高效液相色谱检测银杏茶中总黄酮含量

摘    要:采用正交试验法优选银杏茶中黄酮类化合物的最佳提取条件为25%HCl添加量4 mL,提取温度100℃,提取时间90 min。分析方法采用Eclipse XDB C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-0.4%磷酸溶液(体积比60∶40),流速1.0 mL/min,柱温30℃,检测波长为360 nm。槲皮素、山柰酚、异鼠李素在浓度20μg/mL~100μg/mL范围内线性良好,检出限分别为5.0、8.6、6.7 mg/kg。相对标准偏差(RSD)小于5%(n=6)。本法准确、快速、重现性好,可用于银杏茶的质量控制。

Abstract:The optimum extracting technology of flavonodis in ginkgo biloba tea was investigated by using the factorial and orthogonal design.The optimal conditions included 25 % HCl added 4 mL,extraction temperature 100 ℃,and extraction 1.5 h for each times.The separation was performed on Eclipse XDB C18 column(250 mm×4.6 mm,5 μm).The mobile phase was methanol-0.4 % phosphoric acid(60∶40,volumn ratio)with a flow rate of 1.0 mL/min.The detection wavelength was set at 360 nm and the column teperature was 30 ℃.A linear calibration curve of quercetin,isorhamnetin and keampferol was obtained in the concentration range of 20 μg/mL-100 μg/mL.The qualification limit were 5.0,8.6,6.7 mg/kg.The relative standard deviations(RSDs)were below 5 %(n=6).The method was simple,accurate and can be used for quality control of ginkgo biloba tea.
