摘    要:大豆试样在酸性条件下水解后,采用岛津LC-10AtvpHPLC仪,紫外检测器(SPD-10Avvp),波长 254 nm,色谱柱:Inertsil ODS-3柱(5μm,4.6 mm×250 mm),流动相:甲醇:5% 醋酸溶液30%~70%梯度洗脱,流速:1.0 ml/min,柱温:50℃条件下,测定水解提取物中大豆异黄酮含量。测定结果表明:不同品种、产地的大豆中异黄酮含量为 1.40 mg~3.30 mg/g,大豆胚芽中异黄酮含量最高,为13.18mg/g;其它豆类,如红豆、绿豆、青豆、豌豆等异黄酮含量为0.37mg~1.2mg/g,此测定法能为大豆的选育种、栽培及大豆异黄酮含量评价提供依据。

Abstract:soybean samples were hydrolyzed in the presence of acid. soybean isoflavones content was determined by HPLC. shimadzu LC-10Atv HPLC system with UV detector (wavelength 254 nm) was used. For HPLC separations, a 250 mm×4.6 mm, 5μm Inertsil ODS-C18 column was used. The mobile phases were methanol and 5% acetic acid. The mobile phase gradient pro- gram was used (the ratio was 30% ~70%). The flow of the mobile phase was 1.0 ml/min. The co- lumn temperature was maintained at 50℃ with a column heater. The results showed that iso- flavones content in soybean with different kinds and from different origin was 1.40~3.30 mg/g. The maximum isoflavones content in soybean germ was 13.18 mg/g. Isoflavones content in other beans such as red bean, mung bean, green soybean and pea was 0.37~ 1.2 mg/g. The determina- tion method can provide basis for seed selection, breeding and cultivation of soybean and evalu- ation isoflavones content in soybean.
