Cyclophilins have important homeostatic roles, but following tissue injury, cyclophilin A (CypA) can promote leukocyte recruitment and inflammation, while CypD can facilitate mitochondrial-dependent cell death. This study investigated the therapeutic potential of a selective cyclophilin inhibitor (GS-642362), which does not block calcineurin function, in mouse models of tubular cell necrosis and renal fibrosis. Mice underwent bilateral renal ischemia/reperfusion injury (IRI) and were killed 24 h later: treatment with 10 or 30 mg/kg/BID GS-642362 (or vehicle) began 1 h before surgery. In the second model, mice underwent unilateral ureteric obstruction (UUO) surgery and were killed 7 days later; treatment with 10 or 30 mg/kg/BID GS-642362 (or vehicle) began 1 h before surgery. GS-642362 treatment gave a profound and dose-dependent protection from acute renal failure in the IRI model. This protection was associated with reduced tubular cell death, including a dramatic reduction in neutrophil infiltration. In the UUO model, GS-642362 treatment significantly reduced tubular cell death, macrophage infiltration, and renal fibrosis. This protective effect was independent of the upregulation of IL-2 and activation of the stress-activated protein kinases (p38 and JNK). In conclusion, GS-642362 was effective in suppressing both acute kidney injury and renal fibrosis. These findings support further investigation of cyclophilin blockade in other types of acute and chronic kidney disease. 相似文献

In this study, the effect of co-fermentation and sequential fermentation with a typical halophilic soy sauce yeast Candida versatilis and a non-halophilic dairy bacterium Lactococcus lactis subsp. cremoris in pork hydrolysates was explored for the first time with regard to their viability, physicochemical changes and volatile compound production. It was observed that the growth of C. versatilis was suppressed (only 0.6 - 1.5 log CFU mL−1 increase), whilst L. lactis subsp. cremoris was stimulated (total cell counts exceeded 9.0 log CFU mL−1) in mixed-cultures relative to the respective monocultures. There were no significant differences between co-inoculation and sequential inoculation regarding glucose consumption, organic acid production and free amino acids utilisation. Some distinct volatile compounds such as methionol, 6-methyl-5-hepten-2-one and gamma-nonalactone were found in both co-inoculated and sequentially inoculated samples, with slightly different concentrations. These results suggest that the mixed-inoculation of C. versatilis and L. lactis have a positive impact on the volatile compounds formation in pork hydrolysates. 相似文献

We determined and fine‐tuned the solids transport models appropriate for predicting the single‐phase carrier fluid velocity to transport solid particles in conduits for horizontal, low concentration flow. A database with 538 experimental data points was compiled. A literature review was performed to determine the data ranges, forces, and mechanisms used to develop 44 models, and their velocity predictions were compared against the database using statistics. Using the dimensionless forms of the models and the data, the model parameters were adjusted to improve their accuracy and identify the dominant forces. At low concentrations: for liquid/solid flow from a bed of solids and gas/solid flow from the bottom of pipelines, the particle weight, and inertial and viscous forces dominate; for gas/solid flow from a bed of solids, the particle weight, and inertial, viscous, and adhesive forces play a role; and gaps exist in the data for large‐diameter pipes and high‐density gases. © 2013 American Institute of Chemical Engineers AIChE J, 60: 76–122, 2014 相似文献

ABSTRACT: The electronic states of a singly ionized on-axis double-donor complex (D2+) confined in two identical vertically coupled, axially symmetrical quantum dots in a threading magnetic field are calculated. Energy consumption is a perennial issue in the design of wireless sensor networks (WSNs) which typically rely on portable sources like batteries for power. Recent advances in ambient energy harvesting technology have made it a potential and promising alternative source of energy for powering WSNs. By using energy harvesters with supercapacitors, WSNs are able to operate perpetually until hardware failure and in places where batteries are hard or impossible to replace. In this paper, we study the performance of different medium access control (MAC) schemes based on CSMA and polling techniques for WSNs which are solely powered by ambient energy harvesting using energy harvesters. We base the study on (i) network throughput (S), which is the rate of sensor data received by the sink, (ii) fairness index (F), which determines whether the bandwidth is allocated to each sensor node equally and (iii) inter-arrival time (γ) which measures the average time difference between two packets from a source node. For CSMA, we compare both the slotted and unslotted variants. For polling, we first consider identity polling. Then we design a probabilistic polling protocol that takes into account the unpredictability of the energy harvesting process to achieve good performance. Finally, we present an optimal polling MAC protocol to determine the theoretical maximum performance. We validate the analytical models using extensive simulations incorporating experimental results from the characterization of different types of energy harvesters. The performance results show that probabilistic polling achieves high throughput and fairness as well as low inter-arrival times. 相似文献

We present the main design features for the GISMO-2 bolometer camera, which we build for background-limited operation at the IRAM 30 m telescope on Pico Veleta, Spain. GISMO-2 will operate simultaneously in the 1 and 2 mm atmospherical windows. The 1 mm channel uses a \(32 \times 40\) TES-based backshort under grid (BUG) bolometer array, the 2 mm channel operates with a \(16 \times 16\) BUG array. The camera utilizes almost the entire full field of view provided by the telescope. The optical design of GISMO-2 was strongly influenced by our experience with the GISMO 2 mm bolometer camera, which is successfully operating at the 30 m telescope. GISMO is accessible to the astronomical community through the regular IRAM call for proposals. 相似文献
