摘    要:采用醋酸锂转化法,将一段目的基因转入到酵母体内,破坏酵母PEP4基因的表达。通过对目的基因的扩增和羧肽酶Y酶活检测验证基因敲除情况,并通过传代抗性试验与传代发酵稳定性实验验证突变株遗传稳定性良好。

Abstract:In this paper,we transform a gene into yeast genome so as to disrupt PEP4 gene by LiAc method.The mutant gene can be identified by PCR and testing enzyme activity of CPY.The mutant strain inheritance and ferment stability is well.
