SmartBone® (SB) is a biohybrid bone substitute advantageously proposed as a class III medical device for bone regeneration in reconstructive surgeries (oral, maxillofacial, orthopedic, and oncology). In the present study, a new strategy to improve SB osteoinductivity was developed. SB scaffolds were loaded with lyosecretome, a freeze-dried formulation of mesenchymal stem cell (MSC)-secretome, containing proteins and extracellular vesicles (EVs). Lyosecretome-loaded SB scaffolds (SBlyo) were prepared using an absorption method. A burst release of proteins and EVs (38% and 50% after 30 min, respectively) was observed, and then proteins were released more slowly with respect to EVs, most likely because they more strongly adsorbed onto the SB surface. In vitro tests were conducted using adipose tissue-derived stromal vascular fraction (SVF) plated on SB or SBlyo. After 14 days, significant cell proliferation improvement was observed on SBlyo with respect to SB, where cells filled the cavities between the native trabeculae. On SB, on the other hand, the process was still present, but tissue formation was less organized at 60 days. On both scaffolds, cells differentiated into osteoblasts and were able to mineralize after 60 days. Nonetheless, SBlyo showed a higher expression of osteoblast markers and a higher quantity of newly formed trabeculae than SB alone. The quantification analysis of the newly formed mineralized tissue and the immunohistochemical studies demonstrated that SBlyo induces bone formation more effectively. This osteoinductive effect is likely due to the osteogenic factors present in the lyosecretome, such as fibronectin, alpha-2-macroglobulin, apolipoprotein A, and TGF-β.  相似文献

The purposes of this work were to obtain the accurate elastic modulus of the nanocrystalline WC–Co cemented carbides, and to propose the mechanism for the difference of elastic modulus between the nanocrystalline and conventional polycrystalline cemented carbides. The nanocrystalline cemented carbide was prepared by spark plasma sintering (SPS) technique. The conventional polycrystalline cemented carbides were prepared by SPS and sinter-HIP techniques as references, respectively. The sintered cemented carbides were characterized by X-ray diffractometry, scanning electron microscopy and the transmission electron microscopy with precession electron diffraction technology. The elastic modulus was obtained by averaging the values measured with the continuous stiffness measurement method of the nanoindentation technology. The results show that the nanocrystalline cemented carbide has a relatively low modulus, which could be attributed to the more interface area and higher fraction ratio of the hcp cobalt phase caused by the rapid heating and cooling process during SPS.  相似文献

Inherited cardiomyopathies are frequent causes of sudden cardiac death (SCD), especially in young patients. Despite at the autopsy they usually have distinctive microscopic and/or macroscopic diagnostic features, their phenotypes may be mild or ambiguous, possibly leading to misdiagnoses or missed diagnoses. In this review, the main differential diagnoses of hypertrophic cardiomyopathy (e.g., athlete’s heart, idiopathic left ventricular hypertrophy), arrhythmogenic cardiomyopathy (e.g., adipositas cordis, myocarditis) and dilated cardiomyopathy (e.g., acquired forms of dilated cardiomyopathy, left ventricular noncompaction) are discussed. Moreover, the diagnostic issues in SCD victims affected by phenotype-negative hypertrophic cardiomyopathy and the relationship between myocardial bridging and hypertrophic cardiomyopathy are analyzed. Finally, the applications/limits of virtopsy and post-mortem genetic testing in this field are discussed, with particular attention to the issues related to the assessment of the significance of the genetic variants.  相似文献

Granular materials segregate spontaneously due to differences in particle size, shape, density and flow behaviour. In this paper we experimentally investigate density-difference-driven segregation for a range of density ratios and a range of heavy particle concentrations. The experiments are conducted in an annular shear cell with rotating bumpy bottom that yields an exponential shear profile. The cell is initially filled with a layer of light particles and an upper layer of heavier grains and, on top, a load provides confinement. The segregation process is filmed through the transparent side-wall with a camera, and the evolution of particle concentration in space and time is evaluated by means of post-processing image analysis. We also propose a continuum-approach to model density-driven segregation. We use a segregation-diffusion transport equation, constitutive relations for effective viscosity and friction coefficient, and a segregation velocity analogous to the Stokes’ law. The model, which is validated by comparison with experimental findings, can successfully predict density-driven segregation at different density ratios and volumetric fraction.  相似文献

Cold Sintering Process (CSP) was applied on commercial nanopowders to produce nanostructured TiO2 anatase with nano-to-macro porosity. Nanoporous TiO2 based materials were obtained by applying CSP at 150 °C and pressures up to 500 MPa on three TiO2 nanopowders with different specific surface area (s.s.a. = 50, 90 and 370 m2/g), using water as transient aqueous environment. Although TiO2 is insoluble in water, a density of 68% and s.s.a. = 117 m2/g were achieved from the powder with the highest specific surface area. A post annealing process at 500 °C increased the density up to 73% with a s.s.a. = 59 m2/g, and the crystallites dimensions passed from 110 Å in the powder to 130 Å in CSP material and 172 Å after post annealing. Finally, macroporosity was produced by using thermoplastic polymer beads as sacrificial templates within TiO2 nanopowder during CSP, followed by a debonding at 500 °C.  相似文献

We report the heterologous expression, structure, and antimicrobial activity of a lasso peptide, ubonodin, encoded in the genome of Burkholderia ubonensis. The topology of ubonodin is unprecedented amongst lasso peptides, with 18 of its 28 amino acids found in the mechanically bonded loop segment. Ubonodin inhibits RNA polymerase in vitro and has potent antimicrobial activity against several pathogenic members of the Burkholderia genus, most notably B. cepacia and B. multivorans, causative agents of lung infections in cystic fibrosis patients.  相似文献

Hydrothermal carbonization of sewage sludge was carried out with the aim to evaluate the influence of process severity and initial solid content. Response surface methodology was applied to model yield and C yield responses. Enhanced dewaterability performance was recorded under mild processing conditions. The treatment promoted concentration and immobilization of Pb, Cd, Ni, Zn, and Cu. Variation of the solid content showed a stronger influence than severity on average yield and C yield. Higher heating values (HHVs) and energy retention efficiencies (EREs) of hydrochars obtained at the lowest solid content displayed the lowest values. Hence, the energy requirements of a first dewatering step should be compared with the related improvement in terms of HHV and ERE when sludge is used as feedstock.  相似文献
