霍山蹦蝗中黄酮类化合物提取工艺与检测方法研究

摘    要:考察不同因素对霍山蹦蝗中黄酮类化合物提取的影响。通过正交试验及SPSS分析,得出其优化工艺条件为温度85℃、甲醇浓度80%、时间2h、料液比1∶50(m∶V),提取率为2.02%。运用铝盐显色法和高效液相色谱法分别测出霍山蹦蝗中总黄酮含量为20.198,4.047mg/g(干重),结果表明铝盐显色法可能受杂质酚类干扰,而高效液相色谱法能很好的将不同黄酮类化合物与杂质分离开,因此检测结果更准确。

Abstract:Orthogonal experiment and SPSS analysis were used to investigate the different influence factors of the flavonoids extraction from Sinopodisma houshana Huang.The results showed that a optimum condition of the flavonoids extraction from Sinopodisma houshana was got when solid-liquid ratio equals to 1∶50 with the 80% methanol,and then extracting for 2 h under the extraction temperature of 85 ℃,and the extraction ratio was 2.02%.The total flavonoids content of Sinopodisma houshana were 20.198 mg/g and 4.047 mg/g,respectively(dry weight) by means of aluminium salt-coloration spectrophotometry method and high performance liquid chromatography.The results showed that the high-performance liquid chromatography could separate different flavonoids from impurities in Sinopodisma houshana,while aluminium salt-coloration spectrophotometry method was interfered by other phenols in it.So the high-performance liquid chromatography was a better method to detect flavonoids.
