This paper studies the ozone treatment effect on degradation of aflatoxin B1 (AFB1) in corn with different moisture content (MC). The toxicity of the degradation products (DPs) of the ozone-treated AFB1-Contaminated Corn (ACC) was also evaluated using the human hepatocellular carcinoma cell line (HepG2) as model cells. The degradation rate of AFB1 in corn increases with ozone concentration and treatment time. The results showed that ACC with 13.47% MC was easier to be degraded by ozone than with 20.37% MC. Treated with 90 mg L−1 ozone for 20 min and 40 min, AFB1 in corn with 13.47% MC decreased from 83 μg kg−1 to 18.12 μg kg−1 and 9.9 μg kg−1, respectively, well meeting the China National Standard of AFB1 in corn (20 μg kg−1). In order to evaluate the safety of ozone used on ACC, the impacts of AFB1 as well as untreated and ozone-treated ACC with the same level of AFB1 content on HepG2's survival rate, morphology, and apoptosis were studied. The results showed that ACC had high cell toxicity while the toxicity of ozone-treated ACC had no significant difference with that of the AFB1-free culture solution. It is concluded that ozonation can quickly and effectively degrade AFB1 in corn and diminish ACC's toxicity, and therefore, ozonation is expected to be an effective, fast, and safe method for AFB1 degradation in ACC. 相似文献

Soybean sauce, a Chinese traditional and daily condiment, is often contaminated by aflatoxin B1. An extract-free immunochromatographic assay was proposed based on fluorescent microspheres probe for the' detection of aflatoxin B1 in soybean sauce. The probe was prepared by coupling fluorescent microspheres with anti-aflatoxin B1 antibody by the 1-ethyl-3(3-dimethylaminopropyl) carbodiimides hydrochloride-mediated method. The background from the soybean sauce sample on strip was eliminated because of the optical property of the probe. The sample without extracting procedure was directly detected by diluting with 10% methanol solution. The visible detection limit for the qualitative analysis of aflatoxin B1 in the proposed method was 2.5 μg/L, which was lower than the maximum level of 5 μg/L set by the Chinese government. The results were well agreed with those obtained by liquid chromatography-mass spectrometry (LC-MS). The method showed satisfactory characteristics, such as rapid detection, easy operation, and high sensitivity, and can thus be applied for the large-scale and on-site screening of soybean sauce contaminated with aflatoxin B1. 构建基于磁荧光纳米材料的免疫层析试纸模式,弥补现在免疫层析技术的不足,为更灵敏的免疫学快速检测提供技术支撑。以呕吐毒素(deoxynivalenol,DON)为靶标,采用溶剂热法制备羧基修饰的超顺磁颗粒,碳二亚胺法将磁颗粒、绿色荧光蛋白及DON单克隆抗体进行偶联,一步法制备磁荧光抗体探针,以DON人工抗原(DON-BSA)为检测线建立磁荧光免疫层析试纸。同时用胶体金标记DON单克隆抗体,以DON-BSA为检测线建立胶体金免疫层析试纸;制备的磁荧光抗体探针具有很好的磁性、荧光特性及抗体反应性,基于该探针成功制备了DON磁荧光免疫层析试纸,该试纸回归方程为y=-0.562x+0.921,R2=0.990,IC50为5.611 ng/mL,检出限为1.089 ng/mL;制备了DON胶体金免疫层析试纸,该试纸裸眼检测灵敏度为500 ng/mL;定量检测回归方程为y=-0.543x+1.485,R2=0.991,IC50为65.16 ng/mL,检出限为11.94 ng/mL。DON磁荧光免疫层析试纸的灵敏度是胶体金免疫层析试纸的10.96 倍。本实验建立的磁荧光免疫层析试纸模式可以同时实现样品的富集及荧光信号检测,提高检测灵敏度,并成功用于DON的检测,为磁荧光纳米颗粒广泛应用于免疫层析领域提供参考。 相似文献

A predictive dynamic model for Clostridium perfringens spore germination and outgrowth in cooked pork products during cooling is presented. Cooked, ground pork was inoculated with C. perfringens spores and vacuum packaged. For the isothermal experiments, all samples were incubated in a water bath stabilized at selected temperatures between 10 and 51 °C and sampled periodically. For dynamic experiments, the samples were cooled from 54.4 to 27 °C and subsequently from 27 to 4 °C for different time periods, designated as x and y hours, respectively. The growth models used were based on a model developed by Baranyi and Roberts (1994), which incorporates a constant, referred to as the physiological state constant, q0. The value of this constant captures the cells' history before the cooling begins. To estimate specific growth rates, data from isothermal experiments were used, from which a secondary model was developed, based on a particular form of Ratkowsky's 4-parameter equation. Using the data from dynamic experiments and the Ratkowsky model, an optimal value of q0 (=0.01375) was derived minimizing the mean square error of predictions. However, using this estimate, the model had a tendency to over-predict relative growth when there was observed small amounts of relative growth, and under-predict relative growth when there was observed large relative growth. To provide more fail-safe estimates, rather than using the derived value of q0, a value of 0.04 is recommended. The predictive model with this value of q0 would provide more fail-safe estimates of relative growth and could aid producers and regulatory agencies with determining disposition of products that were subjected to cooling deviations.Industrial relevanceSafe time/temperature for cooling of cooked pork is very important to guard against the pathogen in cooked products. High molecular weight, hyperbranched polyarylenes were synthesized in high isolation yields by the copolycyclotrimerizations of 2,5-diethynylthiophene (1), 4,4′-biphenyldiyne (2), and 2,7-diethynylfluorenes (3) with 1-heptyne (4) and 1-dodecyne (5) using TaCl5–Ph4Sn as the catalyst in toluene. The structures of the polymers were characterized by IR, NMR, TGA, and UV analyses. All the polymers exhibited outstanding thermal stability and emitted strong blue light, whose intensities are higher than that of poly(1-phenyl-1-octyne), a well-known highly emissive polyacetylene. Little red shift was observed in the photoluminescence of the polymer thin films. The polymers strongly attenuated intense pulses of 532 nm laser pulses. 相似文献

Fatty liver (i.e., hepatic lipidosis) is a prevalent metabolic disorder in dairy cows during the transition period, characterized by excess hepatic accumulation of triglyceride (TG), tissue dysfunction, and cell death. Detailed pathological changes, particularly hepatic fibrosis, during fatty liver remain to be determined. Liver fibrosis occurs as a consequence of liver damage, resulting from the excessive accumulation of extracellular matrix, which distorts the architecture of the normal liver, compromising its normal synthetic and metabolic functions. Thus, we aimed to investigate liver fibrosis status and its potential causal factors including oxidative stress, hepatocyte apoptosis, and production of inflammatory cytokines in the liver of cows with fatty liver. Forty-five dairy cows (parity, 3–5) were selected, and liver biopsy and blood were collected on the second week postpartum (days in milk, 10–14 d). On the basis of the degree of lipid accumulation in liver, selected cows were categorized into normal (n = 25; TG <1% wet wt), mild fatty liver (n = 15; 1% ≤ TG <5% wet wt), and moderate fatty liver (n = 5; 5% ≤ TG <10% wet wt). Compared with normal cows, blood concentrations of nonesterified fatty acids and β-hydroxybutyrate, along with alanine aminotransferase and aspartate aminotransferase activities, were greater in the cows with fatty liver (mild and moderate). Hepatic extracellular matrix deposition, as indicated by Picrosirius red staining, was greater in cows with fatty liver than those with normal ones. In addition, we observed an increased proportion of collagen type I fiber in extracellular matrix with increased lipid accumulation in the liver. Compared with normal cows, the area of α-smooth muscle actin (α-SMA)-positive staining along with the mRNA abundance of collagen type I α 1 (COL1A1), ACTA2 (gene encoding α-SMA), and transforming growth factor-β (TGFB) were greater in cows with fatty liver. Compared with normal cows, hepatic contents of malondialdehyde, glutathione disulfide, and 8-isoprostane were greater, whereas total antioxidant capacity, the hepatic content of glutathione, and activities of antioxidant indicators, including superoxide dismutase, glutathione peroxidase, and catalase, were lower in cows with fatty liver. The number of terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling-positive cells and abundance of apoptosis-related molecules BAX, CASP3, CASP8, and CASP9 were greater in cows with fatty liver. However, mRNA abundance of the anti-apoptotic gene BCL2 did not differ. The mRNA abundance of pro-inflammatory cytokines including tumor necrosis factor-α (TNFA), interleukin-1β (IL1B), and interleukin-6 (IL6) was greater in the liver of cows with fatty liver. Overall, the present study indicated that fibrosis is a common pathological response to liver damage and is associated with oxidative stress, hepatocyte death, and inflammation. 相似文献

The combination of catch crop cultivation with its use for biogas production would increase renewable energy production in the form of methane, without interfering with the production of food and fodder crops. The low biomass yield of catch crops has been shown as the main limiting factor for using these crops as co-substrate in biogas plants, since the profit obtained from the sale of methane barely compensates the harvest costs. Therefore, a new agricultural strategy to harvest catch crops together with the residual straw of the main crop was investigated, in order to increase the biomass and the methane yield per hectare. Seven catch crops harvested together with stubble from the previous main crop were evaluated. The effects of stubble height, harvest time and ensiling as a storage method for the different catch crops/straw blends were studied. Biomass yields as TS ranged between 3.2 and 3.6 t ha−1 y−1of which the catch crop constituted around 10% of the total biomass yield. Leaving the straw on the field until harvest of the catch crop in the autumn could benefit methane production from the straw both due to increased biomass yield and an increased organic matter bioavailability of the straw taking place on the field during the autumn months. Ensiling as a storage method could be feasible in terms of energy storage and guaranteeing the feedstock availability for the whole year. This new agricultural strategy may be a good alternative for economically feasible supply of catch crops and straw for biogas production. 相似文献
