In bacteria, selenocysteine (Sec) is incorporated into proteins via the recoding of a particular codon, the UGA stop codon in most cases. Sec-tRNASec is delivered to the ribosome by the Sec-dedicated elongation factor SelB that also recognizes a Sec-insertion sequence element following the codon on the mRNA. Since the excess of SelB may lead to sequestration of Sec-tRNASec under selenium deficiency or oxidative stress, the expression levels of SelB and tRNASec should be regulated. In this bioinformatic study, I analyzed the Rhizobiales SelB species because they were annotated to have a non-canonical C-terminal extension. I found that the open reading frame (ORF) of diverse Alphaproteobacteria selB genes includes an entire tRNASec sequence (selC) and overlaps with the start codon of the downstream ORF. A remnant tRNASec sequence was found in the Sinorhizobium meliloti selB genes whose products have a shorter C-terminal extension. Similar overlapping traits were found in Gammaproteobacteria and Nitrospirae. I hypothesized that once the tRNASec moiety is folded and processed, the expression of the full-length SelB may be repressed. This is the first report on a nested tRNA gene inside a protein ORF in bacteria.  相似文献

We have developed stacked nanocarbon polymer actuators that are composed of several nanocarbon polymer actuator films using nonwoven fabric as insulation layers. The nonwoven fabric prepared through electrospinning methods has extremely-low-density structures, which do not significantly prevent the motions of each nanocarbon actuator layer. Therefore, stacking several thin nanocarbon polymer actuators using nonwoven fabric as insulation layers is expected to increase generated force without decreasing the displacement of a one-layer actuator. We have prepared stacked actuators with one, two, three, four, and seven layers using this method. The displacement and blocking force of these actuators are measured and compared with those of one-layer actuators of different thicknesses. Displacement is weakly dependent on the thickness of the actuator films of the stacked actuators. On the contrary, it decreases considerably as the thickness of the actuator film of the one-layer actuator increases. In both cases, blocking force is proportional to the thickness of actuator films. We have developed a stacked actuator model based on a trilayer actuator model and confirmed the experimental results using the model.  相似文献

Hepatitis B often progresses to decompensated liver cirrhosis requiring orthotopic liver transplantation (OLT). Although newer nucleos(t)ide analogues result in >90% viral and hepatitis activity control, severely decompensated patients still need OLT because of drug-resistant virus, acute exacerbation, or hepatocellular carcinoma. Acute hepatitis B is also an indication for OLT, because it can progress to fatal acute liver failure. After OLT, the hepatitis B recurrence rate is >80% without prevention, while >90% of transplant recipients are clinically controlled with combined hepatitis B immunoglobulin (HBIG) and nucleos(t)ide analogue treatment. However, long-term HBIG administration is associated with several unresolved issues, including limited availability and extremely high cost; therefore, several treatment protocols with low-dose HBIG, combined with nucleos(t)ide analogues, have been investigated. Another approach is to induce self-producing anti-hepatitis B virus (HBV) antibodies using an HBV envelope (HBs) antigen vaccine. Patients who are not HBV carriers, such as those with acutely infected liver failure, are good candidates for vaccination. For chronic HBV carrier liver cirrhosis patients, a successful vaccine response can only be achieved in selected patients, such as those treated with experimentally reduced immunosuppression protocols. The present protocol for post-OLT HBV control and the future prospects of newer treatment strategies are reviewed.  相似文献

Microelectromechanical system (MEMS) actuators essentially have movable silicon structures where the mechanical motion can be activated electronically. The microscanner is one of the most successfully commercialized MEMS devices which are widely used for collecting optical information, manipulating light, and displaying images. While silicon is abundant, it is also brittle and stiff and when microprocessed, defects are not uncommon. These defects result in weakness under torsional stress and this has been the key factor limiting the scanning performance of the microscanner. Here a metallic glass (MG)‐based microscanner is reported with MG as the material for the moving torsion bars. The low elastic modulus, high fracture toughness, and high strength of MG offers, for the first time, an ultralarge rotating angle of 146° with power consumption lowered to the microwatt range, and a smaller driving force and better actuation performance, than conventional single crystal silicon and polycrystalline silicon. The high spatial resolution and large scanning field of the MG‐based microscanner are demonstrated in the tomographic imaging of a human finger. This development of an MG‐based MEMS possibly opens a new field of low‐powered MEMS devices with extreme actuation and enhanced sensing.  相似文献

Although amorphous silica nanoparticles are widely used in the production of food products (e.g., as anticaking agents), there is little information available about their absorption and biological effects after oral exposure. Here, we examined the in vitro intestinal absorption and in vivo biological effects in mice of orally administered amorphous silica particles with diameters of 70, 300, and 1,000 nm (nSP70, mSP300, and mSP1000, respectively) and of nSP70 that had been surface-modified with carboxyl or amine groups (nSP70-C and nSP70-N, respectively). Analysis of intestinal absorption by means of the everted gut sac method combined with an inductively coupled plasma optical emission spectrometer showed that the intestinal absorption of nSP70-C was significantly greater than that of nSP70. The absorption of nSP70-N tended to be greater than that of nSP70; however, the results were not statistically significant. Our results indicate that silica nanoparticles can be absorbed through the intestine and that particle diameter and surface properties are major determinants of the degree of absorption. We also examined the biological effects of the silica particles after 28-day oral exposure in mice. Hematological, histopathological, and biochemical analyses showed no significant differences between control mice and mice treated with the silica particles, suggesting that the silica nanoparticles evaluated in this study are safe for use in food production.  相似文献

This paper presents results from a behavioral demand response trial targeted to both grid and residential peak hours, in which weekly feedback via paper-based reports, real-time feedback via an in-home display, 30-minute tiered rate, and email prompts are adopted to almost 230 residential customers of a condominium in Funabashi, a city located in Greater Tokyo. Through a randomized experiment, we find that the peak saving impact during grid peak hours (1–4 pm, weekdays only) was 11.6 %, given that all the four interventions provided all at once. In addition, the results show that the variation in peak saving effects by household characteristics exists, and the variation differs among packages of peak saving interventions. Furthermore, we analyzed how much informational elements in weekly reports are considered as useful by residential customers. These results suggest that feedback-based approaches for peak saving can promote households’ energy conservation behavior.  相似文献

The authors have established a method of determining mercury and gold in severely polluted environmental samples using prompt gamma-ray analysis (PGA) and instrumental neutron activation analysis (INAA). Since large amounts of mercury are constantly being released into the environment by small-scale gold mining in many developing countries, the mercury concentration in tailings and water has to be determined to mitigate environmental pollution. Cold-vapor atomic absorption analysis, the most pervasive method of mercury analysis, is not suitable because tailings and water around mining facilities have extremely high mercury concentrations. On the other hand, PGA can determine high mercury concentrations in polluted samples as it has an appropriate level of sensitivity. Moreover, gold concentrations can be determined sequentially by using INAA after PGA. In conclusion, the analytical procedure established in this work using PGA and INAA is the best way to evaluate the degree of pollution and the tailing resource value. This method will significantly contribute to mitigating problems in the global environment.  相似文献

Polyaniline (PANI) or carbon black (CB) was used as a conductive additive in porous electrodes of a dry-type carbon nanotube (CNT)-polymer actuator to improve the actuation properties. We estimated the strain from the bending motion of the actuator in the frequency range of 200–0.005 Hz. The generated stress was calculated from the Young’s modulus of the electrode film and the maximum strain. Eight different types of electrode films were prepared by changing the mixed amount of PANI or CB into the CNT electrode film. When a same amount of PANI (50 mg) against CNT (50 mg) was added to the CNT electrode (in the case of CNT/PANI(50/50)), the strain of CNT/PANI(50/50) was increased to be almost three times larger than that of CNT(50) (without any additives). As a result, CNT/PANI(50/50) showed more than five times the generated stress compared to CNT(50). Using 40 mg of CB as an additive, CNT/CB(50/40) showed a more than four times larger stress compared to CNT(50). The electrochemical properties such as conductivity and capacitance were also investigated and discussed. An asymmetrical three-layered actuator element was fabricated to investigate the actuation mechanism. The results imply that the anode contracts and the cathode expands.  相似文献
