酵母富硒蛋白提取条件优化

摘    要:以实验室前期分离的富硒酵母菌株库德里阿兹威氏毕赤酵母(Pichia kudriavzevii)XJ1-3为试验菌株,通过单因素试验和响应面分析优化碱法提取酵母富硒蛋白的提取条件。结果表明,碱法提取酵母富硒蛋白最佳提取条件为NaOH浓度0.1 mol/L,液料比7.5∶1(mL∶g),提取时间140 min。在此优化条件下,酵母富硒蛋白的提取率达到12.62%,酵母富硒蛋白中硒含量为358.9 μg/g。

Abstract:Using selenium-enriched yeast strain Pichia kudriavzevii XJ1-3 isolated by laboratory in the early stage as test strain in this experiment, the alkaline extraction conditions of yeast selenium-enriched protein were optimized through single factor test and response surface analysis. The results showed that the optimal alkaline extraction conditions of yeast selenium-enriched protein were obtained as follows: sodium hydroxide concentration 0.1 mol/L, liquid and material ratio 7.5∶1 (ml∶g), and extraction time 140 min. Under these optimal conditions, the extraction rate of yeast selenium-enriched protein reached 12.62%, and the selenium contents in yeast selenium- enriched protein were 358.9 μg/g.
