Hemorphins are known for their role in the control of blood pressure. Recently, we revealed the positive modulation of the angiotensin II (AngII) type 1 receptor (AT1R) by LVV-hemorphin-7 (LVV-H7) in human embryonic kidney (HEK293) cells. Here, we examined the molecular binding behavior of LVV-H7 on AT1R and its effect on AngII binding using a nanoluciferase-based bioluminescence resonance energy transfer (NanoBRET) assay in HEK293FT cells, as well as molecular docking and molecular dynamics (MD) studies. Saturation and real-time kinetics supported the positive effect of LVV-H7 on the binding of AngII. While the competitive antagonist olmesartan competed with AngII binding, LVV-H7 slightly, but significantly, decreased AngII’s kD by 2.6 fold with no effect on its Bmax. Molecular docking and MD simulations indicated that the binding of LVV-H7 in the intracellular region of AT1R allosterically potentiates AngII binding. LVV-H7 targets residues on intracellular loops 2 and 3 of AT1R, which are known binding sites of allosteric modulators in other GPCRs. Our data demonstrate the allosteric effect of LVV-H7 on AngII binding, which is consistent with the positive modulation of AT1R activity and signaling previously reported. This further supports the pharmacological targeting of AT1R by hemorphins, with implications in vascular and renal physiology. 相似文献

Manganese‐doped tin oxide (SnO2:Mn) thin films were deposited on glass substrates by the sol–gel dip coating technique. The effect on structural, morphological, magnetic, electrical, and optical properties in the films with different Mn concentrations (0–5 mol%) were investigated. X‐ray diffraction patterns (XRD) showed the deterioration of crystallinity with increase in Mn‐doping concentration. Scanning electron microscopy (SEM) studies showed an inhibition of grain growth with an increase in Mn concentration. X ray photoelectron spectroscopy (XPS) revealed the presence of Sn4+ and Mn3+ in SnO2: Mn films. SnO2: Mn films show ferromagnetic and paramagnetic behavior. These SnO2:Mn films acquire n‐type conductivity for 0–3 mol% (SnO2 ‐ Sn0.97Mn0.03O2) ‐doping concentration and p type for 5 mol% Mn‐doping concentration(Sn0.95Mn0.05O2) in SnO2 films. An average transmittance of > 75% (in UV‐Vis region) was observed for all the SnO2:Mn films. Optical band gap energy of SnO2: Mn films were found to vary in the range 3.55 to 3.71 eV with the increase in Mn‐doping concentration. Photoluminescence (PL) spectra of the films exhibited an increase in the emission intensity with increase in Mn‐doping concentration which may be due to structural defects or luminescent centers, such as nanocrystals and defects in the SnO2. Despite the established superiority in finding the global as well as well-spread Pareto optimal (PO) points, the need of more numbers of function evaluations for population based evolutionary optimization techniques leads to a computationally demanding proposal. The case becomes more miserable if the function evaluations are carried out using a first principle based computationally expensive model, making the proposal not fit for online usage of the application. In this work, a Kriging based surrogate model has been proposed to replace a computationally expensive model to save execution time while performing an optimization task. A multi-objective optimization study has been carried out for the bulk vinyl acetate polymerization with long-chain branching using these surrogate as well as expensive models and Kriging PO solutions similar to those found by the first principle models are obtained with a close to 85% savings in function evaluations. 相似文献
