A wide variety of biological activities including the major metabolic actions of insulin is regulated by phosphatidylinositol (PI) 3-kinase. However, the downstream effectors of the various signaling pathways that emanate from PI 3-kinase remain unclear. Akt (protein kinase B), a serine-threonine kinase with a pleckstrin homology domain, is thought to be one such downstream effector. A mutant Akt (Akt-AA) in which the phosphorylation sites (Thr308 and Ser473) targeted by growth factors are replaced by alanine has now been shown to lack protein kinase activity and, when overexpressed in CHO cells or 3T3-L1 adipocytes with the use of an adenovirus vector, to inhibit insulin-induced activation of endogenous Akt. Akt-AA thus acts in a dominant negative manner in intact cells. Insulin-stimulated protein synthesis, which is sensitive to wortmannin, a pharmacological inhibitor of PI 3-kinase, was abolished by overexpression of Akt-AA without an effect on amino acid transport into the cells, suggesting that Akt is required for insulin-stimulated protein synthesis. Insulin activation of p70 S6 kinase was inhibited by approximately 75% in CHO cells and approximately 30% in 3T3-L1 adipocytes, whereas insulin-induced activation of endogenous Akt was inhibited by 80 to 95%, by expression of Akt-AA. Thus, Akt activity appears to be required, at least in part, for insulin stimulation of p70 S6 kinase. However, insulin-stimulated glucose uptake in both CHO cells and 3T3-L1 adipocytes was not affected by overexpression of Akt-AA, suggesting that Akt is not required for this effect of insulin. These data indicate that Akt acts as a downstream effector in some, but not all, of the signaling pathways downstream of PI 3-kinase.  相似文献

We had reported that a double-strand gap (ca. 300 bp long) in a duplex DNA is repaired through gene conversion copying a homologous duplex in a recB21 recC22 sbcA23 strain of Escherichia coli, as predicted on the basis of the double-strand break repair models. We have now examined various mutants for this repair capacity. (i) The recE159 mutation abolishes the reaction in the recB21C22 sbcA23 background. This result is consistent with the hypothesis that exonuclease VIII exposes a 3'-ended single strand from a double-strand break. (ii) Two recA alleles, including a complete deletion, fail to block the repair in this recBC sbcA background. (iii) Mutations in two more SOS-inducible genes, recN and recQ, do not decrease the repair. In addition, a lexA (Ind-) mutation, which blocks SOS induction, does not block the reaction. (iv) The recJ, recF, recO, and recR gene functions are nonessential in this background. (v) The RecBCD enzyme does not abolish the gap repair. We then examined genetic backgrounds other than recBC sbcA, in which the RecE pathway is not active. We failed to detect the double-strand gap repair in a rec+, a recA1, or a recB21 C22 strain, nor did we find the gap repair activity in a recD mutant or in a recB21 C22 sbcB15 sbcC201 mutant. We also failed to detect conservative repair of a simple double-strand break, which was made by restriction cleavage of an inserted linker oligonucleotide, in these backgrounds. We conclude that the RecBCD, RecBCD-, and RecF pathways cannot promote conservative double-strand break repair as the RecE and lambda Red pathways can.  相似文献

In order to examine the mechanics of crack initiation at the free interface edge of a microcomponent on a substrate, delamination tests are carried out for two specimen shapes of Cr microdots on a SiO2 substrate. The microdots of the first specimen are shaped like the frustum of a round cone. The Cr microdots are successfully delaminated from the SiO2 substrate in a brittle manner and the critical load is measured by atomic force microscopy (AFM) with a lateral loading apparatus. Stress analysis reveals that a singular stress field exists near the interface edge and the strength for the crack initiation is governed by the intensified normal stress field. The critical stress intensity parameter is evaluated as KσC ≈ 0.24 MPa m0.39. Similar delamination tests are conducted for microdots shaped like the frustum of an oval cone. The stress distributions at the crack initiation of this specimen shape show a higher normal stress than the first specimen shape in the region near the interface edge of about x < 40 nm, while it is lower in the region of about x > 50 nm (x: distance from the edge). This suggests a limitation of conventional fracture mechanics: namely, the crack initiation in these specimens is not uniquely governed by the intensity of the singular field. It is found that the delamination crack is initiated when the averaged stress σya in the region of 90-130 nm reaches 190-270 MPa, regardless of the specimen shape. This indicates that the dominant stress region of crack initiation is roughly estimated as 90-130 nm and the criterion is given in terms of the averaged stress in the region.  相似文献

LiNi0.5Mn0.5O2 was heat-treated under high oxygen-pressure and Ar-reducing conditions, and then the cathode properties, thermodynamic stability and average and local structures were investigated. From X-ray diffraction and ICP measurements, it was found that the pristine LiNi0.5Mn0.5O2 had a single phase of the layered rock-salt structure although the Ni content was slightly rich compared with the nominal one. These characteristics were kept even after the heat-treatments. Charge–discharge cycle tests clarified that the cycle performance of LiNi0.5Mn0.5O2 was improved by both the reducing and oxidizing treatments. From neutron diffraction and X-ray absorption fine structure measurements, the local distortion around the transition metal, especially Ni, was supposed to be one of the important factors to determine the cathode properties. It was also found that the sample with higher thermodynamic stability exhibited better capacity retention in the discharge–charge cycle tests.  相似文献

Using the instrumented fuel assemblies (IFA) installed in the Japan Power Demonstration Reactor (JPDR)-II core, fluctuations of the inlet and outlet channel flow rates were observed under both conditions of at-power operation and cold core flow circulation. The correlation analysis revealed that the flow fluctuations in any IFA channel showed almost uncorrelated cross-covariance function with other IFA channel flow. To explain the mechanism of the channel flow fluctuations, some hypothetical idea is introduced.  相似文献

In order to increase the photocatalytic activity on TiO2 thin film per its external surface area, the structure of flat thin film was modified by adding a small amount of polyethylene glycol (PEG) to TiO2 sol solution. By firing PEG contained in a TiO2 gel film, a porous structure was developed. The photocatalytic activities of the thin films prepared thus were evaluated by the degradation of 2-propanol in the aqueous solutions under black light illumination. The photocatalytic activity of TiO2 thin film prepared with added PEG 400 by 2.6 wt% or PEG 2000 by 9.5 wt% was increased by about 30% as compared to that prepared without added PEG.  相似文献

Stress fields on elastic-creep bi-material interfaces with different geometry of the interface edge are analyzed by finite element method. The results reveal that the stress highly concentrates near the interface edge at the loading instant and it gradually decreases as the creep-dominated zone expands from the small-scale creep to the large-scale creep. The stress singularity due to creep which resembles the HRR stress singularity appears near the interface edge in all cases. The stress intensity near the interface edge time-dependently decreases and becomes constant when the transition reaches the steady state. The magnitude is scarcely influenced by the edge shape of elastic material, though it depends on the edge shape of creep material. The stress intensity during the transition can be approximately predicted by the J-integral at the loading instant.  相似文献

A superalloy with a thermal barrier coating (TBC) simulating a gas turbine blade is exposed to a high-temperature environment to develop a method for predicting the local temperature and Al content in a bond coat (BC). The Al content decreases with an increase in the test time due to the Al transport induced by the oxidation of the BC and the interdiffusion between the BC and the substrate. This brings about Al-decreased layer (ADL) at the boundary between the BC and the top coat. The thickness of the ADL increases in proportion to the square root of the test time, and the temperature dependence of the growth rate shows an Arrhenius-type behavior. Based on this relation, the local temperature of an in-service blade can be estimated by measuring the ADL thickness when the operation time is known. The Al content decreases in proportion to the ADL thickness. The prediction method of the Al content based on the relation is also presented.  相似文献

Abatement of CO2 emission is one of the most important issues in the 21st century regarding preservation of the earth environment. This paper addresses a utility operations planning problem for distributed energy management systems (DEMSs), where we are to obtain optimal plans that minimize both costs and CO2 emissions. A DEMS consists of multiple entities that seek their own economic profits. In this paper, we give a mathematical formulation of the utility operations planning problem for each entity, and propose an energy trading market, which utlizes a multi‐attribute auction protocol in order to deal with both a price and a CO2 emission rate. Experimental results show that collaboration among entities through the market provides a more profitable plan for each entity and abatement of CO2 emission is also achieved. © 2007 Wiley Periodicals, Inc.
