通过制备纳豆提取液、纳豆激酶粗酶,对纳豆激酶纤溶活性进行研究。结果表明:纳豆激酶纤溶活性最适pH为8.0,pH6~10溶液中40℃以下基本稳定,pH<5失去纤溶活性;模拟胃环境的酸性条件下,粗酶失去纤溶活性,而纳豆浸提液纤溶活性保持25%;SDS-PAGE电泳显示,胰蛋白酶对纳豆激酶成分没有降解作用;体外溶栓作用表明,纳豆激酶溶解纤维蛋白的方式是直接溶解,而不是通过激活纤溶酶原。  相似文献

从固态发酵培养的纳豆中提取纳豆激酶 ,采用盐析、疏水层析、离子交换层析等方法 ,对提取液进行纳豆激酶的分离纯化。经SDS 聚丙烯酰胺凝胶电泳鉴定 ,活性酶蛋白为单一组分 ,并测得其分子质量为 2 8ku。纳豆激酶在pH5 0~ 1 0 0 ,4~ 3 7℃范围内具有较好的稳定性 ,其纤溶活性可被 0 1mmol/L的PMSF完全抑制。体外溶栓实验表明 ,纳豆激酶为纤溶酶而不是纤溶酶原激活剂。  相似文献

目的:以纳豆激酶的作用底物纤维蛋白为配基制备亲和层析胶,分离纯化纳豆激酶。方法:纳豆杆菌发酵液经硫酸铵分段盐析、透析得粗酶,在4℃条件亲和层析法分离纯化,利用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis,SDS-PAGE)对酶纯度进行检验。结果:经亲和层析得纳豆激酶为电泳纯,纯化倍数为8.3,回收率43.9%。纯酶的最适pH值和温度分别为pH7.0~9.0、50℃;温度高于60℃纤溶酶活性迅速下降;在pH6.0~10.0范围内稳定性好;Mg2+对其有微弱激活作用,Cu2+有显著抑制作用。结论:以琼脂糖包埋纤维蛋白制备的层析胶可用于纳豆激酶的快速分离纯化。  相似文献

从木瓜中分离出能溶解纤维蛋白的活性蛋白。木瓜新鲜乳汁经浸提、盐析、透析得蛋白粗提物;SephadexG-75凝胶层析分离蛋白粗提物,收集蛋白质的洗脱液,依序集合为共1~10个样品,纤维蛋白平板法筛选纤溶活性蛋白;SDS-PAGE电泳检测蛋白纯度及分子量;体外观察该蛋白对血凝块的溶解效果。木瓜蛋白粗提物有纤溶作用,经层析后得到的4、5、6号样品均有纤溶活性,5号样品SDS-PAGE电泳为单一蛋白条带,分子量为26ku,以尿激酶为标准标定该蛋白纤溶活性为17.85IU/mL。该蛋白在体外可通过激活纤溶酶原或直接降解纤维蛋白溶解血栓。木瓜中含有一种分子量为26ku的纤溶活性蛋白。  相似文献

采用柱层析技术对盐析后的纳豆激酶发酵液进行分离纯化,以确定层析分离纳豆激酶的技术参数.对比了Sephadex G-50及Sephadex G-7凝胶层析分离纳豆激酶的效果,并进行了不同pH值条件下的层析效果分析.结果表明,在pH值为6.4时,经Sephadex G-75凝胶层析及SP Sepharose FF离子交换层析后,能够得到较高纯度的纳豆激酶,使纳豆激酶纯化了9.2倍,回收率为51.5%.SDS-PAGE电泳显示所得纳豆激酶为单一蛋白带,其分子质量为28ku.  相似文献

纳豆激酶是一种新型食源性纤溶酶 ,具有易于提取 ,无任何毒副作用 ,溶栓效果好 ,体内作用时间长等特点。实验以新鲜制备纳豆为原料 ,经生理盐水浸提 ,硫酸铵分级盐析 ,SephadexG -10 0层析获得纯纳豆激酶活性蛋白。实验表明 ,该酶分子量为2 80 0 0Da ,等电点为 9,在 6 0℃以下 ,pH 5~ 12均较稳定 ,其最适反应pH为 8,最适反应温度为 45℃ ,Mg2 和Co2 离子对酶活有激活作用 ,而Cu2 、Zn2 和Al3 等离子对该酶有明显的抑制作用。纳豆激酶酶学性质的研究对开发纳豆激酶成为新型溶栓制剂具有重要的实践意义  相似文献

从实验室保存的1株高产纳豆激酶菌株出发,进行发酵产酶,确立具有纤溶活力的纳豆激酶的分离纯化工艺,主要通过硫酸铵分级盐析法和Phenyl Sepharose疏水柱层析进行分离纯化,用纤维蛋白平板法测定酶活力,用SDS-PAGE电泳验证为电泳纯,分子质量为28 ku。试验了不同作用温度、pH值和金属离子对NK活力的影响。结果表明,NK的适宜温度为25~50℃,适宜pH值为7.0,Cu2+,Mn2+,Hg2+是其抑制剂,而Mg2+,Ca2+为激活剂。  相似文献

通过硫酸铵分段沉淀、DEAE-32纤维素柱和SephadexG-75柱层析,从福寿螺(Ampullarum crossean)体内分离纯化出一种相对分子质量约为40kDa的纤溶活性蛋白,紫外光谱法测定表明,该蛋白具有纤溶作用,经SDS-PAGE电泳显示为一条带.研究表明,该蛋白作用最适pH为8.0,最适温度为59℃,热稳定性较好,金属离子Na+对酶的活力无影响,而K+、Mg2+、Ca2+可提高此蛋白酶的活力.采用平板法测定此蛋白酶的血纤维蛋白溶解活性,证明此酶具有强烈的纤溶活性.  相似文献

通过PCR方法扩增纳豆激酶成熟肽和纳豆激酶酶原(proNK)基因片段,并分别克隆到表达载体pET28a上,构建与His标签融合表达的重组表达质粒pET28a-NK和pET28a-proNK,转化大肠杆菌宿主菌BL21(DE3),IPTG诱导表达,SDS-PAGE电泳和纤维蛋白平板检测目的蛋白的表达及表达产物的纤溶活性.结果表明,纳豆激酶成熟肽基因在大肠杆菌中的表达产物以包涵体形式存在,表达产物没有纤溶活性;纳豆激酶酶原基因在大肠杆菌中的表达产物能自我剪切,形成有纤溶活性的纳豆激酶,但同时对宿主细胞有降解毒性.  相似文献

通过对纳豆激酶分离纯化工艺的研究,得出:纳豆激酶发酵液经离心、过滤及CM-Sepharose Fast Flow阳离子交换层析纯化后,纯化液经SDS-PAGE电泳为一条单一色带,纯化倍数达29.6.经中试扩大试验,效果基本一致.通过初步的冻干试验,得出最佳赋型剂为1%甘露醇和2%甘氨酸.  相似文献

A 9% whey protein (WP) isolate solution at pH 7.0 was heat-denatured at 80°C for 30 min. Size-exclusion HPLC showed that native WP formed soluble aggregates after heat-treatment. Additions of CaCl2 (10–40 mM), NaCl (50–400 mM) or glucono-delta-lactone (GDL, 0.4–2.0%, w/v) or hydrolysis by a protease from Bacillus licheniformis caused gelation of the denatured solution at 45°C. Textural parameters, hardness, adhesiveness, and cohesiveness of the gels so formed changed markedly with concentration of added salts or pH by added GDL. Maximum gel hardness occurred at 200 mM NaCl or pH 4.7. Increasing CaCl2 concentration continuously increased gel hardness. Generally, GDL-induced gels were harder than salt-induced gels, and much harder than the protease-induced gel.  相似文献

The levels of bisphenol-F-diglycidyl ether (BFDGE) were quantified as part of a European survey on the migration of residues of epoxy resins into oil from canned fish. The contents of BFDGE in cans, lids and fish collected from all 15 Member States of the European Union and Switzerland were analysed in 382 samples. Cans and lids were separately extracted with acetonitrile. The extraction from fish was carried out with hexane followed by re-extraction with acetonitrile. The analysis was performed by reverse phase HPL C with fluorescence detection. BFDGE could be detected in 12% of the fish, 24% of the cans and 18% of the lids. Only 3% of the fish contained BFDGE in concentrations considerably above 1mg/kg. In addition to the presented data, a comparison was made with the levels of BADGE (bisphenol-A-diglycidyl ether)analysed in the same products in the context of a previous study.  相似文献

Microgels are ‘soft’ microscopic cross-linked polymeric particles that are being increasingly exploited in a variety of industries for rheology control, encapsulation and targeted delivery. They are valued because of the ability to tune their functionality to address specific applications in oil recovery, coatings, drug delivery, cosmetics, personal care and foods. Food microgels are typically biopolymer hydrogels in the form of microspheres, nanospheres (also called nanogels), spheroids and fibres. The utilisation of engineered microgels in foods has so far been limited, despite their great potential to address several needs in the food industry, including: satiety control, encapsulation of phytonutrients and prebiotics, texture control for healthier food formulations (e.g. reduced fat products), and targeting delivery to specific areas in the digestive tract. We review the scientific and patent literature on the utilisation and manufacturing methods for producing microgels with an emphasis on micro-hydrogels for food applications.  相似文献

Joubert and Burns prepared a large number of fractions from the high-sulphur proteins of wool and estimated their molecular weights and amino-acid compositions. Their data have been re-examined in order to look for statistically significant interrelations between amino acids and between the proportion of various amino acids and molecular weight. Statistical analysis of the data is also used to examine the credibility of some hypotheses concerning the mechanism of keratin biosynthesis and to provide further evidence for the existence of families of proteins within the high-sulphur fractions of wool.  相似文献

The European Commission's, Quality of Life Research Programme, Key Action 1—Health, Food & Nutrition is mission-oriented and aims, amongst other things, at providing a healthy, safe and high-quality food supply leading to reinforced consumer confidence in the safety of European food. Its objectives also include the enhancing of the competitiveness of the European food supply. Key Action 1 is currently supporting a number of different types of European collaborative projects in the area of risk analysis. The objectives of these projects range from the development and validation of prevention strategies including the reduction of consumers risks; development and validation of new modelling approaches; harmonization of risk assessment principles, methodologies, and terminology; standardization of methods and systems used for the safety evaluation of transgenic food; providing of tools for the evaluation of human viral contamination of shellfish and quality control; new methodologies for assessing the potential of unintended effects of genetically modified (genetically modified) foods; development of a risk assessment model for Cryptosporidium parvum related to the food and water industries; to the development of a communication platform for genetically modified organism, producers, retailers, regulatory authorities and consumer groups to improve safety assessment procedures, risk management strategies and risk communication; development and validation of new methods for safety testing of transgenic food; evaluation of the safety and efficacy of iron supplementation in pregnant women; evaluation of the potential cancer-preventing activity of pro- and pre-biotic ('synbiotic') combinations in human volunteers.
