大豆品种吉林35胚尖再生体系的建立及对农杆菌的敏感性

摘    要:为建立一个简便、高效、稳定的大豆胚尖再生体系,以吉林35为材料,胚尖为外植体,研究氯气、升汞、酒精3种消毒方法对不定芽出芽率的影响和6-BA对不定芽再生率的影响。同时,为检测吉林35的农杆菌易感性,对平安8、东农42、吉林47和吉林35四个大豆品种进行gus基因组织化学染色。研究表明,酒精消毒法对种子伤害较小,可以保证较高的出芽率。6-BA对不定芽再生作用显著,单独使用或配合IBA使用均可获得较高的再生率。gus基因组织化学染色结果表明吉林35的农杆菌易感性强,明显优于其它品种。吉林35胚尖外植体再生率较高且对农杆菌较敏感,是遗传转化良好的受体材料。

Establishment of an embryonic tip regeneration system of Jilin35 soybean and susceptivity to Agrobacterium tumefaciens

Abstract:To establish a convenient efficient and stable soybean regeneration system,using the embryotic tip of Jilin35 as explant,we studied the influence on inductivity of 3 different sterilizing methods including chlorine gas,corrosive sublimate and alcohol.The influence of different 6-BA concentration on shoot regeneration frequencies was also investigated.To determine the susceptivity to A.tumefaciens of Jilin35 histochemical assays of gus activity was carried out on four soybean cultivars Ping' an8,Dongnong42,Jilin47 and Jilin 35.Our research showed that sterilization of alcohol injured seeds less than the other two methods,and higher shoot growth frequencies could be achieved.6-BA significantly increased the shoot regeneration frequencies when 6-BA alone or a combination of 6-BA and IBA were used.The result of GUS assay suggested that Jilin35 showed higher sensitivity than other genotypes.The explant embryonic tip of Jilin35 obtained higher regeneration frequencies and was more sensitive to A.tumefaciens,which could be used as an excellent receptor material of genetic transformation.
