisting of 95 wt% polyolefines and 5 wt% polystyrene was studied. Compositions from this recycled blend and dehydrated zeolite in a narrow concentration range (0–5 wt%) were treated with maleic anhydride-grafted polypropylene and initiator dicumyl peroxide. The compositions were characterized by capillary rheometry, wide-angle X-ray scattering, differential scanning calorimetry, scanning electron microscopy, and mechanical tests. The results show a compatibilizing effect of treated zeolite expressed by increase of shear viscosity and improvement of interfacial interactions and impact strength. The results can open wide possibilities for utilization of treated zeolite in recycling of unsorted polymer wastes.  相似文献
