The Beaver Creek confluence with the main‐stem Klamath River was studied to assess salmonid use in a thermal mixing zone under various summer hydrological and meteorological conditions. Main‐stem flow releases from Iron Gate Dam ranged from 17 cms (615 cfs) to 37 cms (1320 cfs) during the study period and main‐stem water temperatures ranged from 19.5 to 26°C. A grid was constructed to define the thermal refuge as a system of cells. Temperatures were monitored using remote temperature loggers and fish counts were conducted using daytime snorkelling. Most juvenile salmonids were observed moving into the refuge when main‐stem temperatures exceeded 22–23°C. Salmonids in the thermal refuge did not necessarily seek the coolest water, but were generally located in habitats commensurate with species‐specific behavioural needs within their thermal tolerance range. Such ranges largely occurred within refuge areas. Variable meteorological conditions confounded observable biological thermal benefit to fish resulting from higher or lower main‐stem flows. Thermal regime dynamics indicated that under the hydrological and meteorological conditions observed, higher flows from Iron Gate Dam showed some ability to change the structure of the refuge area. It appeared that without the thermal refuge, main‐stem flows alone could not sustain the salmonid population because high water temperatures usually exceeded their published thermal tolerance limits. In this study, the effect of essential oils (thyme and clove) and packaging treatments (vacuum and modified atmosphere) on the storage of hot smoked rainbow trout (Oncorhynchus mykiss) fillets over a period of 120 days at 4°C was investigated. Fillets were subjected to microbiological (total aerobic mesophilic bacteria, psychrotrophic bacteria, lactic acid bacteria, yeast and mould), chemical analyses (pH, thiobarbituric acid reactive substances and total volatile base nitrogen) and quality testing throughout the storage period. Difference in microbiological and chemical changes between samples was found to be significant (p < 0.05) during storage period. Sensory evaluation showed significant differences between the samples, and V2 and M2 samples were assessed as the most acceptable products by the panellists. In conclusion, essential oils treatment to hot smoking had a positive effect on shelf life of rainbow trout, especially clove oil showed more positive effect when compared with thyme oil. As rivers warm, cold‐water fish species may alleviate thermal stress by moving into localized thermal refuges such as cold‐water plumes created by cool tributary inflows. We quantified use of two tributary confluence plumes by juvenile steelhead, Oncorhynchus mykiss, throughout the summer, including how trout positioned themselves in relation to temperature within confluence plumes. At two confluences, Cedar and Elder creeks, along the South Fork Eel River, California, USA, we monitored temperatures using in situ logger grids throughout summer 2016. Fish were counted within confluences via snorkel surveys five times a day on 5 days at each site. We found diel and seasonal dependence on confluence use by steelhead, especially at the Cedar Creek confluence, where mainstem temperatures exceeded 28°C. At this site, fish moved into the confluence on the warmest days and warmest times of the day. Fish observed within the Cedar Creek confluence plume were most common in locations between 20–22°C, rather than the coldest locations (14.5°C). At Elder Creek, where mainstem temperatures remained below 24°C, there was little relationship between mainstem temperature and steelhead presence in the confluence plume. At both sites, steelhead distribution within plumes was influenced by spatial variation of temperature and mean temperature in surveyed grid cells. Our results show that cool tributaries flowing into warmer mainstem reaches (over 24°C) likely create important thermal refuges for juvenile steelhead. As mainstem rivers warm with climate change, cool‐water tributary inputs may become more important for sustaining cold‐water salmonids near the southern end of their range.  相似文献

We developed tetranucleotide-repeat microsatellite markers for the masu salmon (Oncorhynchus masou) complex. 454 pyrosequencing was used to discover repeat motifs, and seven polymorphic microsatellite-primer sets were identified. The number of alleles detected at each locus ranged from four to 24 and the expected heterozygosity varied from 0.57 to 0.92. Cross-subspecies amplification for O. m. masou, O. m. ishikawae and O. m. subsp. was successful. These microsatellites can be utilized in studies of genetic structure, genetic diversity, and intra- and inter-subspecific hybridization, making a contribution to conservation and management of the Oncorhynchus masou complex.  相似文献

This research was focused on the production of polyunsaturated fatty acid concentrates from a farmed rainbow trout (Oncorhynchus mykiss) by‐product (i.e. belly muscle). The effect of different process variables (urea/fatty acids (FA) contents ratio, crystallisation time and temperature and stirring speed of the urea/FA mixture) on the lipid oxidation development during the urea complexation process was investigated. For this purpose, an experimental design (26 runs) following the response‐surface methodology was developed. As a result, peroxide value and TOTOX index showed to be dependent on the crystallisation time and temperature and the urea/FA ratio, while no influence of the crystallisation stirring speed was detected on both indices; additionally, polyene index was affected by the urea/FA ratio and its interaction with the crystallisation time. An optimised desirability score near 1.0 was attained provided values of 2.8 °C, 3.05 h and 3.57 were applied for crystallisation temperature, crystallisation time and urea/FA ratio, respectively.  相似文献

ABSTRACT Laboratory-reared king salmon were harvested using CO2 anesthetizing (CO2) or by rested harvesting techniques using AQUI-S(tm) anesthetizing (AQUI-S). Fish were killed by spiking, and flesh portions were stored in air packs (0°C) for 22 d. Headspace oxygen levels, sensory characteristics (raw and cooked), tensile properties, drip loss, gaping, pH, nucleotide derivatives, color, and bacterial counts were determined after 5 to 22 d of storage. All parameters except tensile properties showed significant trends with storage time. Oxygen levels were consistently lower in CO2 packs whereas, on average, drip was lower in AQUI-S samples, and L*-values and bacterial numbers were higher in AQUI-S samples. Overall, harvesting effects had minimal impact on the quality parameters measured.  相似文献

目的调查广东省市售三文鱼水产品掺假情况。方法应用实时荧光PCR技术检测大西洋鲑鱼和虹鳟鱼的方法,对广东省内批发市场、超市、餐饮寿司店和网络平台等场所的三文鱼掺假情况进行调查。结果98批次样品中,有6批次未检出大西洋鲑鱼,未检出率为6.12%。通过PCR测序结果鉴定,这6批次未检出大西洋鲑鱼的样品中,4批次为虹鳟鱼,1批次为王鲑,1批次为银鲑。结论三文鱼水产品掺假情况较少,但掺假风险仍然存在,建议监管部门加强监管。  相似文献

为了实现大西洋三文鱼和虹鳟鱼的物种掺假快速检测,本研究建立了一种基于双重PCR(普通和荧光)的物种鉴定方法。通过序列分析,分别基于线粒体COⅠ和Cyt b基因设计大西洋三文鱼和虹鳟鱼特异性引物,验证引物的特异性并对双重PCR反应体系进行优化,建立大西洋三文鱼和虹鳟鱼的双重PCR快速鉴定方法。利用本研究设计的引物,仅大西洋三文鱼和虹鳟鱼可以分别扩增出108 bp和207 bp的特异性条带,而其他23种非目标物种均未有扩增条带。经验证,在双重PCR反应体系中,大西洋三文鱼和虹鳟鱼引物的最佳添加量分别为0.1和0.4 μmol/L,熔解温度分别约为81.5 ℃和85 ℃。利用该方法从29份市售“三文鱼”样品中检测到6份大西洋三文鱼和3份虹鳟鱼,且与DNA条形码比对结果一致。本研究建立的大西洋三文鱼和虹鳟鱼双重PCR(普通和荧光)鉴定方法具有良好的特异性和适用性,为大西洋三文鱼和虹鳟鱼物种鉴定提供了可靠的技术手段。  相似文献

The fertilization of freshwater fish occurs in an environment that may negatively affect the gametes; therefore, the specific mechanisms triggering the encounters of gametes would be highly expedient. The egg and ovarian fluid are likely the major sources of these triggers, which we confirmed here for rainbow trout (Oncorhynchus mykiss). The ovarian fluid affected significantly spermatozoa performance: it supported high velocity for a longer period and changed the motility pattern from tumbling in water to straightforward moving in the ovarian fluid. Rainbow trout ovarian fluid induced a trapping chemotaxis-like effect on activated male gametes, and this effect depended on the properties of the activating medium. The interaction of the spermatozoa with the attracting agents was accompanied by the “turn-and-run” behavior involving asymmetric flagellar beating and Ca2+ concentration bursts in the bent flagellum segment, which are characteristic of the chemotactic response. Ovarian fluid created the optimal environment for rainbow trout spermatozoa performance, and the individual peculiarities of the egg (ovarian fluid)–sperm interaction reflect the specific features of the spawning process in this species.  相似文献
