We evaluated the abscopal effect of re-implantation of liquid nitrogen-treated tumor-bearing bone grafts and the synergistic effect of anti-PD-1 (programmed death-1) therapy using a bone metastasis model, created by injecting MMT-060562 cells into the bilateral tibiae of 6–8-week-old female C3H mice. After 2 weeks, the lateral tumors were treated by excision, cryotreatment using liquid nitrogen, excision with anti-PD-1 treatment, and cryotreatment with anti-PD-1 treatment. Anti-mouse PD-1 4H2 was injected on days 1, 6, 12, and 18 post-treatment. The mice were euthanized after 3 weeks; the abscopal effect was evaluated by focusing on growth inhibition of the abscopal tumor. The re-implantation of frozen autografts significantly inhibited the growth of the remaining abscopal tumors. However, a more potent abscopal effect was observed in the anti-PD-1 antibody group. The number of CD8+ T cells infiltrating the abscopal tumor and tumor-specific interferon-γ (IFN-γ)-producing spleen cells increased in the liquid nitrogen-treated group compared with those in the excision group, with no significant difference. The number was significantly higher in the anti-PD-1 antibody-treated group than in the non-treated group. Overall, re-implantation of tumor-bearing frozen autograft has an abscopal effect on abscopal tumor growth, although re-implantation of liquid nitrogen-treated bone grafts did not induce a strong T-cell response or tumor-suppressive effect.  相似文献

In-beam Mössbauer spectroscopy utilizing unstable 57Mn beams is a powerful method to extract physical and chemical properties at the atomic scale. A parallel plate avalanche counter (PPAC), optimized to detect conversion electrons generated by the Mössbauer effect, can be employed to suppress higher-energy background γ rays. However, β rays are emitted by the 57Mn parent nuclei of 57Fe, which can significantly degrade the spectrum quality. In the present work, we have developed a new anti-coincidence-detection system with a thin plastic scintillation counter (0.5 mmt), which can be used to detect β rays and reject them from the recorded PPAC events. To demonstrate the anti-coincidence system, we carried out Mössbauer spectroscopy utilizing 57Mn nuclei that were implanted into a non-magnetic aluminum metal plate at room temperature. Using the anti-coincidence method, we obtained a typical Mössbauer spectrum of high quality, despite a very low number of implanted 57Mn atoms, of ∼5 × 109. The signal to noise ratio of the obtained spectrum was increased remarkably, and the relative peak height above the baseline increased from 10% to 220% using the anti-coincidence method. The developed detection system is applicable to investigation of in situ properties, and avoids the potentially problematic agglomeration of probes in a sample.  相似文献

A detailed morphological study of the microstructure of poly(p-phenylene benzobisoxazole) (PBO) fibres (HM and HM+) and a polypyridobisimidazole (PIPD) (HT) fibre has been undertaken using transmission electron microscopy. Both PBO and PIPD fibres are composed of rigid-rod polymers having p-phenylene rings in the molecular backbone and show high modulus (280–360 GPa) and high strength (4–6 GPa). It is found that the PBO HM+ fibre has the highest degree of molecular orientation of the three fibres and the longest crystal length along the fibre axis, while the PIPD fibre shows a lower degree of orientation and a shorter crystal length than the PBO fibres. To understand the effect of crystalline size and fine structure of the fibres upon mechanical properties, dark-field and high-resolution lattice images were obtained and analysed in detail.  相似文献

Reactive blending has been usually utilized to stabilize morphology and to improve the properties of multi-component polymer blends by generating copolymers in situ at the interface. However, the present study on blends composed of poly(butylene terephthalate) (PBT) and functionalized styrene-acrylonitrile random copolymer (SAN) demonstrates another possibility for this method, i.e. tailoring morphology and thereby controlling the properties of polymer blends. By varying reaction conditions it was demonstrated that blends could be formed having the same ratio of [PBT]/[SAN] but which possessed completely different microstructural forms: a sea-island morphology with and without micelles, a corded dispersed phase morphology, and a highly oriented, layer-like morphology.  相似文献

Pulsed Electric Current Sintering of Silicon Nitride   总被引:1,自引:0,他引:1  
Pulsed electric current sintering (PECS) has been used to densify α-Si3N4 powder doped with oxide additives of Y2O3 and Al2O3. A full density (>99%) was achieved with virtually no transformation to β-phase, resulting in a microstructure with fine equiaxed grains. With further holding at the sintering temperature, the α-to-β phase transformation took place, concurrent with an exaggerated grain growth of a limited number of elongated β-grains in a fine-grained matrix, leading to a distinct bimodal grain size distribution. The average grain size was found to obey a cubic growth law, indicating that the growth is diffusion-controlled. In contrast, the densification by hot pressing was accompanied by a significant degree of the phase transformation, and the subsequent grain growth gave a broad normal size distribution. The apparent activation energy for the phase transformation was as high as 1000 kJ/mol for PECS, almost twice the value for hot pressing (∼500 kJ/mol), thereby causing the retention of α-phase during the densification by PECS.  相似文献

Summary: True spherical capsules are formed by electrostatic polysaccharide interaction between chitosan and gellan gum via polyion complex (PIC) formation in aqueous solutions. Dropwise addition of a chitosan solution into the gellan solution gave spherical capsules whose outside surface was gellan and whose inside was chitosan (chitosanin‐gellanout capsule). Conversely, the addition of gellan into chitosan yields chitosanout‐gellanin capsules. SEM observation revealed a fibrous network spreading along the capsule membrane of the chitosanin‐gellanout capsule. The releasing properties of the capsules were examined using low molecular weight substances and high molecular weight proteins. For low molecular weight substances, the releasing kinetics were affected by the attractive and repulsive interactions between the substances and the inside component of the capsule. The molecular weight of the encapsulated substances also affects the releasing kinetics. These results, together with a simple preparation procedure, indicate the applicability of chitosan‐gellan capsules as drug‐carrier materials having a controlling release mechanism. As an application example, preparation of an actually eatable artificial roe was also described.

The behaviour of low carbon steels has been studied, in particular the initiation and growth of the major crack which led to final fracture of smooth specimens, via surface replica and photomicrograph techniques. From this study, the fracture process and fatigue life prediction characterized by the growth of surface microcracks have been analysed by a new approach unifying the conventional approaches based on the final fracture of materials with the fracture mechanics approach. Knowledge of fracture parameters is critical for non-destructive inspection during service life and the application of fracture mechanics to life prediction and assessment.  相似文献

Rhodococcus erythropolis strains characterized as antibiotic producers can be classified into three groups according to their antibiotic spectrum and growth compatibility. Due to their high genotypic similarity, the taxonomic relationship of these strains has not been elucidated. In this study, ribosomal protein profiling using matrix-assisted laser desorption ionization-mass spectrometry (MALDI-MS) was employed to classify twenty-one strains of R. erythropolis (15 antibiotic producers and 6 non-antibiotic producers). In the first step in this method, a total of 30 intense peaks observed for purified ribosomal subunit proteins of the type strain (R. erythropolis JCM 3201T) were selected as the reference peaks. The mass spectra observed for the cell lysates of each sample strain were then checked as to whether peaks were observed at the same masses of the reference peaks. The results of peak matching were processed by cluster analysis, generating a dendrogram. Four major clusters of the R. erythropolis strains corresponded to three antibiotic groups and the non-antibiotic group. Furthermore, the topology of the dendrogram was highly comparable with the phylogenetic tree based on DNA gyrase subunit B gene (gyrB) sequencing. These results indicate that our proposed ribosomal protein profiling method using MALDI-MS is a potentially reliable and sufficiently high-throughput technique for the taxonomic analysis of closely related bacterial strains without using DNA sequence information.  相似文献
