Effects of the Anti-Tumorigenic Agent AT101 on Human Glioblastoma Cells in the Microenvironmental Glioma Stem Cell Niche

Glioblastoma (GBM) is a barely treatable disease due to its profound chemoresistance. A distinct inter- and intratumoral heterogeneity reflected by specialized microenvironmental niches and different tumor cell subpopulations allows GBMs to evade therapy regimens. Thus, there is an urgent need to develop alternative treatment strategies. A promising candidate for the treatment of GBMs is AT101, the R(-) enantiomer of gossypol. The present study evaluates the effects of AT101, alone or in combination with temozolomide (TMZ), in a microenvironmental glioma stem cell niche model of two GBM cell lines (U251MG and U87MG). AT101 was found to induce strong cytotoxic effects on U251MG and U87MG stem-like cells in comparison to the respective native cells. Moreover, a higher sensitivity against treatment with AT101 was observed upon incubation of native cells with a stem-like cell-conditioned medium. This higher sensitivity was reflected by a specific inhibitory influence on the p-p42/44 signaling pathway. Further, the expression of CXCR7 and the interleukin-6 receptor was significantly regulated upon these stimulatory conditions. Since tumor stem-like cells are known to mediate the development of tumor recurrences and were observed to strongly respond to the AT101 treatment, this might represent a promising approach to prevent the development of GBM recurrences.     

高岭土性质对催化裂化催化剂性能的影响

采用X射线荧光分析仪、X射线衍射仪、N2物理吸附仪、场发射扫描电子显微镜等仪器表征了苏州、广西和美国佐治亚州3个产地的高岭土，并以此3种高岭土为原料制备了模型催化裂化（FCC）催化剂，在ACE评价装置上对比了模型催化剂的反应性能。结果表明:苏州及广西高岭土主要组分为高岭石，佐治亚高岭土主要组分为地开石及珍珠陶土；苏州高岭土呈片状，还含有少量棒状颗粒；广西高岭土呈多层片状，晶粒粒径较大；佐治亚高岭土呈薄片状，晶粒粒径较小；3种高岭土制备的模型催化剂反应活性、Na2O及RE2O3质量分数相近；广西高岭土制备的模型催化剂具有最大的孔体积和磨损指数，但比表面积最小，具有较强的重油转化能力，其目标产物（液化气+汽油）和副产物（干气+焦炭）收率都高于苏州、佐治亚高岭土制备的催化剂的。     

应用独立分量分析的激光测风雷达湍流频谱分解

针对大型风力发电机机组中常见的脉动湍流、风机尾流与涡流等湍流信号,研究了利用自然梯度下降的独立分量分析方法的湍流频谱分离效果,以区分中心风速与湍流信号,提高风机机组的综合工作效率。首先分析了风机组中常见湍流信号的后向散射与频谱分布特点,然后依据这些特点设计了对应的独立分量分析模型。在仿真结果符合要求的基础上,进行了双目激光雷达天线的风速采集与实际分离效果检测。实验结果表明,在大气折射率结构常数C2n≤10-14同时广义大气常数α≥4的通常情况下,利用双目信号能够分离出一个湍流中心和一个中心风速。对1 s内两个谱峰的波动范围进行统计,获得(2.59±0.05)MHz的中心风速以及(1.22±0.19)MHz的湍流中心估计,且二者的平均信噪比分别为25.93 dB和31.01 dB,能够在获得稳定的中心风速估计的同时得到一个较为稳定的湍流中心估计。

为 解 决 远 程 控 制 时 ，操 作 者 无 法 对 操 作 现 场 全 局 掌 握 的 问 题 ，通 过 视 觉 方 法 对 现 场 环 境 进 行 建模 ，利 用 虚 拟 仿 真 技 术 ，实 时 显 示 在 操 作 界 面 。 利 用 视 觉 算 法 处 理 双 目 相 机 获 得 的 目 标 图 像 ，获 取 目 标 的 位置 信 息 。 利 用 仿 真 平 台 对 目 标 位 置 信 息 进 行 二 次 处 理 ，在 现 场 环 境 的 模 拟 模 型 中 将 目 标 物 体 仿 真 建 模 。 最后 在 操 作 界 面 上 利 用 虚 拟 提 示 ，引 导 操 作 者 完 成 任 务 。     

Direct Injection of CRISPR/Cas9-Related mRNA into Cytoplasm of Parthenogenetically Activated Porcine Oocytes Causes Frequent Mosaicism for Indel Mutations

Some reports demonstrated successful genome editing in pigs by one-step zygote microinjection of mRNA of CRISPR/Cas9-related components. Given the relatively long gestation periods and the high cost of housing, the establishment of a single blastocyst-based assay for rapid optimization of the above system is required. As a proof-of-concept, we attempted to disrupt a gene (GGTA1) encoding the α-1,3-galactosyltransferase that synthesizes the α-Gal epitope using parthenogenetically activated porcine oocytes. The lack of α-Gal epitope expression can be monitored by staining with fluorescently labeled isolectin BS-I-B₄ (IB4), which binds specifically to the α-Gal epitope. When oocytes were injected with guide RNA specific to GGTA1 together with enhanced green fluorescent protein (EGFP) and human Cas9 mRNAs, 65% (24/37) of the developing blastocysts exhibited green fluorescence, although almost all (96%, 23/24) showed a mosaic fluorescent pattern. Staining with IB4 revealed that the green fluorescent area often had a reduced binding activity to IB4. Of the 16 samples tested, six (five fluorescent and one non-fluorescent blastocysts) had indel mutations, suggesting a correlation between EGFP expression and mutation induction. Furthermore, it is suggested that zygote microinjection of mRNAs might lead to the production of piglets with cells harboring various mutation types.

嵌入式实时操作系统内核混合代码的自动化验证框架

"如何构造高可信的软件系统"已成为学术界和工业界的研究热点.操作系统内核作为软件系统的基础组件，它的安全可靠是构造高可信软件系统的重要环节.为了确保操作系统内核的安全可靠，将形式化方法引入到操作系统内核验证中，提出了一个自动化验证操作系统内核的框架.该验证框架包括：（1）分别对C语言程序和混合语言程序（C语言和汇编语言）进行验证；（2）在混合语言程序验证中，为汇编程序建立抽象模型，并将C语言程序和抽象模型粘合形成基于C语言验证工具可接收的验证模型；（3）从规范中提取性质，基于该自动验证工具，对性质完成自动验证；（4）该框架不限于特定的硬件架构.成功地运用该验证框架对两种不同硬件平台的嵌入式实时操作系统内核μC/OS-II进行了验证.结果显示：利用该框架在对两个不同的硬件平台上内核验证时，框架的可重复利用率很高，高达到88%，虽然其抽象模型需要根据不同的硬件平台进行重构.在对基于这两种平台的操作系统内核验证中，分别发现了10~12处缺陷.其中，在ARM平台上两处与硬件相关的问题被发现.实验表明，该方法对不同硬件平台的同一个操作系统分析验证具有一定的通用性.     

高密度聚乙烯/丁苯橡胶热塑性硫化胶的形状记忆性能

采用动态硫化法制备了高密度聚乙烯(HDPE)/丁苯橡胶(SBR)热塑性硫化胶(TPV),研究了橡塑比、定型温度、回复温度及增容剂苯乙烯-丁二烯-苯乙烯嵌段共聚物(SBS)对HDPE/SBR TPV形状记忆性能的影响,观察了其微观结构,提出了形状记忆行为机制。结果表明,HDPE/SBR TPV具有典型的"海-岛"结构,拉伸后其表面存在明显的取向结构;SBS能显著提高HDPE/SBR TPV的形状记忆性能,当SBS用量为6份、定型温度和回复温度均为120℃时,HDPE/SBR TPV的形状记忆性能最佳。     

Using a Discrete-Event System Specifications (DEVS) for designing a Modelica compiler

We introduce a new architecture for the design of a tool for modeling and simulation of continuous and hybrid systems. The environment includes a compiler based on Modelica, a modular and a causal standard specification language for physical systems modeling (the tool supports models composed using certain component classes defined in the Modelica Standard Library, and the instantiation, parameterization and connection of these MSL components are described using a subset of Modelica). Models are defined in Modelica and are translated into DEVS models. DEVS theory (originally defined for modeling and simulation of discrete event systems) was extended in order to permit defining these of models. The different steps in the compiling process are show, including how to model these dynamic systems under the discrete event abstraction, including examples of model simulation with their execution results.
