牛乳中骨桥蛋白的分离纯化与鉴定

摘    要:从新鲜牛乳中分离纯化骨桥蛋白,并对其进行鉴定。运用生物化学技术,通过离心脱脂后,再经过离子交换层析、疏水层析、透析等技术达到分离纯化目的;随后对分离纯化得到的产物进行SDS-PAGE电泳、免疫印迹技术鉴定。分离纯化得到分子质量约为60 ku、纯度95%以上的高活性骨桥蛋白。通过上述方法可以得到纯度较高的骨桥蛋白,为将来进一步对骨桥蛋白活性及结构研究奠定了基础。

Abstract:Osteopontin was purified from bovine milk;certain biochemical characteristics were identified. Methods:Biological technique,including centrifugalization,ion-exchange chromatography,hydrophobic chromatography and dialysis was used.Osteopontin from bovine milk were identified by SDS-PAGE and Western Blot.One kind of osteopontin was obtained with high purity over 95% and with molecular weight 60 ku obtained from SDS-PAGE.Osteopontin of higher purification could be obtained through the above- mentioned methods.The research on its activities and structure may be developed on the basis of these re- sults.
