Cyclophilins have important homeostatic roles, but following tissue injury, cyclophilin A (CypA) can promote leukocyte recruitment and inflammation, while CypD can facilitate mitochondrial-dependent cell death. This study investigated the therapeutic potential of a selective cyclophilin inhibitor (GS-642362), which does not block calcineurin function, in mouse models of tubular cell necrosis and renal fibrosis. Mice underwent bilateral renal ischemia/reperfusion injury (IRI) and were killed 24 h later: treatment with 10 or 30 mg/kg/BID GS-642362 (or vehicle) began 1 h before surgery. In the second model, mice underwent unilateral ureteric obstruction (UUO) surgery and were killed 7 days later; treatment with 10 or 30 mg/kg/BID GS-642362 (or vehicle) began 1 h before surgery. GS-642362 treatment gave a profound and dose-dependent protection from acute renal failure in the IRI model. This protection was associated with reduced tubular cell death, including a dramatic reduction in neutrophil infiltration. In the UUO model, GS-642362 treatment significantly reduced tubular cell death, macrophage infiltration, and renal fibrosis. This protective effect was independent of the upregulation of IL-2 and activation of the stress-activated protein kinases (p38 and JNK). In conclusion, GS-642362 was effective in suppressing both acute kidney injury and renal fibrosis. These findings support further investigation of cyclophilin blockade in other types of acute and chronic kidney disease.  相似文献

This paper examines what influences trust in mobile social commerce environment. Drawing on trust-based acceptance model (i.e. cognitive and emotional trust) and online review features (i.e. profile photo, linguistic style, and reported experience), we examine how these factors affect trust in mobile social commerce. Hypotheses were tested using survey data. The results of our model showed that there are significant influences of profile photo, reported experience, cognitive, and emotional trust towards trust in ms-commerce. This work contributes to existing literature by examining the roles of previous trust in mobile payments and online reviews on trust in mobile social commerce.  相似文献

Introducing a carbon single coating is a popular method used to protect SiCf/Ti composites from severe interface reactions. However, carbon coatings lose their protective effect on SiC fibres at high temperature, even after a short period time. As such, given the strong demand for high temperature applications in aeronautics and astronautics a more coating which is more effective at high temperatures is desirable. In order to improve the high temperature interfacial stability of SiCf/Ti composites, a C/TiCx duplex coating system with different C contents in TiCx was introduced to explore the protection of fibres at 1200?°C for 1?h. The results show that the C/quasi-stoichiometric TiC coating system protects the SiC fibres most effectively. Based on insights from the evolution of the interface structure, TiCx has been identified as an interfacial reaction product from the C single coating, exhibiting a gradient in C content and grain size, which is different from a deposited TiC layer with a well-distributed composition and structure. The different coating structure gives rise to different ability to resist C diffusion at high temperatures, in which poor resistance ability appears in TiCx interfacial reaction layer coming from C single coating due to short-circuit diffusion in C-rich fine-grained TiC layer and fast intracrystalline diffusion trigged by amounts of vacancies in sub-stoichiometric coarse-grained TiC layer. Therefore, C/quasi-stoichiometric TiC duplex coatings with a thick, coarse-grained quasi-stoichiometric TiC layer could effectively inhibit C diffusion by comparison to C single coatings, and is more effective than C/rich-carbon TiC duplex coatings due to the existence of short-circuit diffusion in the latter. As such, C/quasi-stoichiometric TiC duplex coatings appear to be an optimal diffusion barrier for SiCf/Ti composites at high temperature.  相似文献

Downstream processing or product recovery plays a vital role in the development of bioprocesses. To improve the bioprocess efficiency, some unconventional methods are much required. The continuous manufacturing in downstream processing makes the Process Analytical Technologies (PATs) as an important tool. Monitoring and controlling bioprocess are an essential factor for the principles of PAT and quality by design. Spectroscopic methods can apply to monitor multiple analytes in real-time with less sample processing with significant advancements. Raman spectroscopy is an extensively used technique as an analytical and research tool owing to its modest process form, non-destructive, non-invasive optical molecular spectroscopic imaging with computer-based analysis. Generally, its application is essential for the analysis and characterization of biological samples, and it is easy to operate with minimal sample. The innovation on various types of enhanced Raman spectroscopy was designed to enhance the Raman analytical technique. Raman spectroscopy could couple with chemometrics to provide reliable alternative analysis method of downstream process analysis. Thus, this review aims to provide useful insight on the application of Raman spectroscopy for PAT in downstream processing of biotechnology and Raman data analysis in biological fields.  相似文献

Superabsorbent polymers (SAPs) were synthesized by grafting acrylic acid and butyl acrylate onto carboxymethyl cellulose (CMC) modified from Ceiba pentandra (L.) Gaertn. (kapok) cotton, with N,N′‐methylenebisacrylamide as a crosslinker and ammonium persulphate as initiator. The effect of distilled water, saline solution, and applied pressure on superabsorbent was investigated. The product exhibited the maximum water absorbency of 554 g/g in distilled water and 96 g/g in saline solution. The SAP achieved the highest water absorbency under load of 83 g/g under applied pressure of 7.6 g/cm2. The kapok cotton modified cellulose‐based SAP exhibited stronger gel strength than the SAP based on commercial CMC. This is probably due to the higher grafting efficiency (78.3%) of the former. The SAP was characterized by FTIR analysis, thermogravimetric analysis, and scanning electron microscopy. Thermogravimetric analysis results showed that the SAP, with AA and BA grafted onto CMC, had better thermal stability than CMC alone. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. A structure–activity relationship (SAR) study of the triosmium carbonyl cluster Os3(CO)10(NCCH3)2 was carried out with a series of clusters of the general formula Os3(CO)12?nLn, cationic osmium clusters and a hemi‐labile maltolato‐Os cluster. The SAR results showed that good solubility in DMSO and at least one vacant site are required for cytotoxicity. In vitro evaluation of these new compounds showed that some are selectively active against estrogen receptor (ER)‐independent MDA‐MB‐231 breast cancer cell lines relative to ER‐dependent MCF‐7 breast cancer cells, suggesting that the compounds have a different biological target specific to MDA‐MB‐231 cells. In particular, the maltolato cluster exhibits strong antiproliferative activity, with an IC50 value of 3 μM after only 24 h incubation. Additionally, biochemical assays conducted with the cationic cluster show that it induces apoptosis, although a biological target has not yet been identified. Further research to establish the molecular targets of these compounds and to develop improved organometallic clusters as potential breast cancer therapeutics is underway.  相似文献

This paper presents fast alternating direction implicit (FADI) method for efficient transient thermal simulation of integrated circuits. The FADI method is formulated from Peaceman–Rachford's ADI and Douglas–Gunn's ADI methods. The update procedure of the proposed method has basic implicit form that features derivative‐free right‐hand side and hence, better efficiency and conciseness. Subsequently, through the basic implicit form of FADI method, the relationship between classical Peaceman–Rachford's and Douglas–Gunn's ADI methods can be clarified and elucidated in detail. A unified boundary condition that can cater to common kinds of boundary conditions in thermal simulation is also introduced. To further accelerate FADI method, the graphics processing unit is also utilized through Compute Unified Device Architecture implementation. It is shown that high efficiency gain can be achieved using the proposed FADI method through large time step size and data parallelism, while maintaining stability and good accuracy. As numerical illustration, an integrated circuit structure with microchannel cooling is demonstrated. Numerical results further ascertain the cooling effect of the microchannels.
