Direct Injection of CRISPR/Cas9-Related mRNA into Cytoplasm of Parthenogenetically Activated Porcine Oocytes Causes Frequent Mosaicism for Indel Mutations

Some reports demonstrated successful genome editing in pigs by one-step zygote microinjection of mRNA of CRISPR/Cas9-related components. Given the relatively long gestation periods and the high cost of housing, the establishment of a single blastocyst-based assay for rapid optimization of the above system is required. As a proof-of-concept, we attempted to disrupt a gene (GGTA1) encoding the α-1,3-galactosyltransferase that synthesizes the α-Gal epitope using parthenogenetically activated porcine oocytes. The lack of α-Gal epitope expression can be monitored by staining with fluorescently labeled isolectin BS-I-B₄ (IB4), which binds specifically to the α-Gal epitope. When oocytes were injected with guide RNA specific to GGTA1 together with enhanced green fluorescent protein (EGFP) and human Cas9 mRNAs, 65% (24/37) of the developing blastocysts exhibited green fluorescence, although almost all (96%, 23/24) showed a mosaic fluorescent pattern. Staining with IB4 revealed that the green fluorescent area often had a reduced binding activity to IB4. Of the 16 samples tested, six (five fluorescent and one non-fluorescent blastocysts) had indel mutations, suggesting a correlation between EGFP expression and mutation induction. Furthermore, it is suggested that zygote microinjection of mRNAs might lead to the production of piglets with cells harboring various mutation types.     

黑龙江省节水增粮行动(2012—2015年)实施方案

文章通过对黑龙江省高效节水现状,存在问题的分析,结合黑龙江省高效节水发展的支撑能力,阐述黑龙江省节水增粮目标、任务及总体布局,为黑龙江省2012—2015年节水增粮实施提供依据。     

高密度聚乙烯/丁苯橡胶热塑性硫化胶的形状记忆性能

采用动态硫化法制备了高密度聚乙烯(HDPE)/丁苯橡胶(SBR)热塑性硫化胶(TPV),研究了橡塑比、定型温度、回复温度及增容剂苯乙烯-丁二烯-苯乙烯嵌段共聚物(SBS)对HDPE/SBR TPV形状记忆性能的影响,观察了其微观结构,提出了形状记忆行为机制。结果表明,HDPE/SBR TPV具有典型的"海-岛"结构,拉伸后其表面存在明显的取向结构;SBS能显著提高HDPE/SBR TPV的形状记忆性能,当SBS用量为6份、定型温度和回复温度均为120℃时,HDPE/SBR TPV的形状记忆性能最佳。     

陈四楼煤矿“123456双重预防体系”的建设与应用

针对煤矿“双重预防体系”如何落地生根问题，结合陈四楼煤矿在风险分级管控和隐患排查治理方面的实用方法和“双重预防体系”的建设经验，深入研究了“双重预防体系”的相关标准、风险隐患事故之间的关系及其各自的产生和发展机理、安全风险的有关辨识评估方法、事故隐患的排查和治理方法，分析了传统安全管理模式与“双重预防体系”的新型安全管理模式的差别，总结出了“123456双重预防体系”这个囊括了事前安全风险辨识、事中隐患排查治理、事后安全现状评估的创新成果，实现了“事前、事中、事后”的全过程控制，对夯实煤矿安全管理根基、促进矿井平稳有序发展发挥着越来越重要的作用，为煤炭行业“双重预防体系”的落地生根提供了借鉴和参考。     

Chemically modified organic/inorganic nanoporous composite particles for the adsorption of reactive black 5 from aqueous solution

In the present work, we report a chemically modified polyacrylamide/silica nanoporous composite adsorbent for the removal of reactive black 5 (RB5) azo dye from aqueous solutions. The composite adsorbent was synthesized in a packed bed and modified by ethylenediamine (EDA). The adsorbent was characterized by Fourier transformation infrared (FT-IR), thermogravimetric analysis (TGA), thermoporometry, Brunauer, Emmett and Teller (BET) method and scanning electron microscopy (SEM). Mechanical stability of the adsorbent was examined in a packed bed by following the back-pressure of the column. Pore diameter of the composite adsorbent in dry and wet states was estimated to be about 18.71 nm and 12.61 nm, respectively. Adsorption experiments were performed in batch mode and effect of various operational parameters on the adsorption capability of the adsorbent was studied systematically. The maximum adsorption capacity of the modified composites was found to be 454.5 mg RB5/g of adsorbent. The equilibrium data were analyzed by Langmuir, Freundlich, Sips, BET and Redlich–Peterson isotherm models and found to fit well to the BET isotherm. The data kinetically followed the pseudo-second-order model. High adsorption capacity, fast removal mechanism, and good mechanical stability are three advantages of the presented composite for the removal of RB5.     

APP软件性能效率研究

性能效率是APP软件的重要质量属性，但目前缺乏APP软件性能效率的通用模型。分析了APP软件的性能特征，基于ISO/IEC 25010标准提出了APP软件的性能效率模型，定义了APP软件性能效率的子特性和度量指标。基于提出的APP软件性能效率模型，通过实验对APP软件的性能效率进行了度量及相关分析。     

腔磁力系统中的诱导透明和快慢光传播

为了在腔磁力系统中实现可控的磁子诱导透明、磁力诱导透明以及快慢光传播,建立了一个混合腔磁力系统.该系统由一个含有YIG球的微波腔和在z方向对球施加一个均匀的偏置磁场组成,并用强泵浦场驱动磁子和弱探测场驱动微波腔.研究表明,通过调节腔与磁子之间的相互作用强度和微波腔与磁子的耗散比,可以增加磁子诱导透明(MIT)、磁力诱导透明(MMIT)的效果和提高快慢光传播的速度.该研究结果可为磁力诱导放大、量子光学操纵和量子信息存储以及灵敏光开关的研究提供参考.     

Red-shift and improved afterglow of Sr3Al2-xBxO5Cl2:Eu2+, Dy3+ phosphors via a cation substitution strategy

Sr₃Al_2-xB_xO₅Cl₂:Eu²⁺, Dy³⁺ (x = 0, 0.2, 0.4) long persistent phosphors were prepared via solid-state process. The pristine Sr₃Al₂O₅Cl₂:Eu²⁺, Dy³⁺ phosphor exhibits orange/red broad band emission around 609 nm, which can be attributed to the electric radiation transitions 4f⁶5 d¹→4f⁷ of Eu²⁺. Upon the same excitation, the B³⁺-doped Sr₃Al_2-xB_xO₅Cl₂:Eu²⁺, Dy³⁺ phosphors display red-shift from 609 nm to 625 nm with increasing B³⁺ concentrations. The XRD patterns show that Al³⁺ can be replaced by B³⁺ in the host lattice at the tetrahedral site, which causes lattice contraction and crystal field enhancement, and thereafter achieves the red-shift on the emission spectrum. The XPS investigation provides direct evidence of the dominant 2-valent europium in the phosphor, which can be ascribed for the broad band emission of the prepared phosphors. The afterglow of all phosphors show standard double exponential decay behavior, and the afterglow of Sr₃Al₂O₅Cl₂:Eu²⁺, Dy³⁺is rather weak, while the sample co-doped with B³⁺shows longer and stronger afterglow, as confirmed after the curve simulation. The analysis of thermally stimulated luminescence showed that, when B³⁺ is introduced, a much deeper trap is created, and the density of the electron trap is also significantly increased. As a result, B³⁺ ions caused redshift and enhanced afterglow for the Sr₃Al_2-xB_xO₅Cl₂:Eu²⁺, Dy³⁺ phosphor.     

多运营商高铁隧道移动网络覆盖方案

高铁移动网络覆盖是国内三大通信运营商的一个重点,而高铁隧道内移动网络覆盖更是运营商的一大难点痛点。文章根据我国中部省份某高铁线路覆盖规划实例,采用“设备+POI+泄漏电缆”模式,即三家运营商信号源设备通过同一POI(point of interface,多系统接入平台)接入,信号输出到泄漏电缆进行隧道覆盖,隧道口场坪站安装宽频切换天线对隧道外进行延伸覆盖,通过链路预算合理布置各运营商主设备信号源,从而实现隧道到室外的无缝覆盖。最后,根据已有成熟网络覆盖解决方案,对未来5G高铁隧道移动网络覆盖方案进行了探讨。
