摘    要:目的 探究表面活性剂十二烷基硫酸钠(sodium dodecyl sulfate, SDS)协助超声波提取甘薯叶总黄酮的最佳工艺条件。方法 以甘薯叶总黄酮提取量为考察指标, 采用Box-Behnken法进行试验设计, 对SDS质量浓度、乙醇体积分数、料液比、超声时间进行响应面优化。结果 各因素对甘薯叶总黄酮提取量的影响程度为C(料液比)>A (SDS质量浓度)>B(乙醇体积分数)>D(超声时间), 甘薯叶总黄酮最佳提取工艺参数为SDS质量浓度1.0%、乙醇体积分数81%、料液比1:50 (g:mL)、超声时间49 min, 在此条件下, 总黄酮提取量为 132.08 mg/g, 与回归模型预测值134.63 mg/g相差1.89%, 表明该模型与实际情况拟合良好。结论 利用表面活性剂十二烷基硫酸钠协同超声提取甘薯叶总黄酮的工艺方法稳定可行, 可为甘薯叶总黄酮的进一步开发利用提供参考依据。

Abstract:Objective To explore the optimum technological conditions for the extraction of total flavonoids from sweet potato leaves by ultrasonic with the aid of the surfactant sodium dodecyl sulfate (SDS). Methods The total flavonoids extracted from sweet potato leaves were used as the inspection index, the Box-Behnken method was used to design the experiment, and response surface optimization was carried out for the mass concentration of SDS, the volume fraction of ethanol, the ratio of material to liquid, and the ultrasound time. Results The degree of influence of extraction factors on the total flavonoids extraction amount from sweet potato leaves was C (material-to-liquid ratio)>A (SDS mass concentration)>B (ethanol volume fraction)>D (ultrasound time), the optimal extraction process parameters of total flavonoids from sweet potato leaves were 1.0% SDS, ethanol volume fraction 81%, material-to-liquid ratio 1:50 (g:mL), and ultrasound time 49 min, under these conditions, the total flavonoid extraction amount was 132.08 mg/g, 1.89% different from the predicted value of the regression model of 134.63 mg/g, indicating that the model fitted well with the actual situation. Conclusion The method of extracting total flavonoids from sweet potato leaves by ultrasonic with the aid of surfactant sodium lauryl sulfate is stable and feasible, which provides a reference for the further development and utilization of total flavonoids from sweet potato leaves.
