Mercury (Hg) is an environmental pollutant that impacts human and ecosystem health. In our previous works, we reported alterations in the properties of Mytilus galloprovincialis protamine-like (PL) proteins after 24 h of exposure to subtoxic doses of toxic metals such as copper and cadmium. The present work aims to assess the effects of 24 h of exposure to 1, 10, and 100 pM HgCl2 on spermatozoa and PL proteins of Mytilus galloprovincialis. Inductively coupled plasma–mass spectrometry indicated accumulation of this metal in the gonads of exposed mussels. Further, RT-qPCR analyses showed altered expression levels of spermatozoa mt10 and hsp70 genes. In Mytilus galloprovincialis, PL proteins represent the major basic component of sperm chromatin. These proteins, following exposure of mussels to HgCl2, appeared, by SDS-PAGE, partly as aggregates and showed a decreased DNA-binding capacity that rendered them unable to prevent DNA damage, in the presence of CuCl2 and H2O2. These results demonstrate that even these doses of HgCl2 exposure could affect the properties of PL proteins and result in adverse effects on the reproductive system of this organism. These analyses could be useful in developing rapid and efficient chromatin-based genotoxicity assays for pollution biomonitoring programs.  相似文献

The current study evaluated exosomes isolated from plasma of heifers bred to have high or low fertility through developing extreme diversity in fertility breeding values, however, key animal traits (e.g., body weight, milk production, and percentage of North American genetics) remained similar between the 2 groups. The exosomes were isolated by a combined ultracentrifugation and size exclusion chromatography approach and characterized by their size distribution (nanoparticle tracking analysis), morphology (transmission electron microscopy), and presence of exosomal markers (immunoblotting). In addition, a targeted mass spectrometry approach was used to confirm the presence of 2 exosomal markers, tumor susceptibility gene 101 and flotillin 1. The number of exosomes from plasma of high fertility heifers was greater compared with low fertility heifers. Interestingly, the exosomal proteomic profile, evaluated using mass spectrometry, identified 89 and 116 proteins in the high and low fertility heifers respectively, of which 4 and 31 were unique, respectively. These include proteins associated with specific biological processes and molecular functions of fertility. Most notably, the tetratricopeptide repeat protein 41-related, glycodelin, and kelch-like protein 8 were identified in plasma exosomes unique to the low fertility heifers. These proteins are suggested to play a role in reproduction; however, the role of these proteins in dairy cow reproduction remains to be elucidated. Their identification underscores the potential for proteins within exosomes to provide information on the fertility status and physiological condition of the cow. This may potentially lead to the development of prognostic tools and interventions to improving dairy cow fertility.  相似文献

Zinc (Zn), the second-most necessary trace element, is abundant in the human body. The human body lacks the capacity to store Zn; hence, the dietary intake of Zn is essential for various functions and metabolism. The uptake of Zn during its transport through the body is important for proper development of the three major accessory sex glands: the testis, epididymis, and prostate. It plays key roles in the initial stages of germ cell development and spermatogenesis, sperm cell development and maturation, ejaculation, liquefaction, the binding of spermatozoa and prostasomes, capacitation, and fertilization. The prostate releases more Zn into the seminal plasma during ejaculation, and it plays a significant role in sperm release and motility. During the maternal, labor, perinatal, and neonatal periods, the part of Zn is vital. The average dietary intake of Zn is in the range of 8–12 mg/day in developing countries during the maternal period. Globally, the dietary intake of Zn varies for pregnant and lactating mothers, but the average Zn intake is in the range of 9.6–11.2 mg/day. The absence of Zn and the consequences of this have been discussed using critical evidence. The events and functions of Zn related to successful fertilization have been summarized in detail. Briefly, our current review emphasizes the role of Zn at each stage of human reproduction, from the spermatogenesis process to childbirth. The role of Zn and its supplementation in in vitro fertilization (IVF) opens opportunities for future studies on reproductive biology.  相似文献

We studied the use of the 3 commonly used reproductive hormones, namely prostaglandins, GnRH, and progesterone, and associated herd-level factors on 760 Dutch dairy farms from 5 veterinary clinics. From 2017 to 2019 we collected data on the sales of reproductive hormones, converted this data into the number of reproductive hormone doses conducted, and expressed this as the annual number of reproductive hormone doses per 100 adult dairy cows. Additional herd-level information was available for 2019. Due to the excess of zeros in the data set (i.e., a substantial number of farms did not use any hormones), we used a zero-inflated negative binomial model to identify related herd-level factors for the use of reproductive hormones. In the entire study period of 2017 to 2019, 5.8% of the dairy farms did not use any reproductive hormones, with the proportion of nonusers varying between 0.0 and 10.3% per veterinary clinic. This proportion was around 13.5% on an annual basis. Prostaglandins were the most frequently used reproductive hormone in Dutch dairy cows (62.9%), followed by GnRH (33.1%) and progesterone (4.0%). Furthermore, participating in a veterinary herd health management program had a significant effect on reproductive hormone use. These farms used more reproductive hormones than farms that did not participate in a herd health management program and were less represented in the group of nonuser farms. Technologies, such as pedometers and automatic milking systems, also had an effect on reproductive hormone use. The presence of pedometers or activity monitors did not reduce the use of the reproductive hormones but was associated with a greater frequency of users. Farms with an automatic milking system used more reproductive hormones than farms with a conventional milking system. With this study, we have made a first step in achieving transparency in the Dutch dairy industry by providing an objective overview of reproductive hormone use on Dutch dairy farms and identifying associations with some herd-level factors.  相似文献

The biology and physiology of Tyrophagus putrescentiae (Schrank) (Sarcoptiformes: Acaridae), also known as the ham, cheese or mold mite, is reviewed along with methods that have been evaluated for managing and controlling this pest. This review was conducted because the ham mite is an important target pest of the dry cured ham industry. Methyl bromide has been historically used to control mite infestations, but is now banned or being phased out of use in most countries because it is an ozone-depleting substance. Only commercially available stockpiles and quarantine and pre-shipment methyl bromide are available for use in countries that require such action. This review compares the effectiveness and feasibility of recently investigated methods to control mite infestations on dry cured ham and to discuss integrated pest management plans for ham mites. The review also proposes methods for conducting a prevention and monitoring-based integrated pest management program that relies on definitive mitigation such as fumigation only when mite numbers exceed a critical action threshold.  相似文献

The Red Devil Cichlid Amphilophus labiatus (Günther 1864), native to Lake Nicaragua and Lake Managua in Central America, is widely cultivated as an attractive, yet aggressive, aquarium fish that has been introduced to several freshwater locations around the world. It was recently introduced into Lake Sentani in Indonesian New Guinea, a naturally rich aquatic habitat home exhibiting a diverse fauna with high endemism. To better understand the invasive potential and impacts of A. labiatus, the present study was undertaken to describe the sex ratio and reproductive potential of A. labiatus in Lake Sentani through determination of its gonad maturation stages, gonado‐somatic index and fecundity. The fish were collected using floating gill nets at six locations around the lake between June and August 2019. Sex identification, gonads type, gonad stage and weight, fecundity were observed in the laboratory. Of the total 542 of A. labiatus collected during the survey, 345 individuals were sampled for sex ratio and reproduction. The overall male:female ratio was skewed towards males in a ratio of 3.28:1. The males were generally found to be in the gonad maturation stage I, while the females were commonly found in gonad maturation stages III and IV for all sampling locations during the study period. The highest male mean GSI value was found for the Yakonde site (0.24 ± 0.21) and at the Ayapo site (1.11 ± 1.01) for the females, with a fecundity of 191–2,720 eggs. The fish have spread to most parts of the lake, being conditioned to potentially spawn at any location, thereby providing an explanation for the observed rapid increase of the A. labiatus population over the last ten years. The results of the present study suggest a high invasive potential for this species in this and other similar lake habitats, warning that their careful management should be undertaken to restrict their further spread in Indonesia and elsewhere.  相似文献

Metallic colors have a unique appearance of glossiness with features such as highlights, contrast, and reflections on their surface, and therefore, metallic objects are very attractive to humans. Especially, gold, silver, and copper colors are familiar metals used as decorative materials, coins, and other furnishings. However, the mechanism and condition of metallic perception have not been fully investigated. There are a few studies for investigating metallic perception using rendered patches or images, but there is no study using real‐world objects. In our previous study, we developed a simple representation technique that made real objects appear to be made of gold by projecting a solid color onto a target nonmetallic object. By using the representation technique, in this study, we have further investigated the perception of metallic appearance such as gold, silver, and copper using real‐world materials, and analyzed the difference between these metallic perceptions. Our results indicate that the perception of the metallic object is different for gold, silver, and copper. Our new findings are as follows: the glitter required for the perception of gold and silver becomes an obstacle to the perception of copper; the metallic perception reveals that learning experience might be strongly affecting; and luminance adjustment is sensitive to the perception of metallic objects.  相似文献

Our primary objective was to evaluate the effect of feeding rumen-protected Met (RPM) in the pre- and postpartum total mixed ration (TMR) on pregnancy per artificial insemination (AI) and pregnancy loss in multiparous Holstein cows. We also evaluated multiple secondary reproductive physiological outcomes before and after AI, including uterine health, ovarian cyclicity, response to synchronization of ovulation, and markers of embryo development and size. A total of 470 multiparous Holstein cows [235 at the University of Wisconsin (UW) and 235 at Cornell University (CU)] were used for this experiment. Experimental treatment diets were applied at the pen level (2 and 4 close-up pens at CU and UW, respectively, and 12 and 6 postfresh pens at CU and UW, respectively); thus, pen was the experimental unit, and cow was the observational unit. Cows were enrolled and randomly assigned to be fed the experimental treatment diets at approximately 4 wk before parturition until 67 d of gestation [147 d in milk (DIM)] after their first service. Close-up dry cow and replicated lactation pens were randomly assigned to treatment diets: RPM, prepartum = 2.83% (UW) and 2.85% (CU), postpartum = 2.58% (UW) and 2.65% (CU); and control (CON), prepartum = 2.30% (UW) and 2.22% (CU), postpartum = 2.09% (UW) and 2.19% (CU; Met as percentage of metabolizable protein). Vaginal discharge and uterine cytology (percentage of polymorphonuclear leucocytes) were evaluated at 35 ± 3 DIM. Cows received timed AI (TAI) at 80 ± 3 DIM after synchronization of ovulation with the Double-Ovsynch protocol. Ovarian cyclicity status, response to synchronization of ovulation, and luteal function were determined by measuring circulating concentrations of progesterone at 35 and 49 ± 3 DIM, 48 and 24 h before TAI, and 8, 18, 22, 25, and 29 d after TAI. Interferon-stimulated gene expression in white blood cells were compared on 18 d after TAI (CU only) and pregnancy-specific protein B concentrations at 22, 25, 29, 32, and 67 d after TAI. Pregnancy status was determined using pregnancy-specific protein B at 25 and 29 d after TAI, and by transrectal ultrasonography at 32, 39, and 67 d after TAI. Embryo and amniotic vesicle size were determined at 32 and 39 d after TAI. Pregnancy per AI (25 d: 64.7 vs. 64.0%, 32 d: 54.3 vs. 55.1% for CON and RPM, respectively) and pregnancy loss (25 to 67 d: 22.6 vs. 19.2% for CON and RPM, respectively) for synchronized cows did not differ. The proportion of cows with purulent vaginal discharge (CON = 7.7 vs. RPM = 4.6%) and cytological endometritis (CON = 20.8 vs. RPM = 23.6%) did not differ. Cyclicity status, ovarian responses to the synchronization protocol, and synchronization rate also did not differ. In addition, fold change for interferon-stimulated genes, concentrations of pregnancy-specific protein B, and embryo size were not affected by treatments. In conclusion, feeding RPM in the pre- and postpartum TMR at the amounts used in this experiment did not affect uterine health, cyclicity, embryo development, or reproductive efficiency in dairy cows.  相似文献

为提高彩色柔性版印刷的色彩阶调再现性,研究了柔性版印刷网点扩大特性、阶调再现特性和色彩再现特性,并对网纹辊线数与印版网点线数的关系进行了研究.在相同的印刷条件下,通过柔性版印刷机进行印刷实验,建立了柔性版印刷profile文件,从而得到柔性版印刷网点扩大曲线、阶调再现曲线和色彩再现特性.同时,采用不同线数的网纹辊对不同网点线数的印版进行印刷,测量了印刷品的阶调再现曲线和网点扩大曲线.实验结果表明:柔性版印刷网点扩大较大,且在亮调到中间调范围(网点百分比为30%~40%)处的网点扩大最严重,从而使得柔性版印刷的阶调再现范围较窄.网纹辊线数与印版网点线数的比值越大,所得到的印刷品阶调再现性越好,其比值应大于3~4.柔性版印刷的色域在红绿色范围内较胶印小,而在蓝色范围内较胶印大.为提高彩色柔性版印刷的质量,应利用profile文件对图象进行分色制版.  相似文献

The purpose of this paper is to investigate the characteristics of the dissemination of information in the community. A variety of possible factors that affect the dissemination of information in Sina Weibo have been discussed. By analyzing the process of the information dissemination in the community of Sina Weibo, we found the information dissemination of Weibo community and the dynamic model are very similar. With the aid of data intensive computing theory, the various features have been mined and modeled. The dynamic model is improved and redefined to characterize the community. Then the SEINR model is proposed. The basic reproductive number, the existence of equilibrium point and the stability of the network are analyzed and proved in detail. By comparing with real data in Weibo community, we show that the SEINR model accurately reflects the dissemination of information community. Furthermore, we investigate the SEINR model in detail to show the influences of different parameters on information dissemination by simulations.  相似文献
