Boxes of beef were examined when product was packed and when boxes were loaded out of five packing plants, when boxes were loaded into and loaded out of seven refrigerated warehouses, and when boxes were received and opened at 21 retail stores. At each stage of handling at each facility, the boxes to be examined were selected at random. For each selected box, the temperature of product at the centre of the box was measured, and the date of packing and the plant of origin were noted. When cuts were packed, the minimum, median and maximum temperatures were about 2, 6 and 18 °C, respectively. Temperatures were successively lower when boxes were loaded out of packing plants, into warehouses and out of warehouses. When loaded out of warehouses, the minimum, median and maximum temperatures were about −2, 1 and 8 °C, respectively. The ranges of temperatures were similar, but the median temperatures were about 2 or 1.5 °C, respectively, when boxes were received at or were opened at retail stores. At packing plants and warehouses, the temperatures of manufacturing and ground beef were lower than those of cuts, but at the retail store the temperatures of all types of product were similar. When boxes were opened at retail stores, the minimum, median and maximum ages of cuts were about 2, 20 and 130 days, respectively; and the corresponding ages for manufacturing and ground beef were 2, 7 and 56 days, respectively. The data indicate that boxed beef is generally cooled to and maintained at temperatures within the range sought by the meat industry. However, cooling to chiller temperatures of product that is packed while warm can take several days; and some product is held for times that are excessive in view of the temperatures of boxed beef.  相似文献

The development of a community of lactic acid bacteria from vacuum-packaged beef was investigated during a 6-week storage trial at 2 degrees C. The lactic acid bacteria population was monitored by using molecular techniques to identify a random sample of isolates at biweekly intervals during the storage trial. The polymerase chain reaction and a randomly amplified polymorphic DNA technique were used to identify and distinguish populations of lactic acid bacteria that developed during the storage trial. At week 0, the population of lactic acid bacteria was 3.5 log cfu/120 cm2 and by week 6, the population reached a maximum of 7.6 log cfu/120 cm2. A sampling from the week 0 population indicated a mixed community of Lactobacillus curvatus, Lactobacillus sakei and Leuconostoc spp. However, the sampling from week 6 indicated the population composition had changed to one where a single Leuconostoc strain predominated. This strain demonstrated antagonism towards the growth of other lactic acid bacteria isolated during the study. Additionally, the strain inhibited the growth of foodborne pathogens Escherichia coli O157:H7 and Listeria monocytogenes. DNA sequence data from the 16S rRNA gene suggested that the isolate may be a Leuconostoc gelidum strain.  相似文献

Deoxynivalenol (DON), a mycotoxin produced by several Fusaruim species, is a worldwide contaminant of foods and feeds. Because of the potential dangers due to accidental or intentional contamination of foods with DON, there is a need to develop a rapid and highly sensitive method for easy identification and quantification of DON. In this study, we have developed and utilized a competitive immunoassay technique to detect DON in various food matrixes and indoor air samples using an array biosensor. A DON-biotin conjugate, immobilized on a NeutrAvidin-coated optical waveguide, competed with the DON in the sample for binding to fluorescently labeled DON monoclonal antibodies. To demonstrate a simple procedure amenable for on-site use, DON-spiked cornmeal, cornflakes, wheat, barley, and oats were extracted with methanol-water (3:1) and assayed without cleanup or preconcentration. The limits of detection ranged from 0.2 ng/mL in buffer to 50 ng/g in oats. The detection limit of DON spiked into an aqueous effluent from an air sampler was 4 ng/mL.  相似文献

The responses to fertiliser‐N of winter wheat and winter barley grown on sandy soils were measured in 72 experiments in England from 1990 to 1994. Yield without fertiliser‐N (Y0) was c 1.1 t ha−1 greater following root crops than following cereals. Following potato crops given organic manures, Y0 was c 1.2 t ha−1 greater than following unmanured potato crops, but Y0 was no greater following sugarbeet to which organic manures had been applied. Only after the two driest winters was there sufficient variation in soil N supply in spring (SNSs) for this to show a relationship with Y0. However, Y0 increased with increasing N mineralisation during the growing season (AM) in the three years it was measured. There was no consistent effect of sowing date on Y0. Following potatoes, yield at optimum fertiliser‐N (Yopt) decreased as sowing date was delayed, but this was not so after cereals, sugarbeet or overall. There was no increase in Yopt with SNSS or AM, but Yopt decreased with increasing moisture stress (S) in June. The mean yield response to NoptY) was c 0.4 and 0.8 t ha−1 smaller following potatoes and sugarbeet respectively than following cereals, but not consistently so as there were large interactions between site, year and previous crop. Following root crops, ΔY was c 0.6 and 1.4 t ha−1 less after sugarbeet and potatoes respectively that had been given organic manures. Without the addition of organic manures, ΔY following potatoes was similar to that following cereals. Regression on SNSS and AM accounted for 28 and 15% respectively of the variance in ΔY. The optimum economic fertiliser‐N application (Nopt) was similar, at c 140 kg ha−1, following cereals and potatoes. Following sugarbeet, cereal Nopt was only c 110 kg ha−1. The differences according to previous crop reported here are consistent with mineralisation of crop residues on sandy soils being more rapid than on other soils; the potato residues were rapidly mineralised in autumn and lost by leaching over winter. Residues from later‐harvested sugarbeet were mineralised during the growing season of the subsequent cereal crop. Fertiliser‐N requirements were, at c 110–140 kg ha−1, smaller than has been found on other soil types, and less than current recommendations for wheat. Requirements were significantly reduced in years of drought stress. No differences were found in Nopt between wheat and barley. These data do not justify the current advice to invariably reduce fertiliser‐N to cereals following potatoes by 20–25 kg ha−1 on these sandy soils. On average a reduction of c 20 kg ha−1 could be made following sugarbeet, with a further reduction of c 40 kg ha−1 N if manures had been applied to the previous sugarbeet crop. A reduction of 40 kg ha−1 N could also be made where cereals followed a potato crop to which manures had been applied. Further refinements on the basis of measurements of soil mineral N could not be justified. Seasonal variation in N response due to drought stress makes recommendations difficult on these soils. Adopting the fertiliser‐N recommendations proposed here would produce N surpluses to the soil of c 37, 10 and 27 kg ha−1 respectively following cereals, sugarbeet and potatoes when cereal grain is removed but straw incorporated. On farms where straw is removed, N surplus would be largely eliminated. Our recommendation that no reduction in fertiliser‐N application to cereal crops grown on sandy soils should be made following potatoes will not increase fertiliser‐N use and is not expected to increase nitrate leaching. Some reduction in nitrate leaching may be achieved if recommendations following cereal crops and sugarbeet are made in accordance with the results reported here. © 2000 Society of Chemical Industry  相似文献

A rapid, high-throughput antimicrobial screening assay was developed using either a physical fluid extraction or a solvent extraction technique coupled to the commercially available Premi®Test. The solvent extraction approach was fully validated for a wide range of tissues and the fluid extraction approach partially validated for porcine muscle. Both procedures can detect a wide range of antimicrobial compounds at or below maximum residue limit concentrations. The use of a solvent extraction provides an enhanced test capable of detecting a wider range of drugs than the fluid extraction approach at or below half maximum residue limit levels in a variety of matrices. Biochemical methods for the class-specific identification of β-lactams and sulphonamides following initial screening were developed and validated. The approach is a significant improvement on existing methodologies as a tool for residues monitoring in surveillance programmes.  相似文献

Tensile properties of unidirectional Celion 6000 graphite/PMR 15 polyimide composites prepared by hot molding and cold molding processes were measured at room temperature and 316°C, the upper use temperature of the polyimide resin, at both 45 and 90° to the fiber axis. The resulting fractures were characterized by scanning electron microscopy and materialographic techniques. Variation in tensile properties with processing history occurred in the elastic modulus and strain to failure for specimens loaded at 90° at 316°C, and in the fracture stress, and hence the in-plane shear stress, for those loaded at 45° at room temperature. Significant plastic deformation was observed in the 45° orientation at 316°C for material produced by both processing methods. In general, fracture occurred by both failure within the matrix and at the fiber-matrix interface; the degree of interfacial failure increased with temperature. Secondary cracking below the primary fracture surface also was observed.  相似文献

Electroretinographic (ERG) spectra show that juvenile goldfish have ultraviolet (UV) sensitivity but adults do not. Chromatic adaptation data suggest mediation by UV cones. ERG spectra from eye cups and spectrophotometry of lenses show that the loss of UV sensitivity with age does not result from lens changes. Our results contribute to a growing literature on UV cone mechanisms and visual development in fish.  相似文献

Human milk triacylglycerols were separated by high-performance liquid chromatography. A 5-μ Supelcosil LC-18 column (Supelco, Inc., Bellefonte, PA) was used with acetone/acetonitrile (64∶36, vol/vol) as mobile phase. Triacylglycerols were tentatively identified based on theoretical carbon number and relative retention time. Despite changes resulting from dietary fat variation, the major component triacylglycerols were those composed of palmitic, oleic and linoleic acids. Triacylglycerols with palmitic, stearic and oleic acids were present as minor components. Fatty acids were quantified by gas chromatography relative to an internal standard. Ratios of n−6/n−3 fatty acids were found to be high than previously reported. Drug delivery to the eye remains a challenge due to low compliance and high amounts of drug loss resulting from anatomical barriers. Contact lens-based delivery systems offer many advantages since their increased corneal residence time can increase drug bioavailability. A releasable, sustained delivery system from a contact lens has been evaluated in the current work. Using a labile lactide spacer, comprised of poly(ethylene glycol) methacrylate, varying numbers of lactide units and succinic anhydride, a model drug, R-roscovitine, was covalently incorporated into model silicone lenses (consisting of 80 mol% N,N-dimethylacrylamide and 20 mol% of a modified silicone). This enabled a greater amount of the drug to remain in the material following extraction with isopropyl alcohol, a common purification procedure used in lens manufacturing. The incorporation of the tether and drug did not substantially alter the bulk properties of the materials, with their water content remaining between 32% and 36% and their transmittance remaining at over 90% at 600 nm. Drug release from these materials exceeded 2 weeks, with the longer lactide tethers showing greater release as expected. This demonstrates that these covalently modified materials have the potential to be used to deliver drugs in a sustained manner. © 2022 Society of Industrial Chemistry.  相似文献
