摘    要:建立测定食用动物油脂中苯甲酸与山梨酸含量的反相高效液相色谱法。采用反相高效液相色谱法,色谱柱为Wondasil C18(4.6mm×250mm,5μm),以甲醇-0.02mol/L乙酸铵溶液(5∶95)为流动相,检测波长为230nm,测定食用动物油脂中苯甲酸与山梨酸的含量。柱温为30℃,流速1.0mL/min,进样量10μL。结果表明,苯甲酸与山梨酸在0~320μg/mL范围内,峰面积与其浓度线性关系良好,相关系数均为0.999 76,平均回收率分别为99.0%、99.1%,重复性试验相对标准偏差分别为0.88%和0.71%(n=9)。本法准确、简便、快速,适用于食用动物油脂中苯甲酸与山梨酸含量的测定,建议用于食用动物油脂的质量控制。

Abstract:To establish RP-HPLC methods for determination of benzoic acid and sorbic acid in edible animal oil, benzoic acid and sorbic acid in edi- ble animal oil were determinated by RP-HPLC with a Wondasil Cls (4.6mm×250mm, 5μm) column. The mobile phase was a mixture of methyl alcohol- 0.02 mol/L ammonium acetate solution (5:95), the detection wavelength was 230 nm, the column temperature was 30℃ and flow rate was 1.0ml/min with the sample size of 10 IxL. Result showed that when the benzoic acid and sorbic acid were within the range of 0-320 μg/mL, the peak area and concentration was in good linear relationship with the correlation coefficient of 0.999 76. The average recovery of benzoic acid and sorbic acid was 99.0% and 99.1% and the relative standard deviation reproducibility test was 0.88% and 0.71%(n=9), respectively. This method was simple, quick, and accurate. It was suitable for the determination of benzoic acid and sorbic acid in edible animal oil and recommended for the quality control in edi- ble animal oil.
